    Crystallization and preliminary crystallographic data for the augmenter of liver regeneration

    A new cellular growth factor termed augmenter of liver regeneration (ALR) has been crystallized. ALR has been shown to have a proliferative effect on liver cells while at the same time producing an immunosuppressive effect on liver-resident natural killer cells and liver-resident mononuclear leukocytes. In addition, ALR appears to play an important role in the synthesis and stabilization of mitochondrial gene transcripts inactively regenerating cells. ALR crystals diffract to beyond 2 Ã… resolution and belong to space group P21212, with a = 125.1, b = 108.1 and c = 38.5 Ã…. Based on four molecules per asymmetric unit, the Matthews coefficient is calculated to be 2.16 Ã…3 Da-1 which corresponds to a solvent content of 43%

    Fresnel zone plate telescopes for X-ray imaging II: numerical simulations with parallel and diverging beams

    We present the results of simulations of shadows cast by a zone plate telescope which may have one to four pairs of zone plates. From the shadows we reconstruct the images under various circumstances. We discuss physical basis of the resolution of the telescope and demonstrate this by our simulations. We allow the source to be at a finite distance (diverging beam) as well as at an infinite distance (parallel beam) and show that the resolution is worsened when the source is nearby. By reconstructing the zone plates in a way that both the zone plates subtend the same solid angles at the source, we obtain back high resolution even for sources at a finite distance. We present simulated results for the observation of the galactic center and show that the sources of varying intensities may be reconstructed with accuracy. Results of these simulations would be of immense use in interpreting the X-ray images from recently launched CORONAS-PHOTON satellite.Comment: 17 pages, 36 figures, Published in Experimental Astronom

    Imaging in hard X-rays of any astrophysical source with high angular resolution is a challenging job. Shadow-casting technique is one of the most viable options for imaging in hard X-rays. We have used two different types of shadow-casters, namely, Coded Aperture Mask (CAM) and Fresnel Zone Plate (FZP) pair and two types of pixellated solid-state detectors, namely, CZT and CMOS in RT-2/CZT payload, the hard X-ray imaging instrument onboard the CORONAS-PHOTON satellite. In this paper, we present the results of simulations with different combinations of coders (CAM & FZP) and detectors that are employed in the RT-2/CZT payload. We discuss the possibility of detecting transient Solar flares with good angular resolution for various combinations.     Minimally invasive mitral valve surgery: a systematic review.

    In the recent years minimally invasive mitral valve surgery (MIMVS) has become a well-established and increasingly used option for managing patients with a mitral valve pathology. Nonetheless, whether the purported benefits of MIMVS translate into clinically important outcomes remains controversial. Therefore, in this paper we provide an overview of MIMVS and discuss results, morbidity, mortality, and quality of life following mitral minimally invasive procedures. MIMVS has been proven to be a feasible alternative to the conventional full sternotomy approach with low perioperative morbidity and short-term mortality. Reported benefits of MIMVS include also decreased postoperative pain, improved postoperative respiratory function, reduced surgical trauma, and greater patient satisfaction. Finally, compared to standard surgery, MIMVS demonstrated comparable efficacy across a range of long-term efficacy measures such as freedom from reoperation and long-term survival

    Small Heat Shock Protein αA-Crystallin Prevents Photoreceptor Degeneration in Experimental Autoimmune Uveitis

    The small heat shock protein, αA-crystallin null (αA−/−) mice are known to be more prone to retinal degeneration than the wild type mice in Experimental Autoimmune Uveoretinitis (EAU). In this report we demonstrate that intravenous administration of αA preserves retinal architecture and prevents photoreceptor damage in EAU. Interestingly, only αA and not αB-crystallin (αB), a closely related small heat shock protein works, pointing to molecular specificity in the observed retinal protection. The possible involvement of αA in retinal protection through immune modulation is corroborated by adaptive transfer experiments, (employing αA−/− and wild type mice with EAU as donors and Rag2−/− as the recipient mice), which indicate that αA protects against the autoimmune challenge by modulating the systemic B and T cell immunity. We show that αA administration causes marked reduction in Th1 cytokines (TNF-α, IL-12 and IFN-γ), both in the retina and in the spleen; notably, IL-17 was only reduced in the retina suggesting local intervention. Importantly, expression of Toll-like receptors and their associated adaptors is also inhibited suggesting that αA protection, against photoreceptor loss in EAU, is associated with systemic suppression of both the adaptive and innate immune responses

    Expression and methylation status of tissue factor pathway inhibitor-2 gene in non-small-cell lung cancer

    Tissue factor pathway inhibitor-2 (TFPI-2) is a Kunitz-type serine proteinase inhibitor that inhibits plasmin-dependent activation of several metalloproteinases. Downregulation of TFPI-2 could thus enhance the invasive potential of neoplastic cells in several cancers, including lung cancer. In this study, TFPI-2 mRNA was measured using a real-time PCR method in tumours of 59 patients with non-small-cell lung cancer (NSCLC). Tumour TFPI-2 mRNA levels appeared well correlated with protein expression evaluated by immunohistochemistry and were 4–120 times lower compared to those of nonaffected lung tissue in 22 cases (37%). Hypermethylation of the TFPI-2 gene promoter was demonstrated by restriction enzyme-polymerase chain reaction in 12 of 40 cases of NSCLC (30%), including nine of 17 for whom tumour TFPI-2 gene expression was lower than in noncancerous tissue. In contrast, this epigenetic modification was shown in only three of 23 tumours in which no decrease in TFPI-2 synthesis was found (P=0.016). Decreased TFPI-2 gene expression and hypermethylation were more frequently associated with stages III or IV NSCLC (eight out of 10, P=0.02) and the TFPI-2 gene promoter was more frequently hypermethylated in patients with lymph node metastases (eight out of 16, P=0.02). These results suggest that silencing of the TFPI-2 gene by hypermethylation might contribute to tumour progression in NSCLC

    C2 prosthesis: anterior upper cervical fixation device to reconstruct the second cervical vertebra

    Destruction of the second cervical vertebra leads to a highly unstable situation. Reconstruction is difficult because the axis plays a central role in rotatory movements and has a unique function in redistributing axial loads. The axis transfers the axial load of the two lateral masses of the atlas to three surfaces on the third cervical vertebra: the two articular facets and the vertebral body. As reconstruction is difficult and the instability in this region is life threatening, pathological processes are often treated less radically compared to other areas of the cervical spine. However, this more moderate approach may result in worse outcomes and prognoses. This paper presents the development of a new implant (C2 prosthesis) and two illustrative cases describing the implementation of this new implant. The C2 prosthesis provides anterior support and therefore allows a more radical surgical approach
