Prevalencia de Dirofilaria immitis en perros de la Fundación Protectora de animales de Dosquebradas, Risaralda, Colombia

La Dirofilaria immitis causante de la dirofilariasis o de la enfermedad del corazón del perro es de distribución mundial, transmitida por mosquitos del género Aedes sp., Anopheles sp., Culex sp. La picadura de cualquier género de estos mosquitos sí es vector, introduce las larvas en el animal afectándolo internamente con problemas comúnmente cardíacos, cutáneos y pulmonares. Se han encontrado algunos casos de este parásito afectando humanos que, a diferencia del perro, no hay una filaremia. Los síntomas más comunes son: dolor retroesternal, tos y hemoptisis. Igualmente se ha encontrado en algunos estudios que estos casos en humanos se han ido extendiendo en lugares cada vez menos propensos, debido a sus vectores y al cambio climático por el que se ha pasado durante la última década. Como objetivo en este estudio se determinó la presencia de este parásito dentro de la ‘Fundación Protectora de animales de Dosquebradas’ con el fin de tener datos más concisos y reales de esta enfermedad, facilitar su diagnóstico, tratamiento y prevención. Igualmente se realizaron algunas correlaciones entre sexo, edad, condición corporal y lugar en el que provenían estos perros para hacer un análisis más adecuado de los afectados y los no afectados. En el estudio se utilizó un kit IDEXX Snap 4Dx plus® para D. immitis por cada muestra de sangre extraída en cada perro, la población muestreada fue de veinte animales

A RNA Interference Screen Identifies the Protein Phosphatase 2A Subunit PR55γ as a Stress-Sensitive Inhibitor of c-SRC

Protein Phosphatase type 2A (PP2A) represents a family of holoenzyme complexes with diverse biological activities. Specific holoenzyme complexes are thought to be deregulated during oncogenic transformation and oncogene-induced signaling. Since most studies on the role of this phosphatase family have relied on the use of generic PP2A inhibitors, the contribution of individual PP2A holoenzyme complexes in PP2A-controlled signaling pathways is largely unclear. To gain insight into this, we have constructed a set of shRNA vectors targeting the individual PP2A regulatory subunits for suppression by RNA interference. Here, we identify PR55γ and PR55δ as inhibitors of c-Jun NH2-terminal kinase (JNK) activation by UV irradiation. We show that PR55γ binds c-SRC and modulates the phosphorylation of serine 12 of c-SRC, a residue we demonstrate to be required for JNK activation by c-SRC. We also find that the physical interaction between PR55γ and c-SRC is sensitive to UV irradiation. Our data reveal a novel mechanism of c-SRC regulation whereby in response to stress c-SRC activity is regulated, at least in part, through loss of the interaction with its inhibitor, PR55γ

Evidence that the Nijmegen breakage syndrome protein, an early sensor of double-strand DNA breaks (DSB), is involved in HIV-1 post-integration repair by recruiting the ataxia telangiectasia-mutated kinase in a process similar to, but distinct from, cellular DSB repair

Retroviral transduction involves integrase-dependent linkage of viral and host DNA that leaves an intermediate that requires post-integration repair (PIR). We and others proposed that PIR hijacks the host cell double-strand DNA break (DSB) repair pathways. Nevertheless, the geometry of retroviral DNA integration differs considerably from that of DSB repair and so the precise role of host-cell mechanisms in PIR remains unclear. In the current study, we found that the Nijmegen breakage syndrome 1 protein (NBS1), an early sensor of DSBs, associates with HIV-1 DNA, recruits the ataxia telangiectasia-mutated (ATM) kinase, promotes stable retroviral transduction, mediates efficient integration of viral DNA and blocks integrase-dependent apoptosis that can arise from unrepaired viral-host DNA linkages. Moreover, we demonstrate that the ATM kinase, recruited by NBS1, is itself required for efficient retroviral transduction. Surprisingly, recruitment of the ATR kinase, which in the context of DSB requires both NBS1 and ATM, proceeds independently of these two proteins. A model is proposed emphasizing similarities and differences between PIR and DSB repair. Differences between the pathways may eventually allow strategies to block PIR while still allowing DSB repair
