    Expression profiling of key pathways in rat liver after a one-year feeding trial with transgenic maize MON810

    In a recent one-year feeding study, we observed no adverse effects on tissue level in organs of rats fed with the genetically-modified maize MON810. Here, we assessed RNA expression levels of 86 key genes of the apoptosis-, NF-кB-, DNA-damage response (DDR)-, and unfolded-protein response (UPR) pathways by RT-qPCR in the rat liver. Male and female rats were fed either with 33% MON810 (GMO), isogenic- (ISO), or conventional maize (CONV) and RNAs were quantified from eight rats from each of the six feeding groups. Only Birc2 transcript showed a significant (p ≤ 0.05) consistent difference of ≥1.5-fold between the GMO and ISO groups in both sexes. Unsupervised cluster analysis showed a strong separation of male and female rats, but no clustering of the feeding groups. Individual analysis of the pathways did not show any clustering of the male or female feeding groups either, though transcript levels of UPR pathway-associated genes caused some clustering of the male GMO and CONV feeding group samples. These differences were not seen between the GMO and ISO control or within the female cohort. Our data therefore does not support an adverse effect on rat liver RNA expression through the long-term feeding of MON810 compared to isogenic control maize

    Longquan celadon: a quantified archaeological analysis of a pan-Indian Ocean industry of the 12th to 15th centuries

    This paper examines the Longquan celadon industry, located in Zhejiang province in China, which flourished mainly between the Southern Song and early Ming dynasties (12th to 15th century). The products of this industry are found on archaeological sites from across China and the Indian Ocean. This paper attempts a quantified analysis of the development of the industry based on archaeological data, focussing on four aspects: production, domestic consumption, overseas consumption and, to a lesser degree, workshop organisation. Although much of the data is still, in many ways, problematic, and many of the conclusions drawn are necessarily tentative, it is possible to demonstrate the value and timeliness of the approach by charting the overall development of this industry and by arguing that the close integration of the four aspects examined indicates that the Longquan celadon industry was an industry of considerable economic significance across much of the Indian Ocean

    The importation of Chinese porcelain and celadon into Europe has long been thought to have first begun around the thirteenth century AD. A unique group of Chinese ceramic sherds from archaeological contexts in Spain dated to between the ninth and eleventh centuries, however, now represents the earliest Chinese wares identified in Europe. Such an unexpectedly early presence on high-status sites in Western Europe probably reflects changing patterns of commerce in the Indian Ocean and the giving of prestigious gifts at the very highest levels of social and political power across the Islamic Mediterranean world

    TRAIL Enhances Shikonin Induced Apoptosis through ROS/JNK Signaling in Cholangiocarcinoma Cells

    Background/Aims: Cholangiocarcinoma (CCA), arising from varying locations within the biliary tree, is the second most common primary liver malignancy worldwide. Shikonin, an active compound extracted from the Chinese herb Zicao, holds anti-bacterial, anti-inflammatory, and anti-tumor activities. However, the effect of shikonin on human cholangiocarcinoma and detailed mechanisms of TRAIL enhancement remains to be elucidated. The purpose of the study was to investigate the protective functions of TRAIL enhancement for shikonin induced apoptosis in cholangiocarcinoma cells. Methods: We use MTT assay, apoptosis assay, caspase activity assay, flow cytometry assay, real time PCR and Western blot to observe the effects of TRAIL on shikonin induced cholangiocarcinoma cells apoptosis and its mechanism. Results: Shikonin inhibited cell viability and induced apoptosis of CCA cells, effects enhanced by TRAIL treatment via activation of caspase-3, -8, -9. Furhermore, TRAIL enhanced anti-proliferation of shikonin and shikonin induced apoptosis through induction of ROS mediated JNK activation, while AKT activation had an effect on shikonin anti-proliferation activity, but not in the TRAIL enhanced counterparts. Finally, shikonin upregulated DR5 expression, an effect essential for TRAIL-enhanced activities of shikonin in RBE cells. Conclusions: Our results revealed that shikonin could inhibit cells viability and induce apoptosis of CCA cells, effects enhanced by TRAIL treatment via ROS mediated JNK signalling pathways, involving up-regulation of DR5 expression. Our results provide further insight into the mechanism underlying the anti-tumor effects of shikonin by TRAIL enhanced in CCA and a new therapeutic strategy to CCA treatment
