    The Gramicidin Dimer Shows Both EX1 and EX2 Mechanisms of H/D Exchange

    We describe the use of H/D amide exchange and electrospray ionization mass spectrometry to study, in organic solvents, the pentadecapeptide gramicidin as a model for protein self association. In methanol-OD, all active H's in the peptide exchange for D within 5 min, indicating a monomer/dimer equilibrium that is shifted towards the fast-exchanging monomer. H/D exchange in n-propanol-OD, however, showed a partially protected gramicidin that slowly converts to a second species that exchanges nearly all the active hydrogens, indicating EX1 kinetics for the H/D exchange. We propose that this behavior is the result of the slower rate of unfolding in n-propanol compared with that in methanol. The rate constant for the unfolding of the dimer is the rate of disappearance of the partially protected species, and it agrees within a factor of two with a value reported in literature. The rate constant of dimer refolding can be determined from the ratio of the rate constant for unfolding and the affinity constant for the dimer, which we determined in an earlier study. The unfolding activation energy is 20 kcal mol−1, determined by performing the exchange experiments as a function of temperature. To study gramicidin in an even more hydrophobic medium than n-propanol, we measured its H/D exchange kinetics in a phospholipids vesicle and found a different H/D amide exchange behavior.     Aggressive Surveillance Is Needed to Detect Endoleaks and Junctional Separation between Device Components after Zenith Fenestrated Aortic Reconstruction

    Background Junctional separation and resulting type IIIa endoleak is a well-known problem after EVAR (endovascular aneurysm repair). This complication results in sac pressurization, enlargement, and eventual rupture. In this manuscript, we review the incidence of this late finding in our experience with the Cook Zenith fenestrated endoprosthesis (ZFEN, Bloomington, IN). Methods A retrospective review was performed of a prospectively maintained institutional ZFEN fenestrated EVAR database capturing all ZFENs implanted at a large-volume, academic hospital system. Patients who experienced junctional separation between the fenestrated main body and distal bifurcated graft (with or without type IIIa endoleak) at any time after initial endoprosthesis implantation were subject to further evaluation of imaging and medical records to abstract clinical courses. Results In 110 ZFENs implanted from October 2012 to December 2017 followed for a mean of 1.5 years, we observed a 4.5% and 2.7% incidence of clinically significant junctional separation and type IIIa endoleak, respectively. Junctional separation was directly related to concurrent type Ib endoleak in all 5 patients. Three patients presented with sac enlargement. One patient did not demonstrate any evidence of clinically significant endoleak and had a decreasing sac size during follow-up imaging. The mean time to diagnosis of modular separation in these patients was 40 months. Junctional separation was captured in surveillance in 2 patients and reintervened upon before manifestation of endoleak. However, the remaining 3 patients completed modular separation resulting in rupture and emergent intervention in 2 and an aortic-related mortality in the other. Conclusions Junctional separation between the fenestrated main and distal bifurcated body with the potential for type IIIa endoleak is an established complication associated with the ZFEN platform. Therefore, we advocate for maximizing aortic overlap during the index procedure followed by aggressive surveillance and treatment of stent overlap loss captured on imaging

    A simple and sensitive spectrophotometric method has been developed for the estimation of Mycophenolic acid. The method is based on the formation of   Ion-Association complex with MYCO formed an ion -association complex with basic dye, SafraninO. The cationic form of the dye SAFO involves in the formation of neutral coloured ion-association complexe with negative charge (acid groups in the drug) which is extractable into chloroform and behaves as a single unit being held together by electrostatic attraction. The absorption maxima were found to be at lMax 520 nm. The method obeys Beer's law within the limits 10-40µg/ml and gives reproducible results. Molar absorptivity value is obtained as8.424x104 L mol-1 cm-1 and recovery was found to be 99.17 ± 0.92 to 99.62 ± 0.27. Interferences of the other ingredients and excipients were not observed.     Possibility of Unconventional Pairing Due to Coulomb Interaction in Fe-Based Pnictide Superconductors: Perturbative Analysis of Multi-Band Hubbard Models

    Possibility of unconventional pairing due to Coulomb interaction in iron-pnictide superconductors is studied by applying a perturbative approach to realistic 2- and 5-band Hubbard models. The linearized Eliashberg equation is solved by expanding the effective pairing interaction perturbatively up to third order in the on-site Coulomb integrals. The numerical results for the 5-band model suggest that the eigenvalues of the Eliashberg equation are sufficiently large to explain the actual high Tc for realistic values of Coulomb interaction and the most probable pairing state is spin-singlet s-wave without any nodes just on the Fermi surfaces, although the superconducting order parameter changes its sign between the small Fermi pockets. On the other hand the 2-band model is quite insufficient to explain the actual high Tc.Comment: 2 pages, 3 figures. Proceedings of the Intl.     Public transport: a large scale fomite of methicillin-resistant Staphylococcus aureus

    Background: The role of public transport as reservoirs of antibiotic-resistant staphylococci was determined.Methods: 200 swabs were collected from 50 public buses (urban and rural) circulating in Davangere, Karnataka. Swabs collected were inoculated on Blood agar, Mannitol salt agar and MacConkey agar plates. After incubation for 24-48 hours, plates were examined for the growth of Staphylococcus aureus. Anti-microbial susceptibility test was performed using oxacillin 1ug disc to detect methicillin resistance as per CSLI guidelines.Results: Out of 40 Staphylococcus aureus isolated 35 isolates were resistant to more than two classes of antibiotics, hence multidrug resistant Staphylococcus aureus. Out of 35 MDR isolates, 18 were resistant to oxacillin and cefoxitin. Minimum inhibitory concentration test revealed that out of 35 MDR isolates, 18 isolates had MIC value of ≥ 4µg/ml.Conclusions: The recovery methicillin-resistant Staphylococcus aureus from public transport system implies a potential risk for transmission of these bacteria in community

    Topological Insulators from Spontaneous Symmetry Breaking Induced by Electron Correlation on Pyrochlore Lattices

    We study an extended Hubbard model with the nearest-neighbor Coulomb interaction on the pyrochlore lattice at half filling. An interaction-driven insulating phase with nontrivial Z_2 invariants emerges at the Hartree-Fock mean-field level in the phase diagram. This topological insulator phase competes with other ordered states and survives in a parameter region surrounded by a semimetal, antiferromagnetic and charge ordered insulators. The symmetries of these phases are group-theoretically analyzed.     Safety evaluation of chromium picolinate in experimental rats

    Background: Chromium, an essential trace mineral plays an important role in the metabolism of carbohydrate, fat and proteins. Chromium picolinate (Cr.Pic) is used in alternative medicine to treat chromium deficiency. Though Cr.Pic is increasingly used to treat diabetes and obesity, studies on its safety profile is limited.Methods: Acute toxicity study was conducted by oral administration of Cr.Pic (2000 mg/kg body weight). The animals were maintained another 14 days with once a day observation. For sub-chronic studies, test groups were treated with Cr.Pic 10 mg/kg/day for 90 days. Tests for hepatic and renal function were conducted. Effect of Cr.Pic on behavioural changes and motor co-ordination was done on every week. Histopathological studies were conducted on day 90 at the end of the experiment.Results: Acute toxicity study of Cr.Pic showed no signs of toxicity and mortality. Absence of any behavioural alteration or mortality during the period of 14 days indicates that Cr.Pic has no latent effect. Similar results were obtained with sub-chronic studies suggesting safety of Cr.Pic. Cr.Pic treated groups showed no changes in learning and motor co-ordination compared to the untreated group. No gross histopathological changes were seen in any group indicating safety of Cr.Pic.Conclusions: The present study conferred safety profile of Cr.Pic from normal results obtained in hepatic function, renal function, behavioural and histopathological studies, suggesting its safety

    Rickettsial neglected zoonoses: prevalence of scrub typhus at central Karnataka

    Background: Fever of unknown Origin (FUO) has many multiple causes such as enteric fever, malaria, dengue, tuberculosis, brucellosis. But scrub typhus is less known cause in Indian scenario. The present study reports the prevalence of scrub typhus at central Karnataka and compares the sensitivity and specificity of Weil-Felix test and the IgM ELISA in the detection of infection.Methods: 368 serum samples of FUO cases were collected. Weil-Felix test was performed and also analyzed for IgM antibodies to Orienta tsutsugamushi by IgM ELISA test along with haematological and biochemical investigations.Results: Out of 368 patients of fever of unknown origin, 94 cases were positive by OXK antigens by Weil Felix test and 61 were positive by ELISA test for ST IgM antibodies. Fever was the most common clinical presentation occurring in ST IgM ELISA positive cases, followed by myalgia in 90.1% cases, headache in 77%, hepatomegaly in 65.5%, splenomegaly in 62.2% and rashes were seen in 29.5% patients. Eschar was seen in 13.1% patients, pneumonia in 3.2% and meningo-encephalitis in 1.6%. Sensitivity and specificity of WFT in relation to IgM ELISA at a titre of 160 was 81.97% and 85.67% respectively.Conclusions: With the growing number of cases detected in India, scrub typhus is fast emerging as a public health threat and also due to limited diagnostics leading to underreporting, Weil Felix test could be used in adjunct with Enzyme-linked immunosorbent assay and blood parameters in the diagnosis of rickettsial diseases

    Coherence effect in a two-band superconductor: Application to iron pnictides

    From a theoretical point of view, we propose an experimental method to determine the pairing symmetry of iron pnictides. We focus on two kinds of pairing symmetries, s+−s_{+-} and s++s_{++}, which are strong candidates for the pairing symmetry of iron pnictides. For each of these two symmetries, we calculate both the density and spin response functions by using the two-band BCS model within the one-loop approximation. As a result, a clear difference is found between the s+−s_{+-}- and s++s_{++}-wave states in the temperature dependence of the response functions at nesting vector Q\bf{Q}, which connects the hole and electron Fermi surfaces. We point out that this difference comes from the coherence effect in the two-band superconductor. We suggest that the pairing symmetry could be clarified by observing the temperature dependence of both the density and spin structure factors at the nesting vector Q\bf{Q} in neutron scattering measurements.Comment: 15 pages, 7 figures, 1 tabl
