    A new Rhus gall aphid species Nurudea zhengii Ren, sp. nov. collected from the Mountain Qixing in Shangrao County, Jiangxi Province, China is described and illustrated from alate viviparous female. The new species differs from the other Nurudea species in the length and proportion of antennal segments, the structure of antennal secondary sensilla, and the flower-like shape of the galls that are formed on its primary host. Its primary host plant is Rhus hypoleuca, whereas other Nurudea species are on R. chinensis

    Research on the Determination of Cadmium in Grain by Rapid Digestion-Standard Addition

    Rapid digestion-standard addition based on graphite furnace atomic absorption spectrometry(GFAAS) for determining cadmium in grain of four kinds of matrix (brown rice, corn, wheat and rice) is established, by studying on digestion conditions, optimizing the proportion of sample and nitric acid, and using digested sample to prepare standard solution. The results show that the correlation coefficients are 0.995 to 0.999. The detection limit of method is 0.003 mg/kg. The relative error between the test result and the standard value is –3.8% to –1.1%. The standard recoveries of cadmium in low, medium and high gradient are 99.1% to 103.4%, and the RSD values of the standard substances and real samples are 1.2% to 2.7%.Compared with the results of microwave digestion method, the detection values of cadmium have no significant difference (P>0.05). The method have shown advantages in terms of accuracy and precision, and it is a rapid and accurate method to calculate cadmium in large quantities of grain samples

    <p>Abstract</p> <p>Background</p> <p>Non-coding RNA molecules, such as microRNAs, may play an important role in carcinogenesis. Recent studies have indicated that microRNAs are involved in initiation and progression of various malignancies. However, little work has been done to compare the microRNA expression patterns in oral cancer. In this study, we constructed an animal model of oral squamous cell carcinoma to investigate expression profiles of microRNAs in oral carcinogenesis.</p> <p>Methods</p> <p>The animal model of oral squamous cell carcinoma was conducted by tri-weekly (Monday, Wednesday, and Friday) painting with 5% DMBA in acetone. Six Syrian hamsters, including three from the treated group and three from the control group, were used as a training group for microRNA microarray analysis. All microarray data were analyzed by Significance Analysis of Microarrays (SAM) and CLUSTER 3.0 software, and this result was further confirmed by qRT-PCR assay.</p> <p>Results</p> <p>Seventeen microRNAs were differentially expressed in oral squamous cell carcinoma. Five microRNAs (hsa-miR-21, hsa-miR-200b, hsa-miR-221, hsa-miR-338, and mmu-miR-762) were significantly upregulated and twelve microRNAs (hsa-miR-16, hsa-miR-26a, hsa-miR-29a, hsa-miR-124a, hsa-miR-125b, mmu-miR-126-5p, hsa-miR-143, hsa-miR-145, hsa-miR-148b, hsa-miR-155, hsa-miR-199a, and hsa-miR-203) were down-regulated in cancer tissues. The expression levels of hsa-miR-21 and hsa-miR-16 seen with Stem-loop qRT-PCR were also seen in microarray analysis in all samples.</p> <p>Conclusion</p> <p>Our findings identified specific microRNA expression in oral squamous cell carcinoma and suggested that microRNAs have a role in oral carcinogenesis.</p

    Essential oil from Chenopodium ambrosioides L. induces mitochondrial-mediated pathway and endoplasmic reticulum stress-related apoptosis in human liver cancer SMMC-7721 cells

    Purpose: To evaluate the cytotoxic effect of essential oil derived from Chenopodium ambrosioides L. in Sichuan Province on human liver cancer SMMC-7721 cells, as well as its possible molecular mechanisms.Methods: Cytotoxicity was characterized by MTT assay and transmission electron microscopy (TEM) of SMMC-7721 cells ultrastructure. The apoptotic effect of the essential oil was evaluated by changes in mitochondrial membrane potential and Western blot assay.Results: MTT assay data indicate that the essential oil was cytotoxic to SMMC-7721 cells, while TEN revealed that there were vacuoles and nucleus fragmentation in the SMMC-7721 cell cytosol, cell swelling, and a large amount of leakage. Mitochondrial membrane potential assay and Western Blot data indicate that the essential oil induced cell apoptosis.Conclusion: The essential oil of Chenopodium ambrosioides L. in Sichuan Province seems to induce apoptosis of human liver cancer SMMC-7721 cells via the mitochondrial-mediated pathway and endoplasmic reticulum stress. Thus, this plant requires further investigation as a potential source of ananti-liver cancer drug.     Clinical Study of Restless Leg Syndrome Accompanied by Psychological Symptoms Induced by High-Dose Treatment With Madopar

    Objectives: Some neurological disorders demonstrate indistinguishable psychological symptoms at an early stage, especially when accompanied by jitters similar to those in Parkinson’s disease. During dopamine replacement therapy, some patients display restless leg syndrome (RLS)-like symptoms. Therefore, we aimed to analyze treatment strategies and the prognosis of RLS caused by high-dose Madopar.Methods: Nine patients who were misdiagnosed with Parkinson’s disease, taking a high dose of Madopar, and showed symptoms of anxiety, depression, and somatization were recruited. Clinical data were collected, and strategies of treatment and prognosis were analyzed.Results: Seven patients demonstrated varying degrees of anxiety and depression, and the other two cases were misdiagnosed as Parkinson’s disease. During Madopar treatment, patients gradually showed aggravated symptoms, including swelling, numbness, pain, and other sensory abnormalities in both lower extremities, which spread to both upper extremities in a few patients. Among the seven patients, symptoms of anxiety, depression, insomnia, and somatization significantly worsened during the observation period. The average time from taking Madopar to the appearance of RLS was 2.6 ± 0.6 months, the average time to clinical diagnosis was 18.17 ± 9.40 months, and the average dosage of Madopar was 1.44 ± 0.21 g per day. Gradually reducing the Madopar dosage and administering a small dose of long-acting dopamine preparation greatly alleviated the symptoms after 3 months.Conclusion: A high dose of Madopar can cause RLS-like symptoms accompanied by anxiety, depression, insomnia, and other mental health symptoms. These symptoms should be more closely monitored by clinicians

    Synthesis of microporous silica nanoparticles to study water phase transitions by vibrational spectroscopy

    Silica can take many forms, and its interaction with water can change dramatically at the interface. Silica based systems offer a rich tapestry to probe the confinement of water as size and volume can be controlled by various templating strategies and synthetic procedures. To this end, microporous silica nanoparticles have been developed by a reverse microemulsion method utilizing zinc nanoclusters encapsulated in hydroxyl-terminated polyamidoamine (PAMAM-OH) dendrimers as a soft template. These nanoparticles were made tunable within the outer diameter range of 20-50 nm with a core mesopore of 2-15 nm. Synthesized nanoparticles were used to study the effects of surface area and microporous volumes on the vibrational spectroscopy of water. These spectra reveal contributions from bulk interfacial/interparticle water, ice-like surface water, liquid-like water, and hydrated silica surfaces suggesting that microporous silica nanoparticles allow a way to probe silica water interactions at the molecular scale

    Integrated application of uniform design and least-squares support vector machines to transfection optimization

    <p>Abstract</p> <p>Background</p> <p>Transfection in mammalian cells based on liposome presents great challenge for biological professionals. To protect themselves from exogenous insults, mammalian cells tend to manifest poor transfection efficiency. In order to gain high efficiency, we have to optimize several conditions of transfection, such as amount of liposome, amount of plasmid, and cell density at transfection. However, this process may be time-consuming and energy-consuming. Fortunately, several mathematical methods, developed in the past decades, may facilitate the resolution of this issue. This study investigates the possibility of optimizing transfection efficiency by using a method referred to as least-squares support vector machine, which requires only a few experiments and maintains fairly high accuracy.</p> <p>Results</p> <p>A protocol consists of 15 experiments was performed according to the principle of uniform design. In this protocol, amount of liposome, amount of plasmid, and the number of seeded cells 24 h before transfection were set as independent variables and transfection efficiency was set as dependent variable. A model was deduced from independent variables and their respective dependent variable. Another protocol made up by 10 experiments was performed to test the accuracy of the model. The model manifested a high accuracy. Compared to traditional method, the integrated application of uniform design and least-squares support vector machine greatly reduced the number of required experiments. What's more, higher transfection efficiency was achieved.</p> <p>Conclusion</p> <p>The integrated application of uniform design and least-squares support vector machine is a simple technique for obtaining high transfection efficiency. Using this novel method, the number of required experiments would be greatly cut down while higher efficiency would be gained. Least-squares support vector machine may be applicable to many other problems that need to be optimized.</p

    The investigation of status of human parasites in children from Yushu

    目的  调查青海省玉树县震后转移安置学生的肠道寄生虫感染情况。方法  粪便直接涂片法检查虫卵。结果  512份粪便样品中,寄生虫阳性粪便31份,感染率为6.05%。其中,结肠内阿米巴20例,感染率为3.91%;蓝氏贾第鞭毛虫8例,感染率为1.56%;蛔虫3例,感染率0.59%。结论  玉树的学生肠道寄生虫的感染率均不高于全国平均水平。Objective: To investigate the status of human parasites in children who have been evacuated here from Yushu after earthquake. Methods: Detect the eggs from smear feces samples. Results: 512 stool samples were collected and examined by microscope. The total infection rate was 6.05%, where the infection rate for entamoeba coli Grassi was 3.91%, Giardia lamblia stile was 1.56%, Ascaris Lumbricoides was 0.59%. Conclusion: Our results suggest that the infection rate of human parasites is not higher than normal rate in China

    <p>Abstract</p> <p>Background</p> <p>Some UL45 gene function of Herpesvirus was reported. While there was no any report of the duck enteritis virus (DEV) UL45 protein as yet.</p> <p>Results</p> <p>The UL45 gene and des-transmembrane domain of UL45 (named UL45Δ gene, 295-675bp of UL45) of DEV were amplified by PCR and subcloned into the prokaryotic expression vector pET-32a(+). The constructed recombinant plasmids were transformed into the host strain BL21(DE3) PLysS and induced by IPTG. SDS-PAGE analysis showed the UL45 gene couldn't express while UL45Δ gene was highly expressed. His Purify Kit or salting-out could purify the protein effectively. Using the purified protein to immunize New-Zealand rabbits and produce polyclonal antibody. The agar diffusion reaction showed the titer of antibody was 1:32. Western blot analysis indicated the purified rabbit anti-UL45Δ IgG had a high level of specificity and the UL45 gene was a part of DEV genome. The transcription phase study of UL45 gene showed that expression of UL45 mRNA was at a low level from 0 to 18 h post-infection (pi), then accumulated quickly at 24 h pi and peaked at 42 h pi. It can be detected till 72 h pi. Besides, western blot analysis of purified virion and different viral ingredients showed that the UL45 protein resided in the purified virion and the viral envelope.</p> <p>Conclusions</p> <p>The rabbit anti-UL45Δ IgG was produced successfully and it can serve as a good tool for penetrating studies of the function of DEV UL45 protein. The transcription phase and protein characteristics analysis indicated that DEV UL45 gene was a late gene and UL45 protein may be a viral envelope protein.</p
