    CD36 gene polymorphism -31118 G > A (rs1761667) is associated with overweight and obesity but not with fat preferences in Mexican children

    CD36 glycoprotein is a candidate receptor involved in the gustatory detection of lipids and emerging evidence has suggested that genetic variations in CD36 may modulate the oral perception threshold to fatty acids. Here, we analyzed the association of -31118 G > A polymorphism in CD36 gene with nutritional status and preferences for fatty foods in Mexican children. Genotyping of SNP rs1761667 was performed in school-age children (n = 63) in addition to sensory tests evaluating the preference and satisfaction score assigned to oil-based sauces of different fatty acid composition. The G allele was associated with high BMI z-score in children (OR = 2.43, 95% (CI 1.02–5.99); p = 0.02) but CD36 genotypes (AA, GA, and GG) did not show significant association with the preference and satisfaction scores assigned to oil-based sauces. The BMI z-score showed no association with the preference to oil-based sauces; however, children with normal weight gave higher satisfaction scores to sauces with a high content of unsaturated fatty acids than to sauces rich in saturated fatty acids (0.56 ± 1.26 vs. 0.06 ± 1.22; p = 0.02).     Study and research of feeding behavior: Roots, development and challenges / Estudio e investigación del comportamiento alimentario: Raíces, desarrollo y retos

    Abstract The present work provides a review of the basis and development of the study and research on feeding behavior (FB). Initial contributions and those that gave identity to this field are highlighted. Among them, the behavior is approached as an essential mean to maintain the state of balance in the organisms, as well as the role of learning as a determinant of food preference, selection and consumption. Subsequently, are presented the arguments that establish FB as an element that integrate the knowledge related to feeding generated by other areas of science. Finally, are stated the main challenges for academics dedicated to the study and research of FB and the role of education on food and nutrition as an optimal solution for food problems. Resumen El presente trabajo expone una revisión de las raíces y el desarrollo del estudio e investigación sobre comportamiento alimentario (CA). Se destacan las aportaciones iniciales y algunas otras que le han venido dotando de identidad a esta área de estudio. Entre ellas, el señalamiento de la conducta como medio indispensable para mantener el estado de equilibrio en los organismos, y el papel del aprendizaje como determinante de la preferencia, la selección y el consumo de alimento. Posteriormente se presentan los argumentos que establecen al CA como elemento integrador del conocimiento que sobre alimentación han generado otras áreas de la ciencia. Finalmente se puntualizan los principales retos a enfrentar por parte de los académicos dedicados al estudio e investigación sobre CA, y se pondera el papel de la educación en alimentación y nutrición como la solución óptima a las problemáticas alimentarias.

    IgG Anti-ghrelin Immune Complexes Are Increased in Rheumatoid Arthritis Patients Under Biologic Therapy and Are Related to Clinical and Metabolic Markers

    Rheumatoid arthritis (RA) is a systemic autoimmune disease associated with increased risk of cardiovascular disease and metabolic alterations. The mechanisms underlying these alterations remain unclear. Ghrelin is a gastrointestinal hormone with potent effects on food intake, body weight, metabolism, and immune response. Recent studies reported the presence of anti-ghrelin autoantibodies in healthy subjects and the levels and affinity of these autoantibodies were altered in anorectic and obese individuals. In this cross-sectional study we analyzed anti-ghrelin autoantibodies in RA patients and evaluated its relationship with clinical, body-composition and metabolic parameters. Clinical measurements of RA patients included the disease activity score-28 (DAS-28), inflammatory biomarkers, autoantibodies (RF and anti-CCP), body composition, glucose and lipid profile. Serum ghrelin levels were measured by enzyme-linked immunosorbent assay (ELISA). Free and total anti-ghrelin autoantibodies quantification (IgG and IgA isotypes) was performed by in-house ELISA. RA patients had lower IgG anti-ghrelin autoantibodies levels and higher immune complexes percentage (IgG+ghrelin) compared to the control group, while the IgA anti-ghrelin autoantibodies showed no significant differences. In the bivariate analysis, the percentage of IgG anti-ghrelin immune complexes positively correlated with BMI and ghrelin whereas in the multivariate regression model, the variables associated were DAS-28, body weight, visceral fat, LDL-C and TG (R2 = 0.72). The percentage of IgA anti-ghrelin immune complexes positively correlated with RF and anti-CCP and the multivariate regression model showed an association with RF and body fat percentage (R2 = 0.22). Our study shows an increased percentage of IgG anti-ghrelin immune complexes in RA patients despite ghrelin levels were similar in both groups, suggesting an increase in the affinity of these autoantibodies toward ghrelin. The associations found in the multiple regression analysis for anti-ghrelin immune complexes support the previously reported functions of these natural autoantibodies as carriers and modulators of the stability and physiological effect of the hormone. However, in RA both the disease activity and the RF appear to influence the formation of these anti-ghrelin immune complexes

    La ingesta de stevia modifica la dinámica del peso corporal, la glucosa y el colesterol en ratas hembras: Algoritmo k-NNs

    El consumo de stevia ha sido promovido por su bajo aporte calórico, su efecto antidiabético y antihipercolesterolémico. Sin embargo, los efectos de la ingesta de stevia parecen no ser los mismos para las ratas hembras respecto de los machos. El propósito de este estudio fue evaluar el efecto de la ingesta de stevia sobre el consumo de alimento, peso corporal y niveles de glucosa, insulina, colesterol y triglicéridos en ratas hembras Wistar durante 13 semanas y realizar un análisis predictivo del peso corporal y la ingesta de alimento a 20 semanas. Se utilizaron 20 ratas hembras adultas, que se dividieron en 2 grupos: control (CG) y stevia (SG), ambos grupos recibieron agua y comida a libre acceso, así como una solución de stevia al 0,2 % para el grupo SG. Se registró diariamente el consumo de alimento, agua y solución de stevia; la medición del peso corporal se realizó semanalmente. Al final de las 13 semanas de experimentación, los animales se sacrificaron para evaluar los parámetros metabolicos. El grupo SG mostró un mayor consumo de alimento, mayor proporción de ganancia de peso corporal, niveles de glucosa y colesterol que el grupo CG. No se encontraron diferencias significativas en los niveles de triglicéridos e insulina. Respecto al análisis predictivo (semanas 14-20), se mantiene un incremento significativo en el consumo de alimento y se observa una tendencia de aumento en la proporción de ganancia de peso corporal. Esto indica que el consumo de stevia en ratas hembras parece no tener los mismos efectos benéficos reportados en machos.Consumption of stevia has been promoted due to its low caloric intake, it’s effects as anti-diabetic and anti-hypercholesterolemic. However, the effects of stevia consumption is apparently not the same in females than males. The purpose of this study was to evaluate the effect of stevia intake on meal consumption, body weight and levels of glucose, insulin, cholesterol and triglycerides in female Wistar rats during 13 weeks and develop a predictive analysis of the body weight and meal intake over 20 weeks. 20 adult female rats were utilized, these were divided into two groups: control (CG) and stevia (SG), both groups received free access to water and food, the SG also received a stevia solution at 0.2%. Consumption of food, water and stevia solution was recorded daily, while weight was recorded weekly. At the end of the 13 weeks of experiment, the subjects were sacrificed to evaluate the metabolic parameters. The SG group showed a higher consumption of food, higher proportion of body weight gain, glucose levels and cholesterol than the CG. No significant differences were found in levels of triglyceride or insulin. Respect to the predictive analysis (weeks 14-20), a significant increase in food consumption is maintained and an increasing trend is observed in the proportion of body weight gain. This indicates that stevia consumption appears not to have the same benefit effects in female rats than male rats

    Altered Expression of TSPAN32 during B Cell Activation and Systemic Lupus Erythematosus

    Systemic lupus erythematosus (SLE) is a chronic inflammatory disease with various clinical features. Autoreactive B cells play a role in disease pathogenesis, through the production of multiple autoantibodies, which form immune complexes and induce the inflammatory response and tissue damage associated with SLE. Recently, tetraspanins, and in particular, TSPAN32, have been recognized to play a central role in immunity, as they are involved in various biological processes, such as the antigen presentation and the activation of lymphocytes. Evidence suggests that tetraspanins could represent in the future a target for therapeutic purposes in patients suffering from autoimmune/immunoinflammatory disorders. In the present study, by performing in silico analyses of high-throughput data, we evaluated the expression levels of TSPAN32 in B cell activation and investigated its modulation in circulating B cells from SLE patients. Our data show that B cell activation is associated with a significant downregulation of TSPAN32. Additionally, significantly lower levels of TSPAN32 were observed in circulating plasmablasts from SLE patients as compared to healthy donor plasmablasts. In addition, type I interferons (IFNs)-related genes were enriched among the genes negatively correlated to TSPAN32, in SLE plasmablasts. Accordingly, IFN-α is able to induce a dose-dependent downregulation of TSPAN32 in B cells. Overall, the data here presented suggest the potential use of TSPAN32 as a diagnostic marker and therapeutic target for the evaluation and management of humoral immune responses in chronic diseases, such as SLE

    The Relevance of Selenium Status in Rheumatoid Arthritis

    Rheumatoid arthritis (RA) is an autoimmune and inflammatory disease that can cause joint damage. Among the environmental risk factors, diet plays an important role because it can aggravate or attenuate inflammation. Selenium (Se) is considered an essential trace element since it is a structural component of antioxidant enzymes; however, its concentration can be affected by diet, drugs and genetic polymorphisms. Studies have reported that RA patients have a deficient diet in some food groups that is associated with parameters of disease activity. Furthermore, it has been shown that there is an alteration in serum Se levels in this population. Although some clinical trials have been conducted in the past to analyze the effect of Se supplementation in RA, no significant results were obtained. Contrastingly, experimental studies that have evaluated the effect of novel Se nanoparticles in RA-induced models have shown promising results on the restoration of antioxidant enzyme levels. In particular, glutathione peroxidase (GPx) is an important selenoprotein that could have a modulating effect on inflammation in RA. Considering that RA patients present an inflammatory and oxidative state, the aim of this review is to give an overview of the current knowledge about the relevance of Se status in RA

    Fiber Consumption Mediates Differences in Several Gut Microbes in a Subpopulation of Young Mexican Adults

    Diet is a determinant for bodyweight and gut microbiota composition. Changes in dietary patterns are useful for the prevention and management of overweight and obesity. We aim to evaluate diet behavior and its potential association with selected gut bacteria and body weight among Mexican young adults. Mexican college students aged between 18 and 25 (normal-weight, overweight, and obese) were recruited. Anthropometric variables were recorded. A validated food frequency questionnaire was applied to all the participants. The percentages of macronutrients, fiber, and energy were calculated, and fecal samples were analyzed by real-time-qPCR to quantify selected gut bacteria. All the participants showed an unbalanced dietary pattern. However, the consumption of fruits, non-fat cereals, and oils and fats without protein were higher in the normal-weight individuals. In the overweight/obese participants, fiber intake did not correlate with the microbial variables, while Kcal from protein and Clostridium leptum correlated positively with Lactobacillus. Similarly, Clostridium coccoides-Eubacterium rectale correlated with Akkermansia muciniphila. In the normal-weight participants, Clostridium leptum and Lactobacillus correlated positively with Clostridium coccoides-Eubacterium rectale and Bifidobacterium, respectively, and Bacteroidetes negatively with Akkermansia muciniphila. In conclusion, a higher fiber intake had a positive impact on body weight and bacterial gut composition in this Mexican population of college students

    Sedentary behavior’s role on health damage has been documented as a promoting factor of pathologies. Scientific evidence shows an increasing percentage of people with sedentary activity especially in large cities causing overweight, obesity, diabetes, hypertension etc. Effects of interrupting sedentary behavior periods with physical activity have been recently evaluated, demonstrating increases on energy expenditure. The objective of this experiment was to compare the effect of two interruption programs of sedentary behavior pattern on caloric expenditure. Participants were exposed to two interruption programs of sedentary behavior. For energy expenditure, heart monitors brand Beurer PM18 model used. First program consisted on 15-minutes periods of sedentary behavior followed by a 2.5 minutes walking break as physical activity. Second program consisted on periods of 30 minutes of sedentary behavior followed by a 5 minutes break of physical activity. Analysis used Student t test for paired samples showed a significant difference in caloric expenditure during sedentary behavior between program 1 and 2. Concluding that increasing the number of interruptions of periods of sedentary behavior has a direct effect on caloric expenditure.El papel de la conducta sedentaria sobre el deterioro del estado de salud ha sido ampliamente documentado como factor promotor de patologías. La evidencia científica demuestra que el porcentaje de personas que mantienen una actividad sedentaria aumenta en las grandes ciudades, y con ello el sobrepeso, la obesidad, la diabetes, la hipertensión, etc. Recientemente se ha evaluado el efecto de interrumpir los periodos de conducta sedentaria con actividad física, demostrando que con ello se aumenta el gasto energético. El objetivo del presente experimento fue comparar el efecto de 2 programas de interrupción del patrón de conducta sedentaria sobre el gasto calórico. Ocho alumnos universitarios se expusieron a 2 programas de interrupción de la conducta sedentaria. Para obtener el gasto energético se utilizaron monitores cardiacos de la marca Beurer modelo pm18. El primer programa consistió en periodos de 15 min de conducta sedentaria seguidos de una interrupción de 2.5 min de caminata como actividad física. El segundo presentó periodos de 30 min de conducta sedentaria seguidos de una interrupción de 5 min de actividad física. El análisis utilizando t de Student para muestras pareadas mostró que existe una diferencia significativa en el gasto calórico durante la conducta sedentaria entre el programa 1 y el 2. Se concluye que aumentar el número de interrupciones en los periodos de conducta sedentaria tiene un efecto directo sobre el gasto calórico
