    Regeneration of okra (Abelmoschus esculentus L.) via apical shoot culture system

    The present study was undertaken to evaluate the most suitable concentration of growth regulators for regeneration of okra (Abelmoschus esculentus L. Monech) via apical shoot culture system. The study of apical shoot culture system was found effective for regeneration of apical shoots. The okra (A. esculentus L. Monech) N-550 line evolved at R&D, Nirmal Seeds Pvt. Ltd., was used as basic material for experiment of regeneration using different auxin and cytokinin hormone combination. Nine to ten days old germinating seedlings were used for isolation of shoot tip explants, isolated shoot tips were cultured on regeneration medium, Murashige and Skoog (MS) medium supplemented with alone indole-3-butyric acid (IBA) (0.25, 0.5, 1.00, 1.5, 2.0 and 2.5 mg/L) and IBA 1.0 mg/L with naphthalene acetic acid (NAA) (0.25, 0.5, 1.00, 1.5, 2.0 and 2.5 mg/L). After regeneration, cultures were inoculated on elongation medium containing MS medium supplemented with kinetin (0.25, 0.5, 1.00, 1.5, 2.0 and 2.5 mg/L). Elongated shoots transferred to rooting medium containing MS medium supplemented with indole-3-butyric acid (IAA) (0.25, 0.5, 1.00, 1.5, 2.0 and 2.5 mg/L). Combination of 1.0 mg/L IBA and 0.5 mg/L NAA were found to be most effective for plant regeneration from apical shoot. Best shoot elongation observed in MS medium supplemented with kinetin 0.5 mg/L. Elongated shoots rooted most effectively in MS medium containing 0.5 mg/L IAA and 1.0 g activated charcoal. The success of apical shoot culture system of okra was encouraged by acclimatization of the plantlets in the field conditions.Key words: Abelmoschus esculentus, apical shoot, auxins, cytokinines, regeneratio

    Potential of optimized NOvA for large theta(13) & combined performance with a LArTPC & T2K

    NOvA experiment has reoptimized its event selection criteria in light of the recently measured moderately large value of theta(13). We study the improvement in the sensitivity to the neutrino mass hierarchy and to leptonic CP violation due to these new features. For favourable values of deltacp, NOvA sensitivity to mass hierarchy and leptonic CP violation is increased by 20%. Addition of 5 years of neutrino data from T2K to NOvA more than doubles the range of deltacp for which the leptonic CP violation can be discovered, compared to stand alone NOvA. But for unfavourable values of deltacp, the combination of NOvA and T2K are not enough to provide even a 90% C.L. hint of hierarchy discovery. Therefore, we further explore the improvement in the hierarchy and CP violation sensitivities due to the addition of a 10 kt liquid argon detector placed close to NOvA site. The capabilities of such a detector are equivalent to those of NOvA in all respects. We find that combined data from 10 kt liquid argon detector (3 years of nu + 3 years of nubar run), NOvA (6 years of nu + 6 years of nubar run) and T2K (5 years of nu run) can give a close to 2 sigma hint of hierarchy discovery for all values of deltacp. With this combined data, we can achieve CP violation discovery at 95% C.L. for roughly 60% values of deltacp.Comment: 22 pages, 24 pdf figures, 5 tables.     Practical computational toolkits for dendrimers and dendrons structure design

    Dendrimers and dendrons offer an excellent platform for developing novel drug delivery systems and medicines. The rational design and further development of these repetitively branched systems are restricted by difficulties in scalable synthesis and structural determination, which can be overcome by judicious use of molecular modelling and molecular simulations. A major difficulty to utilise in silico studies to design dendrimers lies in the laborious generation of their structures. Current modelling tools utilise automated assembly of simpler dendrimers or the inefficient manual assembly of monomer precursors to generate more complicated dendrimer structures. Herein we describe two novel graphical user interface (GUI) toolkits written in Python that provide an improved degree of automation for rapid assembly of dendrimers and generation of their 2D and 3D structures. Our first toolkit uses the RDkit library, SMILES nomenclature of monomers and SMARTS reaction nomenclature to generate SMILES and mol files of dendrimers without 3D coordinates. These files are used for simple graphical representations and storing their structures in databases. The second toolkit assembles complex topology dendrimers from monomers to construct 3D dendrimer structures to be used as starting points for simulation using existing and widely available software and force fields. Both tools were validated for ease-of-use to prototype dendrimer structure and the second toolkit was especially relevant for dendrimers of high complexity and size.Peer reviewe

    Characterization of the Influenza A H5N1 Viruses of the 2008-09 Outbreaks in India Reveals a Third Introduction and Possible Endemicity

    Widespread infection of highly pathogenic avian influenza A H5N1 was reported from backyard and commercial poultry in West Bengal (WB), an eastern state of India in early 2008. Infection gradually spread to Tripura, Assam and Sikkim, the northeastern states, with 70 outbreaks reported between January 2008 and May 2009. Whole genome sequence analysis of three isolates from WB, one isolate from Tripura along with the analysis of hemagglutinin (HA) and neuraminidase (NA) genes of 17 other isolates was performed during this study. In the HA gene phylogenetic tree, all the 2008-09 Indian isolates belonged to EMA3 sublineage of clade 2.2. The closest phylogenetic relationship was found to be with the 2007-09 isolates from Bangladesh and not with the earlier 2006 and 2007 Indian isolates implying a third introduction into the country. The receptor-binding pocket of HA1 of two isolates from WB showed S221P mutation, one of the markers predicted to be associated with human receptor specificity. Two substitutions E119A (2 isolates of WB) and N294S (2 other isolates of WB) known to confer resistance to NA inhibitors were observed in the active site of neuraminidase. Several additional mutations were observed within the 2008-09 Indian isolates indicating genetic diversification. Overall, the study is indicative of a possible endemicity in the eastern and northeastern parts of the country, demanding active surveillance specifically in view of the critical mutations that have been observed in the influenza A H5N1 viruses

    Over the past 20 years, research on the physiology of swallowing has confirmed that the oropharyngeal swallowing process can be modulated, both volitionally and in response to different sensory stimuli. In this review we identify what is known regarding the sensory pathways and mechanisms that are now thought to influence swallowing motor control and evoke its response. By synthesizing the current state of research evidence and knowledge, we identify continuing gaps in our knowledge of these mechanisms and pose questions for future research

    Comprehensive and Integrated Genomic Characterization of Adult Soft Tissue Sarcomas

    Summary Sarcomas are a broad family of mesenchymal malignancies exhibiting remarkable histologic diversity. We describe the multi-platform molecular landscape of 206 adult soft tissue sarcomas representing 6 major types. Along with novel insights into the biology of individual sarcoma types, we report three overarching findings: (1) unlike most epithelial malignancies, these sarcomas (excepting synovial sarcoma) are characterized predominantly by copy-number changes, with low mutational loads and only a few genes (TP53, ATRX, RB1) highly recurrently mutated across sarcoma types; (2) within sarcoma types, genomic and regulomic diversity of driver pathways defines molecular subtypes associated with patient outcome; and (3) the immune microenvironment, inferred from DNA methylation and mRNA profiles, associates with outcome and may inform clinical trials of immune checkpoint inhibitors. Overall, this large-scale analysis reveals previously unappreciated sarcoma-type-specific changes in copy number, methylation, RNA, and protein, providing insights into refining sarcoma therapy and relationships to other cancer types

    Arctic tropospheric ozone: assessment of current knowledge and model performance

    As the third most important greenhouse gas (GHG) after carbon dioxide (CO2) and methane (CH4), tropospheric ozone (O3) is also an air pollutant causing damage to human health and ecosystems. This study brings together recent research on observations and modeling of tropospheric O3 in the Arctic, a rapidly warming and sensitive environment. At different locations in the Arctic, the observed surface O3 seasonal cycles are quite different. Coastal Arctic locations, for example, have a minimum in the springtime due to O3 depletion events resulting from surface bromine chemistry. In contrast, other Arctic locations have a maximum in the spring. The 12 state-of-the-art models used in this study lack the surface halogen chemistry needed to simulate coastal Arctic surface O3 depletion in the springtime; however, the multi-model median (MMM) has accurate seasonal cycles at non-coastal Arctic locations. There is a large amount of variability among models, which has been previously reported, and we show that there continues to be no convergence among models or improved accuracy in simulating tropospheric O3 and its precursor species. The MMM underestimates Arctic surface O3 by 5 % to 15 % depending on the location. The vertical distribution of tropospheric O3 is studied from recent ozonesonde measurements and the models. The models are highly variable, simulating free-tropospheric O3 within a range of ±50 % depending on the model and the altitude. The MMM performs best, within ±8 % for most locations and seasons. However, nearly all models overestimate O3 near the tropopause (∼300 hPa or ∼8 km), likely due to ongoing issues with underestimating the altitude of the tropopause and excessive downward transport of stratospheric O3 at high latitudes. For example, the MMM is biased high by about 20 % at Eureka. Observed and simulated O3 precursors (CO, NOx, and reservoir PAN) are evaluated throughout the troposphere. Models underestimate wintertime CO everywhere, likely due to a combination of underestimating CO emissions and possibly overestimating OH. Throughout the vertical profile (compared to aircraft measurements), the MMM underestimates both CO and NOx but overestimates PAN. Perhaps as a result of competing deficiencies, the MMM O3 matches the observed O3 reasonably well. Our findings suggest that despite model updates over the last decade, model results are as highly variable as ever and have not increased in accuracy for representing Arctic tropospheric O3

    Scintillation light detection in the 6-m drift-length ProtoDUNE Dual Phase liquid argon TPC

    DUNE is a dual-site experiment for long-baseline neutrino oscillation studies, neutrino astrophysics and nucleon decay searches. ProtoDUNE Dual Phase (DP) is a 6  ×  6  ×  6 m 3 liquid argon time-projection-chamber (LArTPC) that recorded cosmic-muon data at the CERN Neutrino Platform in 2019-2020 as a prototype of the DUNE Far Detector. Charged particles propagating through the LArTPC produce ionization and scintillation light. The scintillation light signal in these detectors can provide the trigger for non-beam events. In addition, it adds precise timing capabilities and improves the calorimetry measurements. In ProtoDUNE-DP, scintillation and electroluminescence light produced by cosmic muons in the LArTPC is collected by photomultiplier tubes placed up to 7 m away from the ionizing track. In this paper, the ProtoDUNE-DP photon detection system performance is evaluated with a particular focus on the different wavelength shifters, such as PEN and TPB, and the use of Xe-doped LAr, considering its future use in giant LArTPCs. The scintillation light production and propagation processes are analyzed and a comparison of simulation to data is performed, improving understanding of the liquid argon properties
