    A comparison between faecal sterols and coliform counts in the investigation of sewage contamination in sediments

    Em setembro de 2002, foram coletadas nove amostras de sedimento superficial (0-2 cm) na Enseada de Botafogo (sudoeste da Baía de Guanabara/RJ), a fim de comparar o uso de marcadores químicos (coprostanol) e biológicos (E. coli e coliformes totais) na identificação da contaminação fecal da região. Os resultados obtidos (carbono orgânico - 6,0 to 64,8 mg g-1; coprostanol - 1,4 to 105 µg g-1; E. coli - < 30 to 2400 NMP/10g e coliformes totais - 40 to 9300 NMP/10g) foram iguais ou maiores aos observados em outras áreas contaminadas da Baía de Guanabara. Nas estações próximas à linha de costa, as concentrações de coprostanol e as contagens das bactérias confirmaram que o esgoto doméstico se acumulou no sedimento da enseada. As concentrações de coprostanol se mantiveram altas nas estações distantes das fontes de contaminação, porém o material fecal representou uma menor fração do carbono orgânico. Nessas mesmas estações, redução na contagem de colimetria foi proporcionalmente mais alta, provavelmente pela baixa sobrevivência das bactérias devido ao efeito de luz, salinidade e temperatura. durante o transporte e deposição das partículas de esgoto. Portanto, baseando-se nos resultados obtidos, o coprostanol foi um indicador mais adequado da contaminação fecal nos sedimentos da Enseada de Botafogo.In September 2002, nine sediment samples (0-2 cm) were collected from Botafogo Cove (southwestern part of Guanabara Bay) in order to compare the use of chemical (coprostanol) and biological markers (E. coli and total coliforms) in identifying faecal contamination. The values found (organic carbon - 6.0 to 64.8 mg g-1; coprostanol - 1.4 to 105 µg g-1; E. coli - < 30 to 2400 NMP/10g and total coliforms - 40 to 9300 NMP/10g) were similar to or even higher than those observed in other contaminated areas of Guanabara Bay. In stations close to the shoreline, both coprostanol concentration and bacteria count confirmed that domestic sewage had accumulated in the cove. Coprostanol concentrations were still relatively high in the stations furthest from contamination sources, although faecal material represented a smaller fraction of the total organic carbon originated by dilution in waters containing algae. Reduction was proportionately higher by virtue of the use of the colimetric assessment method. This result may be associated with the effect that environmental variables (light, salinity, temperature) may have on the survival of bacteria during transport and deposition of sewage particles. Consequently, based on these results, coprostanol may be considered the better indicator of the faecal contamination level in Botafogo Cove sediments

    Design, synthesis and the effect of 1,2,3-triazole sialylmimetic neoglycoconjugates on Trypanosoma cruzi and its cell surface trans-sialidase

    This work describes the synthesis of a series of sialylmimetic neoglycoconjugates represented by 1,4-disubstituted 1,2,3-triazole-sialic acid derivatives containing galactose modified at either C-1 or C-6 positions, glucose or gulose at C-3 position, and by the amino acid derivative 1,2,3-triazole fused threonine-3-O-galactose as potential TcTS inhibitors and anti-trypanosomal agents. This series was obtained by Cu(I)-catalysed azide-alkyne cycloaddition reaction ('click chemistry') between the azido-functionalized sugars 1-N(3)-Gal (commercial), 6-N(3)-Gal, 3-N(3)-Glc and 3-N(3)-Gul with the corresponding alkyne-based 2-propynyl-sialic acid, as well as by click chemistry reaction between the amino acid N(3)-ThrOBn with 3-O-propynyl-GalOMe. the 1,2,3-triazole linked sialic acid-6-O-galactose and the sialic acid-galactopyranoside showed high Trypanosoma cruzi trans-sialidase (TcTS) inhibitory activity at 1.0 mM (approx. 90%), whilst only the former displayed relevant trypanocidal activity (IC(50) 260 mu M). These results highlight the 1,2,3-triazole linked sialic acid-6-O-galactose as a prototype for further design of new neoglycoconjugates against Chagas' disease. (C) 2011 Elsevier B.V.     Quantification and distribution of vibrio species in water from an estuary in Ceará-Brazil impacted by shrimp farming

    Vibrios were quantified and their distribution determined for the estuary of the Coreaú river, in Northeastern Brazil, based on 24 water samples collected between June and October 2005. The most probable number of vibrios per 100 mL ranged from 230 x 10³ to 240 x 10(11). The pH value was the environmental factor most strongly associated with the abundance of vibrios. Sixty-two vibrio strains were isolated belonging to 15 species (6 of which observed in June-September and 8 in October). The most frequently isolated species were V. parahaemolyticus and V. cholerae.Foram realizadas análises concernentes à quantificação e distribuição de Vibrio em 24 amostras de água do estuário do Rio Coreaú (CE) no período de junho a novembro de 2005. O Número Mais Provável (NMP/100 mL) de Vibrio oscilou de 230 x 10³ a 240 x 10(11). O pH foi o fator ambiental que mais favoreceu ao aumento da microbiota de Vibrio. Foram isoladas 62 cepas de Vibrio, sendo observada uma distribuição de sete espécies nos meses de junho a setembro, e oito espécies no mês de outubro. As espécies mais freqüentes nos isolamentos foram V. parahaemolyticus e V. cholerae

    Estudo dos efeitos de diferentes doses de surfactante exógeno para o tratamento da síndrome de aspiração de mecônio em coelhos recém-nascidos

    OBJECTIVE: To evaluate the effects of 2 different doses of exogenous surfactant on pulmonary mechanics and on the regularity of pulmonary parenchyma inflation in newborn rabbits. METHOD: Newborn rabbits were submitted to tracheostomy and randomized into 4 study groups: the Control group did not receive any material inside the trachea; the MEC group was instilled with meconium, without surfactant treatment; the S100 and S200 groups were instilled with meconium and were treated with 100 and 200 mg/kg of exogenous surfactant (produced by Instituto Butantan) respectively. Animals from the 4 groups were mechanically ventilated during a 25-minute period. Dynamic compliance, ventilatory pressure, tidal volume, and maximum lung volume (P-V curve) were evaluated. Histological analysis was conducted using the mean linear intercept (Lm), and the lung tissue distortion index (SDI) was derived from the standard deviation of the means of the Lm. One-way analysis of variance was used with a = 0.05. RESULTS: After 25 minutes of ventilation, dynamic compliance (mL/cm H2O · kg) was 0.87 ± 0.07 (Control); 0.49 ± 0.04 (MEC*); 0.67 ± 0.06 (S100); and 0.67 ± 0.08 (S200), and ventilatory pressure (cm H2O) was 9.0 ± 0.9 (Control); 16.5 ± 1.7 (MEC*); 12.4 ± 1.1 (S100); and 12.1 ± 1.5 (S200). Both treated groups had lower Lm values and more homogeneity in the lung parenchyma compared to the MEC group: SDI = 7.5 ± 1.9 (Control); 11.3 ± 2.5 (MEC*), 5.8 ± 1.9 (S100); and 6.7 ± 1.7 (S200) (*P < 0.05 versus all the other groups). CONCLUSIONS: Animals treated with surfactant showed significant improvement in pulmonary mechanics and more regularity of the lung parenchyma in comparison to untreated animals. There was no difference in results after treatment with either of the doses used.OBJETIVO: Avaliar os efeitos de duas diferentes doses de surfactante exógeno sobre a mecânica pulmonar e sobre a regularidade da expansão do parênquima pulmonar em coelhos recém-nascidos. MÉTODO: Coelhos recém-nascidos foram traqueostomizados e randomizados em quatro grupos de estudo: grupo-Controle, sem aspiração de mecônio; grupo MEC, com aspiração de mecônio e sem tratamento com surfactante exógeno; grupos S100 e S200, ambos com aspiração de mecônio e tratados respectivamente com 100 e 200 mg/kg de surfactante exógeno (produzido e fornecido pelo Instituto Butantan). Os animais dos 4 grupos foram ventilados por 25 minutos. A mecânica pulmonar foi avaliada a partir dos valores de complacência dinâmica, pressão ventilatória, volume-corrente e volume pulmonar máximo (curva P-V). A análise histológica foi feita calculando-se o diâmetro alveolar médio (Lm) e o índice de distorção através do desvio padrão do Lm. Utilizou-se ANOVA One Way com a = 0,05. RESULTADOS: Após 25 minutos de ventilação, os valores de complacência dinâmica (ml/cm H2O.kg) foram: 0,87± 0,07 (Controle); 0,49±0,04 (MEC*); 0,67±0,06 (S100) e 0,67±0,08 (S200) e de pressão ventilatória (cm H2O): 9,0± 0,9 (Controle); 16,5±1,7 (MEC*); 12,4±1,1 (S100) e 12,1±1,5 (S200). Ambos os grupos tratados tiveram padrão de expansão do parênquima mais homogêneo em relação aos animais não tratados: índice de distorção de 7,5± 1,9 (Controle); 11,3±2,5 (MEC*); 5,8±1,9 (S100) e 6,7±1,7 (S200) (*p < 0,05 vs outros grupos). CONCLUSÕES: Animais tratados com surfactante mostraram melhora significativa da mecânica pulmonar e maior homogeneidade do padrão de expansão pulmonar comparados ao grupo não tratado. Não houve influência das doses de surfactante utilizadas

    α-Selective glycosylation affords mucin-related GalNAc amino acids and diketopiperazines active on Trypanosoma cruzi

    AbstractThis work addresses the synthesis and biological evaluation of glycosyl diketopiperazines (DKPs) cyclo[Asp-(αGalNAc)Ser] 3 and cyclo[Asp-(αGalNAc)Thr] 4 for the development of novel anti-trypanosomal agents and Trypanosoma cruzi trans-sialidase (TcTS) inhibitors. The target compounds were synthetized by coupling reactions between glycosyl amino acids αGalNAc-Ser 7 or αGalNAc-Thr 8 and the amino acid (O-tBu)-Asp 17, followed by one-pot deprotection-cyclisation reaction in the presence of 20% piperidine in DMF. The protected glycosyl amino acid intermediates 7 and 8 were, in turn, obtained by α-selective, HgBr2-catalysed glycosylation reactions of Fmoc-Ser/Thr benzyl esters 12/14 with αGalN3Cl 11, being, subsequently, fully deprotected for comparative biological assays. The DKPs 3 and 4 showed relevant anti-trypanosomal effects (IC50 282–124μM), whereas glycosyl amino acids 1 and 2 showed better TcTS inhibition (57–79%) than the corresponding DKPs (13–25%)

    Atlantic mammal traits: a dataset of morphological traits of mammals in the atlantic forest of south America

    Measures of traits are the basis of functional biological diversity. Numerous works consider mean species-level measures of traits while ignoring individual variance within species. However, there is a large amount of variation within species and it is increasingly apparent that it is important to consider trait variation not only between species, but also within species. Mammals are an interesting group for investigating trait-based approaches because they play diverse and important ecological functions (e.g., pollination, seed dispersal, predation, grazing) that are correlated with functional traits. Here we compile a data set comprising morphological and life history information of 279 mammal species from 39,850 individuals of 388 populations ranging from −5.83 to −29.75 decimal degrees of latitude and −34.82 to −56.73 decimal degrees of longitude in the Atlantic forest of South America. We present trait information from 16,840 individuals of 181 species of non-volant mammals (Rodentia, Didelphimorphia, Carnivora, Primates, Cingulata, Artiodactyla, Pilosa, Lagomorpha, Perissodactyla) and from 23,010 individuals of 98 species of volant mammals (Chiroptera). The traits reported include body mass, age, sex, reproductive stage, as well as the geographic coordinates of sampling for all taxa. Moreover, we gathered information on forearm length for bats and body length and tail length for rodents and marsupials. No copyright restrictions are associated with the use of this data set.     Methicillin-resistant Staphylococcus aureus (MRSA) carriage in a dermatology unit

    OBJECTIVE: The aim of this study was to characterize Staphylococcus aureus (MRSA) carriage in a dermatology unit. METHODS: This was a prospective and descriptive study. Over the course of 26 weeks, surveillance cultures were collected weekly from the anterior nares and skin of all patients hospitalized in a 20-bed dermatology unit of a tertiary-care hospital. Samples from healthcare workers (HCWS) were cultured at the beginning and end of the study. Colonized patients were put under contact precautions, and basic infection control measures were enforced. Staphylococcus aureus colonization pressure was determined monthly. Colonized and non-colonized patients were compared, and isolates were evaluated for antimicrobial susceptibility, SCCmec type, virulence factors, and type. RESULTS: Of the 142 patients evaluated, 64 (45%) were colonized by MRSA (39% hospital acquired; 25% community acquired; 36% indeterminate). Despite isolation precautions, hospital-acquired Staphylococcus aureus occurred in addition to the continuous entry of Staphylococcus aureus from the community. Colonization pressure increased from 13% to 59%, and pemphigus and other bullous diseases were associated with MRSA colonization. Eleven out of 71 HCWs (15%) were Staphylococcus aureus carriers, although only one worker carried a persistent clone. Of the hospital-acquired MRSA cases, 14/28 (50%) were SCCmec type IV (3 PFGE types), 13 were SCCmec type III (46%), and one had an indeterminate type. These types were also present among the community-acquired Staphylococcus aureus isolates. SSCmec type IV isolates were shown to be more susceptible than type III isolates. There were two cases of bloodstream infection, and the pvl and tst virulence genes were absent from all isolates. CONCLUSIONS: Dermatology patients were colonized by community- and hospital-acquired Staphylococcus aureus. Half of the nosocomial Staphylococcus aureus isolates were SCCmec type IV. Despite the identification of colonized patients and the subsequent contact precautions and room placement, Staphylococcus aureus colonization continued to occur, and colonization pressure increased. Pemphigus and other bullous diseases were associated with Staphylococcus aureus

    Discriminating among Earth composition models using geo-antineutrinos

    It has been estimated that the entire Earth generates heat corresponding to about 40 TW (equivalent to 10,000 nuclear power plants) which is considered to originate mainly from the radioactive decay of elements like U, Th and K, deposited in the crust and mantle of the Earth. Radioactivity of these elements produce not only heat but also antineutrinos (called geo-antineutrinos) which can be observed by terrestrial detectors. We investigate the possibility of discriminating among Earth composition models predicting different total radiogenic heat generation, by observing such geo-antineutrinos at Kamioka and Gran Sasso, assuming KamLAND and Borexino (type) detectors, respectively, at these places. By simulating the future geo-antineutrino data as well as reactor antineutrino background contributions, we try to establish to which extent we can discriminate among Earth composition models for given exposures (in units of ktâ‹…\cdot yr) at these two sites on our planet. We use also information on neutrino mixing parameters coming from solar neutrino data as well as KamLAND reactor antineutrino data, in order to estimate the number of geo-antineutrino induced events.Comment: 24 pages, 10 figures, final version to appear in JHE
