    Infection studies of Diplodia zeae (Schw.) Lev. and control of seedling blights of corn

    The two most important symptoms of the corn disease caused by Diplodia zeae are seedling blight and ear rot. Artificial inoculation of the ears was effected in all stages of their development but most readily in the milk stage. Infection of seedlings from Diplodia-infected kernels usually takes place in the natural wounds at the points of production of the secondary radicles near the attachment of the mesocotyl to the scutellum. Field experiments showed that Diplodia-infected seed in 1925 and 1927 produced 4.5 and 1.3 times as many weak plants, respectively, as did nearly disease-free seed. Plants that are weak due to Diplodia zeae and Basisporium gallarum early in the season often produce normal ears if there is no competition in the hill but seldom produce normal ears if there is competition with strong plants. The visible-root sand culture method, devised as a means of following the spread of Diplodia in the underground parts of the corn seedling, has been found of value in correlating micro-organism injury or disinfectant injury with top growth and in determining the effect of moisture and temperature on the development of Diplodia seedling blight. Fewer plants developed from Diplodia-infected seed at 15 °-19° C. than at 20°-24° C. There was less visible seedling blight at the lower temperature, due to the large number of seedlings that were killed before they reached the surface of the soil. Results of seed treatment experiments in 1926 using nine mercury disinfectants on nearly disease-free seed, showed that yields were not increased or decreased significantly. However, in 1927, treated, high quality, selected seed corn entered in the state yield contest gave significant increases. In 1926, Diplodia-infected treated seed showed significant increases ranging from 6.8 to 13.9 bushels per acre. The yield from the highest yielding, Diplodiainfected, treated seed was within about six bushels of that from the nearly disease-free seed. In 1927, three mercury dust disinfectants (Bayer Dust, Merko and Semesan Jr.) increased the yield of Diplodia-infected seed from 21.3 to 30.2 percent and of Basisporium-infected seed from 15.5 to 19.6 percent. The benefits derived from treatment of seed corn vary greatly with the amount of infection in the seed, with the climatic and soil conditions at planting time and apparently with the strain of corn used

    Predicting Future Changes in Muskegon River Watershed Game Fish Distributions under Future Land Cover Alteration and Climate Change Scenarios

    Future alterations in land cover and climate are likely to cause substantial changes in the ranges of fish species. Predictive distribution models are an important tool for assessing the probability that these changes will cause increases or decreases in or the extirpation of species. Classification tree models that predict the probability of game fish presence were applied to the streams of the Muskegon River watershed, Michigan. The models were used to study three potential future scenarios: (1) land cover change only, (2) land cover change and a 3°C increase in air temperature by 2100, and (3) land cover change and a 5°C increase in air temperature by 2100. The analysis indicated that the expected change in air temperature and subsequent change in water temperatures would result in the decline of coldwater fish in the Muskegon watershed by the end of the 21st century while cool‐ and warmwater species would significantly increase their ranges. The greatest decline detected was a 90% reduction in the probability that brook trout Salvelinus fontinalis would occur in Bigelow Creek. The greatest increase was a 276% increase in the probability that northern pike Esox lucius would occur in the Middle Branch River. Changes in land cover are expected to cause large changes in a few fish species, such as walleye Sander vitreus and Chinook salmon Oncorhynchus tshawytscha, but not to drive major changes in species composition. Managers can alter stream environmental conditions to maximize the probability that species will reside in particular stream reaches through application of the classification tree models.     Evaluation of hypoxia in an experimental rat tumour model by [18F]Fluoromisonidazole PET and immunohistochemistry

    This study aimed to evaluate tumour hypoxia by comparing [(18)F]Fluoromisonidazole uptake measured using positron emission tomography ([(18)F]FMISO-PET) with immunohistochemical (IHC) staining techniques. Syngeneic rhabdomyosarcoma (R1) tumour pieces were transplanted subcutaneously in the flanks of WAG/Rij rats. Tumours were analysed at volumes between 0.9 and 7.3 cm(3). Hypoxic volumes were defined using a 3D region of interest on 2 h postinjection [(18)F]FMISO-PET images, applying different thresholds (1.2-3.0). Monoclonal antibodies to pimonidazole (PIMO) and carbonic anhydrase IX (CA IX), exogenous and endogenous markers of hypoxia, respectively, were used for IHC staining. Marker-positive fractions were microscopically measured for each tumour, and hypoxic volumes were calculated. A heterogeneous distribution of hypoxia was observed both with histology and [(18)F]FMISO autoradiography. A statistically significant correlation (P<0.05) was obtained between the hypoxic volumes defined with [(18)F]FMISO-PET and the volumes derived from the PIMO-stained tumour sections (r=0.9066; P=0.0001), regardless of the selected threshold between 1.4 and 2.2. A similar observation was made with the CA IX staining (r=0.8636; P=0.0006). The relationship found between [(18)F]FMISO-PET and PIMO- and additionally CA IX-derived hypoxic volumes in rat rhabdomyosarcomas indicates the value of the noninvasive imaging method to measure hypoxia in whole tumours.Journal ArticleSCOPUS: ar.jinfo:eu-repo/semantics/publishe

    Vascular architecture and hypoxic profiles in human head and neck squamous cell carcinomas

    Tumour oxygenation and vasculature are determinants for radiation treatment outcome and prognosis in patients with squamous cell carcinomas of the head and neck. In this study we visualized and quantified these factors which may provide a predictive tool for new treatments. Twenty-one patients with stage III–IV squamous cell carcinomas of the head and neck were intravenously injected with pimonidazole, a bioreductive hypoxic marker. Tumour biopsies were taken 2 h later. Frozen tissue sections were stained for vessels and hypoxia by fluorescent immunohistochemistry. Twenty-two sections of biopsies of different head and neck sites were scanned and analysed with a computerized image analysis system. The hypoxic fractions varied from 0.02 to 0.29 and were independent from T- and N-classification, localization and differentiation grade. No significant correlation between hypoxic fraction and vascular density was observed. As a first attempt to categorize tumours based on their hypoxic profile, three different hypoxia patterns are described. The first category comprised tumours with large hypoxic, but viable, areas at distances even greater than 200 ÎŒm from the vessels. The second category showed a typical band-like distribution of hypoxia at an intermediate distance (50–200 ÎŒm) from the vessels with necrosis at greater distances. The third category demonstrated hypoxia already within 50 ÎŒm from the vessels, suggestive for acute hypoxia. This method of multiparameter analysis proved to be clinically feasible. The information on architectural patterns and the differences that exist between tumours can improve our understanding of the tumour micro-environment and may in the future be of assistance with the selection of (oxygenation modifying) treatment strategies. © 2000 Cancer Research Campaig

    Benznidazole biotransformation and multiple targets in <i>Trypanosoma</i> cruzi revealed by metabolomics

    &lt;b&gt;Background&lt;/b&gt;&lt;p&gt;&lt;/p&gt; The first line treatment for Chagas disease, a neglected tropical disease caused by the protozoan parasite Trypanosoma cruzi, involves administration of benznidazole (Bzn). Bzn is a 2-nitroimidazole pro-drug which requires nitroreduction to become active, although its mode of action is not fully understood. In the present work we used a non-targeted MS-based metabolomics approach to study the metabolic response of T. cruzi to Bzn.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Methodology/Principal findings&lt;/b&gt;&lt;p&gt;&lt;/p&gt; Parasites treated with Bzn were minimally altered compared to untreated trypanosomes, although the redox active thiols trypanothione, homotrypanothione and cysteine were significantly diminished in abundance post-treatment. In addition, multiple Bzn-derived metabolites were detected after treatment. These metabolites included reduction products, fragments and covalent adducts of reduced Bzn linked to each of the major low molecular weight thiols: trypanothione, glutathione, Îł-glutamylcysteine, glutathionylspermidine, cysteine and ovothiol A. Bzn products known to be generated in vitro by the unusual trypanosomal nitroreductase, TcNTRI, were found within the parasites, but low molecular weight adducts of glyoxal, a proposed toxic end-product of NTRI Bzn metabolism, were not detected.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Conclusions/significance&lt;/b&gt;&lt;p&gt;&lt;/p&gt; Our data is indicative of a major role of the thiol binding capacity of Bzn reduction products in the mechanism of Bzn toxicity against T. cruzi

    Developing User‐Friendly Habitat Suitability Tools from Regional Stream Fish Survey Data

    We developed user‐friendly fish habitat suitability tools (plots) for fishery managers in Michigan; these tools are based on driving habitat variables and fish population estimates for several hundred stream sites throughout the state. We generated contour plots to show patterns in fish biomass for over 60 common species (and for 120 species grouped at the family level) in relation to axes of catchment area and low‐flow yield (90% exceedance flow divided by catchment area) and also in relation to axes of mean and weekly range of July temperatures. The plots showed distinct patterns in fish habitat suitability at each level of biological organization studied and were useful for quantitatively comparing river sites. We demonstrate how these plots can be used to support stream management, and we provide examples pertaining to resource assessment, trout stocking, angling regulations, chemical reclamation of marginal trout streams, indicator species, instream flow protection, and habitat restoration.     Environmental changes and violent conflict

    This letter reviews the scientific literature on whether and how environmental changes affect the risk of violent conflict. The available evidence from qualitative case studies indicates that environmental stress can contribute to violent conflict in some specific cases. Results from quantitative large-N studies, however, strongly suggest that we should be careful in drawing general conclusions. Those large-N studies that we regard as the most sophisticated ones obtain results that are not robust to alternative model specifications and, thus, have been debated. This suggests that environmental changes may, under specific circumstances, increase the risk of violent conflict, but not necessarily in a systematic way and unconditionally. Hence there is, to date, no scientific consensus on the impact of environmental changes on violent conflict. This letter also highlights the most important challenges for further research on the subject. One of the key issues is that the effects of environmental changes on violent conflict are likely to be contingent on a set of economic and political conditions that determine adaptation capacity. In the authors' view, the most important indirect effects are likely to lead from environmental changes via economic performance and migration to violent conflict. © 2012 IOP Publishing Ltd
