Different histories but similar genetic diversity and structure for black walnut in Indiana and Missouri
—Missouri and Indiana have markedly different histories of glaciation and recolonization by forest trees. These states also differ in land use patterns and degree of anthropogenic landscape change such as forest fragmentation. To determine the overall effects of these and other demographic differences on the levels of genetic diversity and structure in black walnut (Juglans nigra L.) more than 550 total black walnut trees from nine populations in Indiana and 10 in Missouri were sampled and analyzed using 12 nuclear microsatellite loci. Although genetic diversity parameters such as allelic richness and expected heterozygosity were high overall, they varied little among populations and their mean values for the two states were not significantly different. Pairwise genetic distance values between all population pairs ranged from 0.012-0.159, but no significant pattern of isolation by distance was detected. The estimate of the degree of genetic differentiation between states (FPT = 0.0009) was very small and not significant, indicating that differences between states explained an inconsequential portion of the total variance. The observed low levels of local and regional genetic structure indicate that high levels of pollen flow have buffered black walnut from the genetic consequences of founder effects and genetic drift in both geologic and recent time scales
An alternative pathway for Alu retrotransposition suggests a role in DNA double-strand break repair
AbstractThe Alu family is a highly successful group of non-LTR retrotransposons ubiquitously found in primate genomes. Similar to the L1 retrotransposon family, Alu elements integrate primarily through an endonuclease-dependent mechanism termed target site-primed reverse transcription (TPRT). Recent studies have suggested that, in addition to TPRT, L1 elements occasionally utilize an alternative endonuclease-independent pathway for genomic integration. To determine whether an analogous mechanism exists for Alu elements, we have analyzed three publicly available primate genomes (human, chimpanzee and rhesus macaque) for endonuclease-independent recently integrated or lineage specific Alu insertions. We recovered twenty-three examples of such insertions and show that these insertions are recognizably different from classical TPRT-mediated Alu element integration. We suggest a role for this process in DNA double-strand break repair and present evidence to suggest its association with intra-chromosomal translocations, in-vitro RNA recombination (IVRR), and synthesis-dependent strand annealing (SDSA)

Impact of HCV testing and treatment on HCV transmission among men who have sex with men and who inject drugs in San Francisco: A modelling analysis
BACKGROUND: Men who have sex with men who ever injected drugs (ever MSM-IDU) carry a high hepatitis C virus (HCV) burden. We estimated whether current HCV testing and treatment in San Francisco can achieve the 2030 World Health Organization (WHO) HCV elimination target on HCV incidence among ever MSM-IDU. METHODS: A dynamic HCV/HIV transmission model among MSM was calibrated to San Francisco data, including HCV antibody (15.5%, 2011) and HIV prevalence (32.8%, 2017) among ever MSM-IDU. MSM had high HCV testing (79%-86% ever tested, 2011-2019) and diagnosed MSM had high HCV treatment (65% ever treated, 2018). Following coronavirus disease 2019 (COVID-19)-related lockdowns, HCV testing and treatment decreased by 59%. RESULTS: Among all MSM, 43% of incident HCV infections in 2022 were IDU-related. Among ever MSM-IDU in 2015, HCV incidence was 1.2/100 person-years (95% credibility interval [CrI], 0.8-1.6). Assuming COVID-19-related declines in HCV testing/treatment persist until 2030, HCV incidence among ever MSM-IDU will decrease by 84.9% (95% CrI, 72.3%-90.8%) over 2015-2030. This decline is largely attributed to HCV testing and treatment (75.8%; 95% CrI, 66.7%-89.5%). Slightly greater decreases in HCV incidence (94%-95%) are projected if COVID-19 disruptions recover by 2025 or 2022. CONCLUSIONS: We estimate that HCV incidence will decline by >80% over 2015-2030 among ever MSM-IDU in San Francisco, achieving the WHO target
Life In The Cold: An Investigation Of Polar Regions
Polar areas provide unique environments that, though they may seem extreme and uninhabitable, are flourishing with life. These areas around the North and South poles include deep oceans, shallow shelf regions, tundra, mountain ranges and vast glaciers. With the increasing effects of global climate change, a basic knowledge of polar regions is crucial to understand future impacts and implications. The purpose of this book is to give a broad background of polar biology, and also provide details on specific examples through case studies. Topics included throughout this book are: Ice, Life in Polar Regions, Species Interactions, and Anthropogenic Impacts.
The students in the Polar Biology course (MAR 464) at the University of New England have researched and reviewed scientific literature to educate readers about these regions. The class, comprised of fourteen junior and senior Marine Science, Ocean Studies and Marine Affairs, and Environmental Sciences students, selected the different topics, presented the material, wrote the chapters, and assembled the final versions into this book. This book cannot be all inclusive, but we think it will provide an excellent broad overview of the most important aspects of Polar Biology and will stimulate the reader to dive into the material further.https://dune.une.edu/marinesci_studproj/1001/thumbnail.jp
