An orthologue of bacteroides fragilis NanH is the principal sialidase in tannerella forsythia
Sialidase activity is a putative virulence factor of the anaerobic periodontal pathogen Tannerella forsythia, but it is uncertain which genes encode this activity. Characterization of a putative sialidase, SiaHI, by others, indicated that this protein alone may not be responsible for all of the sialidase activity. We describe a second sialidase in T. forsythia (TF0035), an orthologue of Bacteroides fragilis NanH, and its expression in Escherichia coli. Sialidase activity of the expressed NanH was confirmed by using 2′-(4-methylumbelliferyl)-α-d-N-acetylneuraminic acid as a substrate. Biochemical characterization of the recombinant T. forsythia NanH indicated that it was active over a broad pH range, with optimum activity at pH 5.5. This enzyme has high affinity for 2′-(4-methylumbelliferyl)-α-d-N-acetylneuraminic acid (Km of 32.9 ± 10.3 μM) and rapidly releases 4-methylumbelliferone (Vmax of 170.8 ± 11.8 nmol of 4-methylumbelliferone min−1 mg of protein−1). E. coli lysates containing recombinant T. forsythia NanH cleave sialic acid from a range of substrates, with a preference for α2-3 glycosidic linkages. The genes adjacent to nanH encode proteins apparently involved in the metabolism of sialic acid, indicating that the NanH sialidase is likely to be involved in nutrient acquisition
Marked mitigation of transplant vascular sclerosis in FasL(gld) (CD95L) mutant recipients. I. The role of alloantibodies in the development of chronic rejection
Background. In the acute rejection of allografts, the interaction between Fas (CD95) and its ligand (FasL; CD95L) has been shown to be involved in mediating apoptotic cell death. The role, however, of these molecules in the pathogenesis of transplant vascular sclerosis is as yet undetermined. The present study was therefore designed to address this issue. Material. C3H/HEJ FasL(gld) (FasL-; H2(k)) spontaneously mutant mice were used either as donors or recipients of aortic allografts; wild-type C57BI/6 (B6; H2b) were used as corresponding recipients or donors (n=6/group), respectively. Controls included aortas transplanted across appropriate allogeneic and syngeneic strain combinations. For histopathological evaluations, the grafts were harvested at day 40 after transplantation, at which time, splenocytes and sera were also obtained for mixed leukocyte reaction and complement- mediated microcytotoxicity assays, respectively. Results. Similar to aortas obtained from allogeneic controls, allografts harvested from FasL-→B6 recipients had morphological evidence of chronic rejection characterized by circumferential intimal thickening with partial disruption of the elastic membranes. Correspondingly, heightened antidonor cellular reactivity was also witnessed in these recipients. On the contrary, B6 allografts harvested from the majority of C3H→FasL- recipients exhibited marked preservation of aortic morphology. Although these recipients had diminished antidonor cellular proliferation, the titers of alloantibodies were markedly elevated. Conclusion. The presence of FasL-expressing functional cytotoxic T cells is required for the pathogenesis of transplant vascular sclerosis. The significant reduction and/or absence of chronic rejection with the concomitant retention of antidonor humoral response in C3H FasL- recipients of B6 aortas prompt us to suggest that perhaps posttransplantation vasculopathy is initiated by cell-mediated cytotoxicity with its perpetuation facilitated by alloantibodies
Oxygen isotopic ratios in cool R Coronae Borealis stars
We investigate the relationship between R Coronae Borealis (RCB) stars and
hydrogen-deficient carbon (HdC) stars by measuring precise 16O/18O ratios for
five cool RCB stars. The 16O/18O ratios are derived by spectrum synthesis from
high-resolution (R=50,000) K-band spectra. Lower limits to the 16O/17O and
14N/15}N ratios as well as Na and S abundances (when possible) are also given.
RCB stars in our sample generally display less 18O than HdC stars - the derived
16O/18O ratios range from 3 to 20. The only exception is the RCB star WX CrA,
which seems to be a HdC-like star with 16O/18O=0.3. Our result of a higher
16O/18O ratio for the RCB stars must be accounted for by a theory of the
formation and evolution of HdC and RCB stars. We speculate that a late
dredge-up of products of He-burning, principally 12C and 16O, may convert a
18O-rich HdC star into a 18O-poor RCB star as the H-deficient star begins its
final evolution from a cool supergiant to the top of the white dwarf cooling
track.Comment: Accepted for publication in the Astrophysical Journal; 21 pages, 3

Improved surgical technique for the establishment of a murine model of aortic transplantation.
Aortic allotransplantation is a reliable procedure to study the evolvement of chronic rejection in mice. The progressive nature of this process in mice is characterized by diffuse and concentric myointimal proliferation which is inevitably associated with variable degrees of luminal constriction. These vascular changes are comparable to those that are witnessed in organ allografts undergoing chronic rejection in humans, underscoring its utility as a model of choice for the study of the development of this lesion. Whilst improved surgical technique has resulted in markedly enhanced graft survival, the results are far from being acceptable. Realizing this limitation, we embarked on developing a modified technique for aortic transplantation which would allow for improved graft survival in mice. A bypass conduit was created by end-to-side anastomosis of a segment of the donor's thoracic aorta into the infrarenal portion of the recipient's abdominal aorta. Using this technique, the graft survival was >98% with evidence in allotransplanted aorta of morphological changes pathognomonic of chronic rejection. On the contrary, no histopathological anomalies were discerned in aortic grafts transplanted across syngeneic animals. This modified surgical approach ameliorates the unacceptably high graft loss associated with earlier techniques, further extending the utility of this model as a tool to study the molecular and cellular mechanisms rudiment to the evolvement of chronic rejection
Hierarchical Re-estimation of Topic Models for Measuring Topical Diversity
A high degree of topical diversity is often considered to be an important
characteristic of interesting text documents. A recent proposal for measuring
topical diversity identifies three elements for assessing diversity: words,
topics, and documents as collections of words. Topic models play a central role
in this approach. Using standard topic models for measuring diversity of
documents is suboptimal due to generality and impurity. General topics only
include common information from a background corpus and are assigned to most of
the documents in the collection. Impure topics contain words that are not
related to the topic; impurity lowers the interpretability of topic models and
impure topics are likely to get assigned to documents erroneously. We propose a
hierarchical re-estimation approach for topic models to combat generality and
impurity; the proposed approach operates at three levels: words, topics, and
documents. Our re-estimation approach for measuring documents' topical
diversity outperforms the state of the art on PubMed dataset which is commonly
used for diversity experiments.Comment: Proceedings of the 39th European Conference on Information Retrieval

The performance of modified jatropha-based nanofluid during turning process
The industry's extensive use of petroleum-based metalworking fluid (MWF) harms the environment
and humans. The production of bio-based MWF, especially from crude jatropha oil (CJO), has therefore taken
numerous initiatives. This aimed to formulate newly modified jatropha oil (MJO) with the addition of 0.05wt.%
hBN and 0.05wt.% MoS2 as the nanofluid for MWF. The performance of the nanofluids was determined
through the turning process in terms of cutting temperature, workpiece surface roughness, tool life and tool
wear of the tool lubricated by the nanofluids. The performance of the nanofluid samples was compared with
the synthetic ester (SE). From the results, after conducted 100mm axial cutting length MJO+hBN+MoS2
recorded the lowest in cutting temperature and surface roughness compared to all samples. The result shows
that MJO+hBN+MoS2 has longer tool life (6500mm) compared to SE (6000mm). Abrasion and adhesion were
observed as the dominant tool wear mechanism. In conclusion, MJO+hBN+MoS2 shows better machining
performance and has the potential to be an environmentally friendly metalworking fluid
Magnetic Field resulting from non-linear electrical transport in single crystals of charge-ordered Pr Ca MnO}
In this letter we report that the current induced destabilization of the
charge ordered (CO) state in a rare-earth manganite gives rise to regions with
ferromagnetic correlation. We did this experiment by measurement of the I-V
curves in single crystal of the CO system
PrCaMnO and simultanously measuring the magnetization
of the current carrying conductor using a high T SQUID working at T = 77K.
We have found that the current induced destabilization of the CO state leads to
a regime of negative differential resistance which leads to a small enhancement
of the magnetization of the sample, indicating ferromagnetically aligned
moments.Comment: 4 pages LateX, 4 eps figure
