    Fantasia' e `Ruiva' são dois cultivares de uva de mesa desenvolvidos pelo Departamento de Agricultura dos E.U.A., lançadas na Califórnia em 1989 e introduzidos no Brasil, no mesmo ano, pelos autores. No final do inverno de 1991, garfos dos dois cultivares foram enxertados sobre três diferentes porta-enxertos, a saber: Kober 5BB, Ripária do Traviú (106-8 Mgt) e IAC 766 `Campinas', no Centro Experimental de Campinas, do IAC. Garfos também foram distribuídos para alguns viticultores selecionados em diferentes regiões paulistas. Resultados de observações preliminares são apresentados. Para a familiarização pelos viticultores, bem como facilitar a pronúncia em português, os cultivares foram renomeados como sendo `Fantasia' para "Fantasy Seedless" e `Ruiva' para "Crimson Seedless". `Fantasia' é uma uva preta, de ciclo precoce a médio, com cachos médios (350-550g), de 13-20cm de comprimento e de compacidade média a solta. Os bagos são naturalmente grandes, apesar de apirenas, pesando em média 4-9g, com diâmetro de 17 a 22mm e 20-30mm de comprimento e ovais. `Ruiva' é uma uva avermelhada, de ciclo médio, com cachos médios a grandes (460-620g), com 18-30cm de comprimento e levemente compactos. Os bagos são naturalmente grandes, apesar de apirenas, pesando em média 3,5-8,0g, diâmetro de 16-21mm e 18-30mm de comprimento e cilíndricas a ovais. A duração do ciclo vegetativo (da poda à colheita) em Campinas, para ambos os cultivares foi aproximadamente o mesmo, independentemente do porta-enxerto utilizado. Por outro lado, o vigor, avaliado por intermédio do diâmetro do tronco e do peso de ramos podados, foi mais pronunciado para Ruiva enxertado sobre Kober 5BB. O comportamento de ambos cultivares em São Paulo, avaliado nas condições enunciadas, mostrou elevado potencial produtivo, aliado a excelentes características comerciais.<br>"Fantasy Seedless" and "Crimson Seedless" are two vinifera table grape cultivars released in 1989 by the USDA, Fresno, CA, USA, and introduced in Brazil by the authors. At the end of winter of 1991, cuttings of both cultivars were grafted on three different rootstocks at Campinas, SP, Brazil. Cuttings were also distributed for selected growers of various regions in the São Paulo State. Results of preliminary observations are presented here. "Fantasy Seedless" is an early to mid-season black seedless grape with cluster medium in size (350-550g), with a length of 13-20cm and a medium to loose compactness. The berries are naturally large, averaging 4-9g, 17-22mm in diameter, 20-30mm long, and are mostly oval. "Crimson Seedless" is a mid season red seedless grape with cluster medium in size (460-620g) and length (18-30cm), and are slightly compact. The berries are naturally large, averaging 3.5-8.0g, 16-21mm in diameter, 18-30mm long, and are cylindrical to oval in shape. The duration of the cycle (pruning to harvest) in Campinas, for both cultivars was about the same whether grafted on Kober 5BB, 106-8 Mgt or IAC 766, but vigor evaluated through trunk diameter and pruned weight, was higher for Crimson Seedless grafted on Kober 5BB. The performance of both cultivars in the São Paulo State showed that they can be commercially recommended. Both varieties were renamed in Portuguese as follows: Fantasia for "Fantasy Seedless" and Ruiva for "Crimson Seedless"

    The objetive of this work was to rescue immature embryos of apple rootstocks Malus prunifolia (Marubakaido) and Malus pumila (M9) after 40-60 days of pollination and to put them into MS culture media supplemented with agar (6 g L-1) and casein hydrolysate (500 mg L-1). Embryos originated from interspecific crosses and open pollination showed differences in the in vitro responses, depending on the female parent, the developmental stage of the embryo, and the culture medium composition. Embryos of the M. pumila rootstock, rescued within 40 days after pollination and put in culture medium supplemented with indolacetic acid (IAA), gibberellic acid (GA3), kinetin and maltose, resulted in a normal development of plantlets. However, embryos originating from hand-pollination, cultivated in medium supplemented with 14 µM IAA, 5 µM kinetin and 1.5 µM Ga3 (MS1), mainly those of M. prunifolia x M. pumila, showed a high percentage of rusted embryos (96.2%). Embryos from open pollination of M. prunifolia and M. pumila formed calluses. It was possible to identify the influence of the female parent by the enhanced development of M. pumila shoots derived from open or hand-pollination. The crossing of responsive species and the use of the technique of embryo culture provided a rapid and uniform germination and, consequently, the development of fully normal seedlings.<br>O objetivo deste trabalho foi resgatar embriões imaturos de porta-enxertos de macieira Malus prunifolia (Marubakaido) e Malus pumila (M9) depois de 40 e 60 dias de polinização e colocá-los em meio de cultura MS suplementado com ágar (6 g L-1) e hidrolisado de caseína (500 mg L-1). Embriões originados do cruzamento interespecífico dirigido e de polinização aberta mostraram diferenças significativas in vitro, tendo sido observadas diferenças quanto ao progenitor feminino, quanto à fase de desenvolvimento do embrião e à composição do meio de cultura. Embriões do porta-enxerto M. pumila, resgatados aos 40 dias depois da polinização e colocados em meio de cultura suplementado com ácido indolacético (AIA), ácido giberélico (GA3), cinetina e maltose, resultaram em desenvolvimento normal das plantas. Porém, foi observada alta porcentagem de embriões oxidados (96,2%), originados de polinização dirigida, cultivados em meio com 14 µM de AIA, 5 µM de cinetina e 1,5 µM de GA3 (MS1), principalmente para o cruzamento M. prunifolia e M. pumila. Embriões de polinização aberta de M. prunifolia x M. pumila formaram calos. Foi possível identificar a influência do progenitor feminino, pelo incremento de brotações de M. pumila derivada de polinização aberta e dirigida. O cruzamento de espécies responsivas e o uso da técnica de cultura de embrião promoveram germinação rápida e uniforme e, por conseguinte, o desenvolvimento de mudas normais

    Growth regulators and darkness increase efficiency on in vitro culture of immature embryos from peppers

    [EN] Common pepper (Capsicum annuum L.) is one of the most important vegetables in the world, and extensive breeding efforts are being made to develop new improved strains of this species. In this regard, in vitro culture of immature embryos may help breeders accelerate breeding cycles and overcome interspecific barriers, among other applications. In this study, we have optimized a protocol for in vitro culture of immature embryos of C. annuum. Levels of indole-3-acetic acid (IAA) and zeatin have been tested to improve the efficiency (germination rates) of this technique in C. annuum embryos at the four main immature stages (i.e. globular, heart, torpedo, and early cotyledonary) from four varietal types of this species (California Wonder, Piquillo, Guindilla, and Bola). The effect of 5-day initial incubation in the dark was also tested on the most efficient hormone formulation. On average, relatively low levels of both IAA and zeatin (0.01 mg L-1 each) (M-1) provided the highest germination rates, particularly in the advanced stages (torpedo and cotyledonary). To a lesser extent, the lack of these growth regulators (M-0) or high IAA (0.2 mg L-1)/low zeatin (0.01 mg L-1) (M-2) combination also had a positive response. On the contrary, high zeatin levels (0.2 mg L-1) produced very low germination rates or callus development (efficiency 0-7 %). Different responses were also found between genotypes. Thus, considering the best media (M-0, M-1, M-2), Bola embryos had the highest rates. M-1 plus 5-days of initial dark incubation (M-1-D) improved the efficiency rates at all embryo stages, particularly in the earliest (globular) embryos which increased from 3 % to > 20 %.Juan P.
