    Expression of an innate immune element (mouse hepcidin-1) in baculovirus expression system and the comparison of its function with synthetic human hepcidin-25

    Hepcidin is an innate immune element which decreases the iron absorption from diet and iron releasing from macrophage cell. In contrast to the chemical iron chelators, there has been limited effort applied to the specific use of hepcidin as a new drug for decreasing the iron overload. Hepcidin is produced in different biological systems. For instance, E-coli is used for human hepcidin expression, however, post-translational modification is impaired. We have used a simple baculovirus expression system (BES) to improve the hepcidin folding and activity. Hepcidin Messenger Ribonucleic acid (mRNA) was isolated from mouse liver cells and its complementary Deoxyribonucleic acid (cDNA) was produced and amplified. PFastBac HTB vector was used for recombinant bacmid production. Recombinant baculovirus was produced using SF-9 cell line. The mouse hepcidin-1 protein was expressed in a large quantity and functional tests were performed for this recombinant peptide. The yield of hepcidin in BES was 20 μg/mL and anti-histidine (anti-His) tag antibody was used for the confirmation of hepcidin on western blot nitrocellulose paper. Functional tests showed that mouse hepcidin accumulates iron in the macrophage cell line J774A.1 up to 63%. In addition, our data showed that the mouse hepcidin-1 has less toxicity compared to the synthetic human hepcidin-25 (p = 0.000). © 2011 by School of Pharmacy

    Hepcidin is a peptide hormone that plays an important role in iron metabolism. We have produced a recombinant mouse hepcidin-1 by using baculovirus expression system. Its expression yield was 25 μg/ml when cell culture media were supplemented with a protease inhibitor cocktail. The recombinant mouse hepcidin-1 and synthetic human hepcidin-25 had similar effects on reducing ferroportin expression in J774A cell line and in peritoneal macrophages. However, synthetic human hepcidin-25 was more efficient than recombinant mouse hepcidin-1 in reducing iron concentration in blood circulation (p < 0.01). © 2013 Springer Science+Business Media Dordrecht.     Synergistic Accumulative Effect of Salicylic Acid and Dibutyl Phthalate on Paclitaxel Production in Corylus avellana Cell Culture

    Suspension cell cultures of Corylus avellana were challenged with salicylic acid and its combined use with dibutyl phthalate solvent. Salicylic acid with concentrations of 12.5, 25 and 50 mg L–1 and 10% (v/v) dibutyl phthalate were used and added on day 8 and 10 of subculture, respectively. The results showed that growth, viability and protein content of cells were decreased by the treatments, compared to control. In all treatments, hydrogen peroxide content and lipid peroxidation rate of cells increased, compared to those of the control cells. Activity of phenylalanine ammonia-lyase increased by salicylic acid and, dibutyl phthalate exaggerated effect of salicylic acid. While flavonoids content decreased by the treatments, paclitaxel content increased significantly. The extracellular paclitaxel was more affected, compared to cell-associated paclitaxel and all treatments increased paclitaxel release and specific yield compared to that of the control. The most production of paclitaxel and specific yield of it were observed under effect of combined use of salicylic acid (50 mg L–1) and dibutyl phthalate, suggesting a synergistic accumulative effect

    Investigation of the field-induced ferromagnetic phase transition in spin polarized neutron matter: a lowest order constrained variational approach

    In this paper, the lowest order constrained variational (LOCV) method has been used to investigate the magnetic properties of spin polarized neutron matter in the presence of strong magnetic field at zero temperature employing AV18AV_{18} potential. Our results indicate that a ferromagnetic phase transition is induced by a strong magnetic field with strength greater than 1018 G10^{18}\ G, leading to a partial spin polarization of the neutron matter. It is also shown that the equation of state of neutron matter in the presence of magnetic field is stiffer than the case in absence of magnetic field.Comment: 23 pages, 9 figures Phys. Rev.     Elucidating social networking apps decisions: Performance expectancy, effort expectancy and social influence

    Purpose: The purpose of this paper is to investigate the determinants of behavioural intention and use behaviour towards social networking apps. Exogenous latent constructs, namely, performance expectancy, effort expectancy and social influence are the key antecedents proposed based on the unified theory of acceptance and use of technology to predict the usage intention and behaviour of social networking apps (i.e. endogenous latent constructs). Experience as a moderator is the extended construct to explain social networking apps user’s behavioural intention. Design/methodology/approach: To target young generation (Millennial), a cross-sectional data collection approach was conducted to collect data from the social networking apps users (i.e. Facebook, WhatsApp, WeChat, Twitter, Instagram, YouTube, Snapchat and others) whereby a total of 384 valid questionnaires were obtained from six universities in Malaysia. Statistical analysis using partial least squares path modelling approach and a variance-based structural equation modelling (VB-SEM) techniques is performed to analyse the measurement and structural relationship. Findings: The findings indicate that performance expectancy, effort expectancy and social influence determine behavioural intention, and behavioural intention impacts social networking apps use behaviour. Moreover, the moderation analysis reveals that the relationship between effort expectancy and behavioural intention is moderated by experience, whereas the relationship between social influence and behavioural intention is not moderated by experience. Originality/value: While the surge of social networking apps has gained tremendous popularity among Millennial as an attractive market segment, previous studies mainly have focussed on intention and behaviour of online users in general. Despite apps and related technologies which have opened a new era of effective communications in marketing, social networking apps usage intention and behaviour focussing on Millennial is not well understood in the current literature. This study contributes and sheds lights on the current issue of social networking apps usage intention and behaviour and looks into a key rising market segment, the Millennial users. © 2018, Emerald Publishing Limited

    How can patient journey in surgical wards of a referral hospital be improved?

    Background: We studied the patient journey in surgical wards in order to find an effective and efficient way of scheduling in surgical wards.Methods: We applied Root cause analysis (RCA) model within three months in a referral hospital. After understanding root causes of the events occurred through a focus discussion group, required interventions were proposed according to literatures, experiences, and preference of the stakeholders. Possible interventions were also analyzed based on its ability to reduce contributing factors in the events and the belief of process-owner that if interventions can be implemented.Results: The results were provided for five main steps: 1) the most important root cause was “not prioritizing patients and pre-scheduling the number of surgical procedures in the days before”. 2) Constraints indicated that workforce weren't allocated proportionally to the number of surgical operations in varying shift lengths, increased numbers of on-calls physicians increased related costs, the admission of patients in VIP wards have been getting a high priority, and surgeon compensation based on fee for service method was challenging. 3) The current situation of allocating three rooms on average for each physician can be changed depending on numbers of surgeries. 4) Proposed interventions are establishing a computer registration system, reforming payment methods, setting up an electronic waiting list, development of scheduling guidelines, and Applying MIP model.Conclusions: Implementing of scheduling reforms requires a comprehensive action plan system and predefined functional indicators.     Phase Change Material Systems for High Temperature Heat Storage

    Efficient, cost effective, and stable high-temperature heat storage material systems are important in applications such as high-temperature industrial processes (metal processing, cement and glass manufacturing, etc.), or electricity storage using advanced adiabatic compressed air energy storage. Incorporating phase change media into heat storage systems provides an advantage of storing and releasing heat at nearly constant temperature, allowing steady and optimized operation of the downstream processes. The choice of, and compatibility of materials and encapsulation for the phase change section is crucial, as these must guarantee good and stable performance and long lifetime at low cost. Detailed knowledge of the material properties and stability, and the coupled heat transfer, phase change, and fluid flow are required to allow for performance and lifetime predictions. We present coupled experimental-numerical techniques allowing prediction of the long-term performance of a phase change material-based high-temperature heat storage system. The experimental investigations focus on determination of material properties (melting temperature, heat of fusion, etc.) and phase change material and encapsulation interaction (stability, interface reactions, etc.). The computational investigations focus on an understanding of the multi-mode heat transfer, fluid flow, and phase change processes in order to design the material system for enhanced performance. The importance of both the experimental and numerical approaches is highlighted and we give an example of how both approaches can be complementarily used for the investigation of long-term performance

    Pattern of Deliberate Self-Poisoning in Gorgan, North of Iran

    Background: Suicide is a global public health problem. Deliberate Self-Poisoning (DSP) is one of the most common methods of suicide in many countries. This study was designed to identify the trends and characteristics of DSP in Gorgan.Methods: The study was carried out retrospectively in 5 Azar Hospital. It included 549 patients who were hospitalized in the hospital due to DSP from March 2008 to March 2015. Data were obtained from medical records. Stata software and Pearson's chi-squared test were used for data analysis.Results: Of 549 patients, 51% were females and 50.27% were aged 20–29 years. The majority of patients (76.68%) lived in urban areas. Poisoning occurred mostly in summer and the peak was observed in August. Most of the poisoning agents were pharmaceuticals (80.51%). Among the pharmaceuticals, benzodiazepines were involved most often. Overall, 21 patients (3.83%) died. The highest number of deaths was due to aluminum phosphide poisoning (76.19%). In addition, family quarrel was the main cause of DSP (43.17%). There were significant differences between the causes of DSP and demographics. Characteristics including gender, age groups, marital status, employment status and educational status.Conclusion: Deliberate self-poisoning (DSP) with drugs has recently been a serious social problem, especially in the younger generation in Gorgan and there is an urgent need for a prevention plan
