    Peptide characterization of mature fluorotic and control human enamel

    Exposure to high fluoride levels during amelogenesis causes enamel fluorosis. This study aimed to determine and compare the amino acid sequences in the enamel of fluorotic and control teeth. This investigation included enamel samples obtained from erupted and non-erupted third molars with either TF grade 4-6 (n=7) fluorosis or no sign of fluorosis (controls, n=7). The samples were kept frozen at -20 °C until protein extraction. Samples were etched and processed with a cocktail of proteinase inhibitors and immediately analyzed. Matrix Assisted Laser Desorption/Ionization-Time-Of-Flight/Time-of-Flight Mass Spectrometry (MALDI-TOF/TOF) followed by MASCOT search aided the peptides analysis. The more abundant peptides bore the N-terminal amelogenin sequences WYQSIRPPYP (which is specific for the X-encoded amelogenin) and MPLPPHPGHPGYINF (which does not show sexual dimorphism) were not different in control or fluorotic enamel. There was no missing proteolytic cleavage in the fluorotic samples, which suggested that the increased amount of protein described in fluorotic enamel did not stem from the decreased ability of proteinases to cleave the proteins in humans. This study showed how to successfully obtain peptide from superficial enamel. Nenhuma alteração na proteólise ocorreu nas amostras fluoróticas, o que sugere que o aumento na quantidade de proteínas existentes nas amostras fluoróticas não está correlacionada a habilidade das proteinases em clivar as proteínas em humanos.     Effect of biofunctionalized implant surface on osseointegration: a histomorphometric study in dogs

    Among the different properties that influence bone apposition around implants, the chemical or biochemical composition of implant surface may interfere on its acceptance by the surrounding bone. The aim of this study was to investigate if a biofunctionalization of implant surface influences the bone apposition in a dog model and to compare it with other surfaces, such as a microstructured created by the grit-blasting/acid-etching process. Eight young adult male mongrel dogs had the bilateral mandibular premolars extracted and each one received 6 implants after 12 weeks, totaling 48 implants in the experiment. Four groups of implants were formed with the same microrough topography with or without some kind of biofunctionalization treatment. After histomorphometric analysis, it was observed that the modified microstructured surface with a "low concentration of the bioactive peptide" provided a higher adjacent bone density (54.6%) when compared to the other groups (microstructured + HA coating = 46.0%, microstructured only = 45.3% and microstructured + "high concentration of the bioactive peptide" = 40.7%), but this difference was not statistically significant. In conclusion, biofunctionalization of the implant surface might interfere in the bone apposition around implants, especially in terms of bone density. Different concentrations of bioactive peptide lead to different results.Entre as diferentes propriedades de uma superfície capazes de influenciar a deposição óssea ao redor de implantes, a composição química e bioquímica pode atuar no reconhecimento do tecido ósseo circundante. O presente trabalho investigou a influência da biofuncionalização de superfícies de implante na deposição óssea ao redor dos mesmos em um modelo animal, comparando-as com outras superfícies, como a microtexturizada obtida pelo processo de jateamento e ataque ácido. Metodologicamente, os pré-molares mandibulares bilaterais de 8 cães foram extraídos e após 12 semanas foram instalados 6 implantes em cada cão, constituindo uma amostra de 48 implantes. Dos 4 grupos experimentais de diferentes superfícies, todos continham a mesma microtopografia rugosa, porém possuindo ou não alguma biofuncionalização. A análise histomorfométrica revelou que a superfície microtexturizada modificada pela adição de baixa concentração peptídica obteve uma maior densidade óssea adjacente (54,6%) quando comparada aos outros grupos (microtexturizada + HA = 46%, somente microtexturizada = 45,3% e microtexturizada com adição de alta concentração peptídica = 40,7%), no entanto estas diferenças numéricas não foram estatisticamente significantes. Dentro deste contexto, conclui-se que a biofuncionalização da superfície de implantes pode interferir na aposição óssea, em particular na densidade óssea, e que diferentes concentrações peptídicas podem conduzir a diferentes resultados.FAPES

    Iron-binding properties of sugar cane yeast peptides

    AbstractThe extract of sugar-cane yeast (Saccharomyces cerevisiae) was enzymatically hydrolysed by Alcalase, Protex or Viscozyme. Hydrolysates were fractionated using a membrane ultrafiltration system and peptides smaller than 5kDa were evaluated for iron chelating ability through measurements of iron solubility, binding capacity and dialyzability. Iron-chelating peptides were isolated using immobilized metal affinity chromatography (IMAC). They showed higher content of His, Lys, and Arg than the original hydrolysates. In spite of poor iron solubility, hydrolysates of Viscozyme provided higher iron dialyzability than those of other enzymes. This means that more chelates of iron or complexes were formed and these kept the iron stable during simulated gastro-intestinal digestion in vitro, improving its dialyzability

    Brazilian community restaurants’ low-income food handlers : association between the nutritional status and the presence of non-communicable chronic diseases

    This cross-sectional study aimed primarily to determine the association between the nutritional status and the presence of non-communicable chronic diseases (NCDs) among community restaurants’ food handlers, since their access to food can influence their body mass index (BMI). The study discusses the socio-demographic status of participants, dietary intake, the prevalence of overweightness/obesity, and self-reported diagnosed NCDs. In 36 Community Restaurants (CRs) from all of the Brazilian regions, we collected data from 559 food handlers. We used a questionnaire to collect socio-demographic data and the reported diagnosed chronic diseases. For the anthropometric evaluation with Body Mass Index calculation, we measured the weight and the height of the individuals. They were all weighed before having lunch at the CR, without shoes and coats. Associations between variables were analyzed by the chi-square test and Poisson regression at a significance level of 5%, considering health as the outcome variable. Most of the food handlers were female (63.1%), married or with a partner (51.7%), and overweight (59.9%). Among the food handlers that presented diagnosed NCDs (n = 96, 17.2% of food handlers), 45.8% (n = 44) presented hypertension and 12.5% (n = 12) type 2 diabetes mellitus. There was a significant association between BMI and NCD status in the studied population. The total daily sodium intake of food handlers was higher than the recommendations of the World Health Organization (WHO), especially from the CR lunch, which may raise the risk of chronic diseases such as hypertension (the most prevalent non-communicable disease found in our study). Despite that, in general, the CRs provide access to cheap and adequate meals to their workers, considering energy intake and the proportion of macronutrients. In this population, overweightness and obesity were prevalent; there was an association of obesity with chronic disease in the study population. Therefore, it is necessary for better menu planning for CRs to guarantee sodium reduction throughout time

    Factors affecting the retention efficiency and physicochemical properties of spray dried lipid nanoparticles loaded with lippia sidoides essential oil

    Essential oils (EOs) are widely used in various industrial sectors but can present several instability problems when exposed to environmental factors. Encapsulation technologies are effective solutions to improve EOs properties and stability. Currently, the encapsulation in lipid nanoparticles has received significant attention, due to the several recognized advantages over conventional systems. The study aimed to investigate the influence of the lipid matrix composition and spray-drying process on the physicochemical properties of the lipid-based nanoparticles loaded with Lippia sidoides EO and their retention efficiency for the oil. The obtained spray-dried products were characterized by determination of flow properties (Carr Index: from 25.0% to 47.93%, and Hausner ratio: from 1.25 to 1.38), moisture (from 3.78% to 5.20%), water activity (<0.5), and powder morphology. Zeta potential, mean particle size and polydispersity index, of the redispersed dried product, fell between 25.9 mV and 30.9 mV, 525.3 nm and 1143 nm, and 0.425 and 0.652, respectively; showing slight differences with the results obtained prior to spray-drying (from 16.4 mV to 31.6 mV; 147 nm to 1531 nm; and 0.459 to 0.729).     Spouted bed dried Rosmarinus officinalis extract: a novel approach for physicochemical properties and antioxidant activity

    In this study, a conical-cylindrical spouted bed dryer with Teflon® beads as spouting material was used for producing powdered rosemary (Rosmarinus officinalis L.) extract. The influence of the inlet drying gas temperature (Tgi) and the percentage ratio between the feed rate of concentrated liquid extract by the maximum evaporation capacity of the spouted bed (Ws/Wmax) on selected physicochemical properties of the finished products were investigated. Antioxidant properties of the concentrated liquid extract and dried extracts were also evaluated by the 2.2-diphenyl-1-picrylhydrazyl radical scavenging (DPPH) and lipid peroxidation induced by Fe2+/citrate (LPO) methods; and compared with the values obtained for a lyophilized extract (used as a control). Colloidal silicon dioxide (Tixosil® 333) and maltodextrin (DE 14) at a 2:1 ratio was added to the concentrated extract before drying (4.4% w/w) to improve the drying performance. The drying variables Ws/Wmax and Tgi have statistically significant influence on total polyphenols and total flavonoid contents of the dried powders. The concentrated extract (on dry basisbeing absolute solid content) showed superior antioxidant activity (AA) compared to both the spouted bed dried and the lyophilized extracts; exhibiting IC50 values of 0.96 ± 0.02, 2.16 ± 0.04 and 3.79 ± 0.05 µg mL1 (DPPH method) and 0.22 ± 0.01, 1.31± 0.01 and 2.54 ± 0.02 µg mL1 (LPO method), respectively. These results of AA are comparable to values obtained for quercetin, a flavonoid compound often used as a reference standard due to its potent antioxidant activity; with IC50 of 1.17 µg mL1 (DPPH) and 0.22 µg mL1 (LPO). However, the dried rosemary extracts are about 13.5 times more concentrated than the initial concentrated extract (dry weight), with a concentration of total flavonoids and polyphenols compounds ranging from 4.3 to 12.3 and from 1.2 to 4.7 times higher than the concentrated extract values (wet basis).     Estudo da partição de fitase produzida por Aspergillus niger var. phoenicis utilizando bioconversão extrativa em sistemas de duas fases aquosas PEG/citrato

    Existem diversos métodos tradicionais que são utilizados para extratir biomoléculas produzidas por fermentação convencional. Um método alternativo é o sistema de duas fases aquosas, o qual foi desenvolvido para a extração de bioprodutos. A bioconversão extrativa trata-se de um sistema de duas fases aquosas que integra cultivo microbiológico à produção e recuperação do bioproduto. Fitases são fosfatases específicas que estão envolvidas na catálise do ácido fitico. O objetivo deste trabalho foi estudar a partição da fitase produzida por A. niger var. phoenicis por bioconversão extrativa utilizando PEG/citrato. Realizou-se um planejamento fatorial completo 25, estudando as seguintes variáveis: massa molar do PEG, concentração do PEG, concentração de citrato, pH e agitação, onde obteve-se como variável-resposta o coeficiente de partição em atividade (KATIV). Neste trabalho conseguiu-se um coeficiente de partição de 25,77 utilizando MPEG (8000 g/mol), CPEG, (26,0% m/m), CCIT (20,0% m/m), pH (6,0) e agitação (100 rpm). Através dos resultados obtivos, pode-se concluir que a fitase utilizada no presente estudo apresenta uma tendência de particionar para a fase superior do sistema (K>1). A técnica de fermentação extrativa utilizando SDFA PEG/citrato demonstrou ser promissora para extração de fitase produzida por A. niger var. phoenicis, podendo ser aplicada na composição de rações comerciais
