    : Galangal (Alpinia galanga) is one of the plants that is often used for cancer therapy. Galangal contains different active ingredients, one of which is 1 'acetoxy chavicol acetate (ACA) which serves as an anticancer through its action as an anti-inflammatory agent, induction of apoptosis and inhibition of proliferation. Benzo(a)pyrenes (BAP) are five-ringed Polycyclic Aromatic Hydrocarbons (PAHs), a group of mutagenic and carcinogenic organic compounds. This study aimed to determine the effect of galangal extract on histological features of mice's breast induced by benzo(a)pyrene. This was an experimental laboratory study using mice as experimental animals. Subjects were 15 mice, divided into negative control group (5 mice) and two treatment groups (5 mice each). All mice were fed with standard pellet throughout the experiment. Mice in group A (negative control) received no treatment; mice in group B were administered with benzo(a)pyrene for 14 days then were given no treatment for the next 14 days; and mice in group C were induced by benzo(a)pyrene and were given galangal extract for 14 days then were given no treatment for the next 14 days. The result of this study revealed changes in mice breast histological features in the form of hyperplasia of cuboid epithelial cells of lactiferous ducts in group B and C, however the manifestation in group C was less pronounced than that was seen in group B. Conclusion: Oral administration of galangal extract subcutaneously was able to inhibit the cuboidal ductal lactiferious epithelial cell hyperplasia of mice breast induced by benzo(a)pyrene

    UAS in the NAS: Survey Responses by ATC, Manned Aircraft Pilots, and UAS Pilots

    NASA currently is working with industry and the Federal Aviation Administration (FAA) to establish future requirements for Unmanned Aircraft Systems (UAS) flying in the National Airspace System (NAS). To work these issues NASA has established a multi-center UAS Integration in the NAS project. In order to establish Ground Control Station requirements for UAS, the perspective of each of the major players in NAS operations was desired. Three on-line surveys were administered that focused on Air Traffic Controllers (ATC), pilots of manned aircraft, and pilots of UAS. Follow-up telephone interviews were conducted with some survey respondents. The survey questions addressed UAS control, navigation, and communications from the perspective of small and large unmanned aircraft. Questions also addressed issues of UAS equipage, especially with regard to sense and avoid capabilities. From the ATC and military ATC perspective, of particular interest is how mixed-operations (manned/UAS) have worked in the past and the role of aircraft equipage. Knowledge gained from this information is expected to assist the NASA UAS in the NAS project in directing research foci thus assisting the FAA in the development of rules, regulations, and policies related to UAS in the NAS

    Small Unmanned Aircraft Systems Integration into the National Airspace System Visual-Line-of-Sight Human-in-the-Loop Experiment

    As part of the Unmanned Aircraft Systems (UAS) in the National Airspace System (NAS) project, research on integrating small UAS (sUAS) into the NAS was underway by a human-systems integration (HSI) team at the NASA Langley Research Center. Minimal to no research has been conducted on the safe, effective, and efficient manner in which to integrate these aircraft into the NAS. sUAS are defined as aircraft weighing 55 pounds or less. The objective of this human system integration team was to build a UAS Ground Control Station (GCS) and to develop a research test-bed and database that provides data, proof of concept, and human factors guidelines for GCS operations in the NAS. The objectives of this experiment were to evaluate the effectiveness and safety of flying sUAS in Class D and Class G airspace utilizing manual control inputs and voice radio communications between the pilot, mission control, and air traffic control. The design of the experiment included three sets of GCS display configurations, in addition to a hand-held control unit. The three different display configurations were VLOS, VLOS + Primary Flight Display (PFD), and VLOS + PFD + Moving Map (Map). Test subject pilots had better situation awareness of their vehicle position, altitude, airspeed, location over the ground, and mission track using the Map display configuration. This configuration allowed the pilots to complete the mission objectives with less workload, at the expense of having better situation awareness of other aircraft. The subjects were better able to see other aircraft when using the VLOS display configuration. However, their mission performance, as well as their ability to aviate and navigate, was reduced compared to runs that included the PFD and Map displays

    Methylation-Specific MLPA (MS-MLPA): simultaneous detection of CpG methylation and copy number changes of up to 40 sequences

    Copy number changes and CpG methylation of various genes are hallmarks of tumor development but are not yet widely used in diagnostic settings. The recently developed multiplex ligation-dependent probe amplification (MLPA) method has increased the possibilities for multiplex detection of gene copy number aberrations in a routine laboratory. Here we describe a novel robust method: the methylation-specific MLPA (MS-MLPA) that can detect changes in both CpG methylation as well as copy number of up to 40 chromosomal sequences in a simple reaction. In MS-MLPA, the ligation of MLPA probe oligonucleotides is combined with digestion of the genomic DNA–probe hybrid complexes with methylation-sensitive endonucleases. Digestion of the genomic DNA–probe complex, rather than double-stranded genomic DNA, allowed the use of DNA derived from the formalin treated paraffin-embedded tissue samples, enabling retrospective studies. To validate this novel method, we used MS-MLPA to detect aberrant methylation in DNA samples of patients with Prader–Willy syndrome, Angelman syndrome or acute myeloid leukemia

    Characteristics of contralateral carcinomas in patients with differentiated thyroid cancer larger than 1 cm

    Purpose: Traditionally, total thyroidectomy has been advocated for patients with tumors larger than 1 cm. However, according to the ATA and NCCN guidelines (2015, USA), patients with tumors up to 4 cm are now eligible for lobectomy. A rationale for adhering to total thyroidectomy might be the presence of contralateral carcinomas. The purpose of this study was to describe the characteristics of contralateral carcinomas in patients with differentiated thyroid cancer (DTC) larger than 1 cm. Methods: A retrospective study was performed including patients from 17 centers in 5 countries. Adults diagnosed with DTC stage T1b-T3 N0-1a M0 who all underwent a total thyroidectomy were included.     Intragenic deletions and a deep intronic mutation affecting pre-mRNA splicing in the dihydropyrimidine dehydrogenase gene as novel mechanisms causing 5-fluorouracil toxicity

    Dihydropyrimidine dehydrogenase (DPD) is the initial enzyme acting in the catabolism of the widely used antineoplastic agent 5-fluorouracil (5FU). DPD deficiency is known to cause a potentially lethal toxicity following administration of 5FU. Here, we report novel genetic mechanisms underlying DPD deficiency in patients presenting with grade III/IV 5FU-associated toxicity. In one patient a genomic DPYD deletion of exons 21–23 was observed. In five patients a deep intronic mutation c.1129–5923C>G was identified creating a cryptic splice donor site. As a consequence, a 44 bp fragment corresponding to nucleotides c.1129–5967 to c.1129–5924 of intron 10 was inserted in the mature DPD mRNA. The deleterious c.1129–5923C>G mutation proved to be in cis with three intronic polymorphisms (c.483 + 18G>A, c.959–51T>G, c.680 + 139G>A) and the synonymous mutation c.1236G>A of a previously identified haplotype. Retrospective analysis of 203 cancer patients showed that the c.1129–5923C>G mutation was significantly enriched in patients with severe 5FU-associated toxicity (9.1%) compared to patients without toxicity (2.2%). In addition, a high prevalence was observed for the c.1129–5923C>G mutation in the normal Dutch (2.6%) and German (3.3%) population. Our study demonstrates that a genomic deletion affecting DPYD and a deep intronic mutation affecting pre-mRNA splicing can cause severe 5FU-associated toxicity. We conclude that screening for DPD deficiency should include a search for genomic rearrangements and aberrant splicing
