    Effect of the type of acetic fermentation process on the chemical composition of prickly pear vinegar (Opuntia ficus-indica).

    BACKGROUND: In several countries, the cactus plant (Opuntia ficus-indica (L). Mill) knows renewed attention due to its ecological, socio-economic, and environmental role. In this study, prickly pear vinegar was produced employing two types of acetification processes: surface and submerged culture. Both acetification processes were performed at different temperatures (30 °C, 37 °C, 40 °C) by using two different species of thermo-tolerant acetic acid bacteria (Acetobacter malorum and Gluconobacter oxydans). Polyphenols and volatiles compounds analyzed by UPLC/DAD and SBSE-GC/MS, respectively, were considered as the main variables to determine the effect of the acetification process on the quality of the vinegar. RESULTS: As a result, fifteen polyphenols and seventy volatile compounds were identified and quantified in the vinegar samples produced by both acetification processes. The results showed that the surface acetification method led to an increase in the concentration of phenolic components, which was higher than that in the submerged process. However, a significant increase of volatile compounds predominated by esters and acids was observed when submerged culture acetification was employed, whereas alcohols were predominant in surface culture vinegars. Moreover, the multivariate statistical analysis showed that the components that mostly contributed to the differentiation between all vinegar samples were the volatile compounds. CONCLUSION: It has been proved that prickly pear vinegar could be successfully produced at higher temperatures than usual, by employing thermo-tolerant bacteria, and that the type of acetification method significantly affects the final quality of the vinegar produced.     Influence of Different Bacteria Inocula and Temperature Levels on the Chemical Composition and Antioxidant Activity of Prickly Pear Vinegar Produced by Surface Culture

    This work intends to determine the effect on the aroma profile, phenolic content and antioxidant activity of prickly pear vinegars produced by the surface culture at two different fermentation temperatures and using different acetic acid bacteria (AAB) inocula. Prickly pear wine was fermented at two temperature levels (30 and 37 degrees C) by using bacteria inocula containing Acetobacter, Gluconobacter or a mixture of bacteria isolated from Sherry vinegars. Eighty-five individual volatile compounds from different families and sixteen polyphenolic compounds have been identified. It was confirmed that the highest temperature tested (37 degrees C) resulted in a lower concentration of volatile compounds, while no significant effect on the vinegars' volatile composition could be associated with the AAB inoculum used. Contrariwise, the highest content of polyphenolic compounds was detected in those vinegars produced at 37 degrees C and their concentration was also affected by the type of AAB inoculum used. Prickly pear wine displayed greater antioxidant activity than juices or vinegars, while the vinegars obtained through the mixture of AAB from Sherry vinegar showed higher antiradical activity than those obtained through either of the two AAB genera used in this study. It can be therefore concluded that, although the volatile content of vinegars decreased when fermented at a higher temperature, vinegars with a higher content in polyphenols could be obtained by means of partial fermentations at 37 degrees C, as long as thermotolerant bacteria were employed

    Production of prickly pear (Opuntia ficus-indica) vinegar in submerged culture using Acetobacter malorum and Gluconobacter oxydans: Study of volatile and polyphenolic composition

    Prickly pear (Opuntia ficus-indica (L). Mill.) vinegar has been produced by submerged culture using scale laboratory fermenters at three acetification temperatures (30 ◦C, 37 ◦C, and 40 ◦C). Pure thermotolerant Acetic Acid Bacteria (AAB) which had been previously submitted to molecular identification by PCR–RFLP of the 16S–23S rDNA regions and 16S–23S ITS rDNA sequencing, were used. The AAB used were identified as Acetobacter malorum and Gluconobacter oxydans. The acetic fermentation achieved by A. malorum at 30 ◦C and 37 ◦C was more efficient, in terms of acidity, than that accomplished by G. oxydans. The chemical analysis revealed the presence of 85 individual volatile compounds and 17 polyphenolic compounds. The concentration of approximately half the volatile compounds was significantly affected by fermentation temperature, with clearly lower concentrations as temperature increased, whereas few significant differences were observed when comparing the vinegars produced by the two AAB species. Regarding phenolic compounds, significant increases were registered when temperature changed from 30 ◦C to 40 ◦C. Furthermore, the vinegars produced by A. malorum presented a greater concentration of phenolic compounds than those produced by G. oxydans

    Screening and Characterization of New Acetobacter fabarum and Acetobacter pasteurianus Strains with High Ethanol-Thermo Tolerance and the Optimization of Acetic Acid Production

    The production of vinegar on an industrial scale from different raw materials is subject to constraints, notably the low tolerance of acetic acid bacteria (AAB) to high temperatures and high ethanol concentrations. In this study, we used 25 samples of different fruits from seven Moroccan biotopes with arid and semi-arid environmental conditions as a basic substrate to isolate thermo- and ethanol-tolerant AAB strains. The isolation and morphological, biochemical and metabolic characterization of these bacteria allowed us to isolate a total number of 400 strains with characters similar to AAB, of which six strains (FAGD1, FAGD10, FAGD18 and GCM2, GCM4, GCM15) were found to be mobile and immobile Gram-negative bacteria with ellipsoidal rod-shaped colonies that clustered in pairs and in isolated chains. These strains are capable of producing acetic acid from ethanol, growing on peptone and oxidizing acetate to CO2 and H2O. Strains FAGD1, FAGD10 and FAGD18 show negative growth on YPG medium containing D-glucose > 30%, while strains GCM2, GCM4 and GCM15 show positive growth. These six strains stand out on CARR indicator medium as isolates of the genus Acetobacter ssp. Analysis of 16S rDNA gene sequencing allowed us to differentiate these strains as Acetobacter fabarum and Acetobacter pasteurianus. The study of the tolerance of these six isolates towards pH showed that most of the six strains are unable to grow at pH 3 and pH 9, with an ideal pH of 5. The behavior of the six strains at different concentrations of ethanol shows an optimal production of acetic acid after incubation at concentrations between 6% and 8% (v/v) of ethanol. All six strains tolerated an ethanol concentration of 16% (v/v). The resistance of the strains to acetic acid differs between the species of AAB. The optimum acetic acid production is obtained at a concentration of 1% (v/v) for the strains of FAGD1, FAGD10 and FAGD18, and 3% (v/v) for GCM2, GCM4 and GCM15. These strains are able to tolerate an acetic acid concentration of up to 6% (v/v). The production kinetics of the six strains show the highest levels of growth and acetic acid production at 30 degrees C. This rate of growth and acetic acid production is high at 35 degrees C, 37 degrees C and 40 degrees C. Above 40 degrees C, the production of acid is reduced. All six strains continue to produce acetic acid, even at high temperatures up to 48 degrees C. These strains can be used in the vinegar production industry to minimize the load on cooling systems, especially in countries with high summer temperatures

    Semi-automated repetitive sequence-based PCR amplification for species of the Scedosporium apiospermum complex

    International audiencePurpose: The Scedosporium apiospermum species complex usually ranks second among the filamentous fungi colonizing the airways of patients with cystic fibrosis (CF), but little is known about the molecular epidemiology of the airway colonization.Methods: Polymerase chain reaction (PCR) amplification of repetitive sequences (rep-PCR) was applied to the retrospective analysis of a panel of isolates already studied by random amplification of polymorphic DNA (RAPD) and comprising 63 isolates recovered from sputa from 9 CF patients. Results were compared to those obtained previously by RAPD, and herein by beta-tubulin (TUB) gene sequencing and Multilocus Sequence Typing (MLST).Results: Within the panel of isolates studied, S. apiospermum sensu stricto and Sce-dosporium boydii, as expected, were the predominant species with 21 and 36 isolates, respectively. Four isolates from one patient were identified as Scedosporium auranti-acum, whereas two isolates belonged to the Pseudallescheria ellipsoidea subgroup of S. boydii. rep-PCR analysis of these isolates clearly differentiated the three species and P. ellipsoidea isolates, whatever the rep-PCR kit used, and also permitted strain differentiation. When using the mold primer kit, results from rep-PCR were in close agreement with those obtained by MLST. For both S. apiospermum and S. boydii, 8 genotypes were differentiated by rep-PCR and MLST compared to 10 by RAPD. All S. aurantiacum isolates shared the same RAPD genotype and exhibited the same rep-PCR profile and sequence type.Conclusions: These results illustrate the efficacy of rep-PCR for both species identification within the S. apiospermum complex and genotyping for the two major species of this comple

    A novel approach for adsorption of organic dyes from aqueous solutions using a sodium alginate/titanium dioxide nanowire doped with zirconium cryogel beads

    The presence of organic dyes in wastewater raises significant environmental and human health concerns, owing to their high toxicity. In light of this, a novel adsorbent material with porous cryogel architecture was developed and employed for the effective removal of organic dyes from an aqueous solution. Initially, a titanium dioxide nanowire doped with zirconium HZTO was synthesized by the hydrothermal process. Subsequently, the beads (SA/HZTO) of sodium alginate and HZTO were successfully prepared through a cross-linking process, employing Ca2+ ions as the crosslinking agent. Structural analysis of SA/HZTO beads was performed using FTIR, SEM, and EDX techniques. We systematically examined the impact of different conditions, including the initial dye concentration, pH, contact time, and adsorbent dosage, on the adsorption process. Batch experiments, both in signal and binary systems, were conducted to rigorously assess the dye adsorption capabilities. Kinetic modeling revealed that the adsorption process adhered to the pseudo-second-order kinetic model. Remarkably, the prepared beads exhibited impressive adsorption capacities of 26 and 29 mg/g toward methylene blue (MB) and safranin (SF), respectively. SA/HZTO beads have demonstrated excellent adsorption properties, offering a promising avenue for the development of low-cost, efficient, and reusable adsorbent to remove dyes from wastewater

    Association of respiratory symptoms and lung function with occupation in the multinational Burden of Obstructive Lung Disease (BOLD) study

    Background Chronic obstructive pulmonary disease has been associated with exposures in the workplace. We aimed to assess the association of respiratory symptoms and lung function with occupation in the Burden of Obstructive Lung Disease study. Methods We analysed cross-sectional data from 28 823 adults (≥40 years) in 34 countries. We considered 11 occupations and grouped them by likelihood of exposure to organic dusts, inorganic dusts and fumes. The association of chronic cough, chronic phlegm, wheeze, dyspnoea, forced vital capacity (FVC) and forced expiratory volume in 1 s (FEV1)/FVC with occupation was assessed, per study site, using multivariable regression. These estimates were then meta-analysed. Sensitivity analyses explored differences between sexes and gross national income. Results Overall, working in settings with potentially high exposure to dusts or fumes was associated with respiratory symptoms but not lung function differences. The most common occupation was farming. Compared to people not working in any of the 11 considered occupations, those who were farmers for ≥20 years were more likely to have chronic cough (OR 1.52, 95% CI 1.19–1.94), wheeze (OR 1.37, 95% CI 1.16–1.63) and dyspnoea (OR 1.83, 95% CI 1.53–2.20), but not lower FVC (β=0.02 L, 95% CI −0.02–0.06 L) or lower FEV1/FVC (β=0.04%, 95% CI −0.49–0.58%). Some findings differed by sex and gross national income. Conclusion At a population level, the occupational exposures considered in this study do not appear to be major determinants of differences in lung function, although they are associated with more respiratory symptoms. Because not all work settings were included in this study, respiratory surveillance should still be encouraged among high-risk dusty and fume job workers, especially in low- and middle-income countries.publishedVersio

    Cohort Profile: Burden of Obstructive Lung Disease (BOLD) study

    The Burden of Obstructive Lung Disease (BOLD) study was established to assess the prevalence of chronic airflow obstruction, a key characteristic of chronic obstructive pulmonary disease, and its risk factors in adults (≥40 years) from general populations across the world. The baseline study was conducted between 2003 and 2016, in 41 sites across Africa, Asia, Europe, North America, the Caribbean and Oceania, and collected high-quality pre- and post-bronchodilator spirometry from 28 828 participants. The follow-up study was conducted between 2019 and 2021, in 18 sites across Africa, Asia, Europe and the Caribbean. At baseline, there were in these sites 12 502 participants with high-quality spirometry. A total of 6452 were followed up, with 5936 completing the study core questionnaire. Of these, 4044 also provided high-quality pre- and post-bronchodilator spirometry. On both occasions, the core questionnaire covered information on respiratory symptoms, doctor diagnoses, health care use, medication use and ealth status, as well as potential risk factors.     Dans la gestion de l’édentement antérieur, l’esthétique est l’un des paramètres essentiels. Il dépend de l’étendue de l’édentement, de la valeur de la crête alvéolaire du maxillaire supérieur ainsi que de la qualité du guide antérieur. Mais la recherche de l’esthétique dento-labiale a surtout une dimension humaine qui dépasse le cadre des techniques opératoires. Le praticien fait donc face à un double défi : – recopier la nature pour créer l’illusion du naturel en denture artificielle, tout en respectant les différents paramètres biologiques (le guide antérieur, la santé parodontale...); – répondre à la demande immédiate du patient qui obéit au canon socio-culturel occidental de dents jeunes, claires, petites et bien rangées. Face à ce défi, la prise en compte d’une chronologie raisonnée de réalisation du plan de traitement s’avère judicieuse pour faciliter les étapes thérapeutiques et diminuer la temporisation. Nous allons exposer, à travers un cas clinique, les différentes étapes (parodontales et prothétiques) de la gestion d’une réhabilitation prothétique antérieure
