    Developing a model for e-prints and open access journal content in UK further and higher education

    A study carried out for the UK Joint Information Systems Committee examined models for the provision of access to material in institutional and subject-based archives and in open access journals. Their relative merits were considered, addressing not only technical concerns but also how e-print provision (by authors) can be achieved – an essential factor for an effective e-print delivery service (for users). A "harvesting" model is recommended, where the metadata of articles deposited in distributed archives are harvested, stored and enhanced by a national service. This model has major advantages over the alternatives of a national centralized service or a completely decentralized one. Options for the implementation of a service based on the harvesting model are presented

    Methods to Isolate Possible Bacteriophage for Micrococcus Luteus and Acinetobacter Baumannii

    The increasing prevalence of antibiotic-resistant strains of bacteria has led to a crisis in treatment options. Acinetobacter baumannii is an example of a bacterium that has developed a dangerous level of multidrug resistance. Not only does it have genes allowing for the resistance to antibiotics, but it also produces a biofilm that protects it. In recent years, A. baumannii has become a major contributor to nosocomial infections making it critical to develop new treatment methods. Micrococcus luteus, while typically not thought of as a pathogen, is also developing a resistance to antibiotics. M. luteus is capable of forming a biofilm on its own making it worrisome as it has increasingly been noted as an opportunistic pathogen. One potential new treatment of antibiotic resistance is the development of bacteriophage therapy, using bacterial viruses to target the infection and treat it. This study examines methods for isolating novel bacteriophage from dairy cattle feces, specifically for the biofilm producers A. baumannii and M. luteus

    A realist interview study of a participatory public mental health project “#KindnessByPost”

    Background: #KindnessByPost (KbP) is a participatory public health initiative in which people anonymously send and receive cards containing messages of goodwill with others also taking part in the programme. Quantitative evaluations of KbP consistently find evidence of improvements to people’s mental wellbeing and feelings of loneliness after participation and three months later. Our aim in the present study is to develop a programme theory of KbP, which describes for whom the KbP intervention improves mental wellbeing, other reported impacts, in which contexts it has these effects, and the mechanisms by which it works. Methods: We use a realist interviewing methodology to develop the programme theory. We conducted a focus group with the KbP executive team, and 20 one-to-one interviews with KbP participants. During analysis, a co-production working group iteratively developed a Theory of Change model comprising context-mechanism-outcome statements [CMOs] to map out the mechanisms present in KbP. Results: We developed 145 CMO statements, which we condensed and categorized into 32 overarching CMOs across nine thematic topics: access to scheme; pathways to involvement; resources; culture; giving post; receiving post; content of received post; community; long term impact. These CMOs set out pathways through which KbP benefited participants, including from doing something kind for someone else, of receiving post and appreciating the effort that went into it, and from the creative process of creating post and writing the messages inside them. Effects were sustained in part through people keeping the cards and through the social media communities that emerged around KbP. Discussion: Both giving and receiving post and the sense of community benefited participants and improved their mood and feelings of connectedness with others. Connection with a stranger, rather than friends or family, was also an important feature of the initiative for participants. Our wide range of CMO pathways by which KbP produced positive outcomes may mean that the intervention is applicable or adaptable across many communities and settings. Taken together with evidence from the quantitative evaluations, KbP is potentially an effective, low-cost, and highly scalable public health intervention for reducing loneliness and improving wellbeing.</p

    Replacement of the Trabecular Meshwork Cells—A Way Ahead in IOP Control?

    Glaucoma is one of the leading causes of vision loss worldwide, characterised with irreversible optic nerve damage and progressive vision loss. Primary open-angle glaucoma (POAG) is a subset of glaucoma, characterised by normal anterior chamber angle and raised intraocular pressure (IOP). Reducing IOP is the main modifiable factor in the treatment of POAG, and the trabecular meshwork (TM) is the primary site of aqueous humour outflow (AH) and the resistance to outflow. The structure and the composition of the TM are key to its function in regulating AH outflow. Dysfunction and loss of the TM cells found in the natural ageing process and more so in POAG can cause abnormal extracellular matrix (ECM) accumulation, increased TM stiffness, and increased IOP. Therefore, repair or regeneration of TM’s structure and function is considered as a potential treatment for POAG. Cell transplantation is an attractive option to repopulate the TM cells in POAG, but to develop a cell replacement approach, various challenges are still to be addressed. The choice of cell replacement covers autologous or allogenic approaches, which led to investigations into TM progenitor cells, induced pluripotent stem cells (iPSCs), and mesenchymal stem cells (MSCs) as potential stem cell source candidates. However, the potential plasticity and the lack of definitive cell markers for the progenitor and the TM cell population compound the biological challenge. Morphological and differential gene expression of TM cells located within different regions of the TM may give rise to different cell replacement or regenerative approaches. As such, this review describes the different approaches taken to date investigating different cell sources and their differing cell isolation and differentiation methodologies. In addition, we highlighted how these approaches were evaluated in different animal and ex vivo model systems and the potential of these methods in future POAG treatment

    Image-based monitoring for early detection of fouling in crystallisation processes

    Fouling or encrustation is a significant problem in continuous crystallisation processes where crystal deposits at surfaces impede heat transfer, increase flow resistance and reduce product quality. This paper proposes an automatic algorithm to detect early stages of fouling using images of vessel surfaces from commodity cameras. Statistical analysis of the pixel intensity variation offers the ability to distinguish appearance of crystals in the bulk solution and on the crystalliser walls. This information is used to develop a fouling metric indicator and determine separately induction times for appearance of first crystals at the surfaces and in the bulk. A method to detect process state changes using Bayesian online change point detection is also proposed, where the first change point is used to determine induction time either at the surface or in the bulk, based on real-time online measurements without using any predetermined threshold which usually varies between experiments and depends on data acquisition equipment. This approach can be used for in situ monitoring of early signs of encrustation to allow early warning for corrective actions to be taken when operating continuous crystallisation processes

    We Are Not Pontius Pilate: Acknowledging Ethics and Policy

    A new AI system is being developed to optimize vaccination strategies based on the structure and shape of a community's social contact network. The technology is minimally constrained and not bound by preconceived notions or human biases. With this come novel outside the box strategies; however, the system is only capable of optimizing what it is instructed to optimize, and does not consider any ethical or political concerns. With the growing concern for systematic discrimination as a result of artificial intelligence, we acknowledge a number of relevant issues that may arise as a consequence of our new technology and categorize them into three classes. We also introduce four normative ethical approaches that are used as a framework for decision-making. Despite the focus on vaccination strategies, our goal is to improve the discussions surrounding public concern and trust over artificial intelligence and demonstrate that artificial intelligence practitioners are addressing these concerns.Natural Sciences and Engineering Research Council of Canad

    Newly Developed and Validated Eosinophilic Esophagitis Histology Scoring System and Evidence that it Outperforms Peak Eosinophil Count for Disease Diagnosis and Monitoring

    Eosinophilic esophagitis is diagnosed by symptoms, and at least 15 intraepithelial eosinophils per high power field in an esophageal biopsy. Other pathologic features have not been emphasized. We developed a histology scoring system for esophageal biopsies that evaluates eight features: eosinophil density, basal zone hyperplasia, eosinophil abscesses, eosinophil surface layering, dilated intercellular spaces, surface epithelial alteration, dyskeratotic epithelial cells and lamina propria fibrosis. Severity (grade) and extent (stage) of abnormalities were scored using a 4 point scale (0 normal; 3 maximum change). Reliability was demonstrated by strong to moderate agreement among 3 pathologists who scored biopsies independently (p≀0.008). Several features were often abnormal in 201 biopsies (101 distal, 100 proximal) from 104 subjects (34 untreated, 167 treated). Median grade and stage scores were significantly higher in untreated compared to treated subjects (p≀0.0062). Grade scores for features independent of eosinophil counts were significantly higher in biopsies from untreated compared to treated subjects (basal zone hyperplasia p≀0.024 and dilated intercellular spaces p≀0.005), and were strongly correlated (r-square\u3e0.67). Principal components analysis identified 3 principal components that explained 78.2% of the variation in the features. In logistic regression models, 2 principal components more closely associated with treatment status than log distal peak eosinophil count (r-square 17, area under the curve 77.8 vs r-square 9, area under the curve 69.8). In summary, the eosinophilic esophagitis histology scoring system provides a method to objectively assess histologic changes in the esophagus beyond eosinophil number. Importantly, it discriminates treated from untreated patients, uses features commonly found in such biopsies, and is utilizable by pathologists after minimal training. These data provide rationales and a method to evaluate esophageal biopsies for features in addition to peak eosinophil count
