    Wirkstoffsuche und Charakterisierung mit BIACORE®

    BIACORE Systeme nutzen die Oberflächen-Plasmonen-Resonanz (SPR) zur Messung von Wechselwirkungen zwischen Biomolekülen. Dabei wird dem an einem Sensor Chip immobilisiertem Molekül der zu testende Reaktionspartner mittels eines Fließinjektionssystems zugeführt. Diese Technologie ist in den zehn Jahren seit Ihrer Einführung zu einem unverzichtbaren Werkzeug geworden, um Wechselwirkungen zwischen Proteinen und anderen Makromolekülen qualitativ (Spezifität) und quantitativ (Konzentration, Affinitätskonstanten, Geschwindigkeitskonstanten) zu beschreiben [Grundlagen in 1]. Fortschritte in der Meßtechnik und der Anwendungsentwicklung erlauben heute auch die direkte Messung von Substanzen mit einem Molekulargewicht von weniger als 200 Da. Bei der Suche nach pharmazeutischen Wirkstoffen werden Substanzen mit relative geringem Molekulargewicht durch verschiedene Prozesse modifiziert, um eine optimale Bindung an die Zielstruktur zu erreichen. Die Einführung von Markierungen ist oft ein größerer Eingriff als die ursprünglich geplante Modifikation. Daher ist gerade bei der Suche nach Wirkstoffen bzw. bei deren weitergehender Charakterisierung die SPR Biosensorik als markierungsfreie Technik ein wichtiges Werkzeug. [Übersicht in 2]. Biacore hat zusammen mit seinen Kooperationspartnern eine Reihe verschiedener Testsysteme untersucht und mit der Entwicklung von spezialisierten BIACORE Systemen begonnen. Im Vortrag werden BIACORE Anwendungen zur Untersuchung von HIV-Protease- und Thrombininhibitoren vorgestellt. Es handelt sich hierbei um Beispiele für das Sekundär Screening mit BIACORE. Weitere Themen im Prozeß der Wirkstoffsuche sind 'lead optimisation' und 'admission, distribution, metabolisation, excretion' (ADME). Hierzu wird die Bindung von Wirkstoffen an humanes Serumalbumin (HSA), an alpha-1-Glycoprotein (AGP), sowie an Lipidoberflächen gezeigt [3]. Zum Abschluß wird ein Ausblick auf die Entwicklung der High Performance Systeme und die SPR Array-Technologie gegeben

    Soil fertility management for organic rice production in the Lao PDR

    Rice is the most important agricultural commodity of the Lao People's Democratic Republic (Lao PDR), produced largely using traditional methods with limited inputs of fertilizers and other chemicals. The country has a wide diversity in rice production systems and rice varieties, with over 3000 different varieties recorded. The rich diversity and the production environment and methods are favorable for organic rice production. Investigations were carried out to describe soil fertility conditions, management practices, opportunities and problems associated with organic production methods for rice. Soils used for rice production are mostly of low fertility, with low organic matter and N-availability. In spite of this, virtually no fertilizer inputs are used for upland rice production. Inorganic fertilizer inputs for lowland rice production have increased rapidly over the past decade, but are still below 20 kg ha−1. The most important nutrient sources are rice straw and manure from buffalo and cattle. Chromolaena odorata plays an important role in nutrient cycling in upland rice systems and is sometimes added to lowland fields. In a range of fertility management studies, yield increase ranged from 2 to 89% for manure, straw or rice husk applied at modest rates (3 t ha−1), 32-156% for modest rates of inorganic fertilizer (60 kg N ha−1) and 36-167% for combined application of manure or crop residues with inorganic fertilizer. The response to locally produced commercial organic fertilizer was poor. The most promising inputs and strategies available to optimize yields in organic rice production systems are (1) optimizing use of locally available nutrients, mostly from manure, crop residues and weed biomass, (2) N addition through green manure and legumes growing in rotation and (3) additions of P through guano or rock-phosphate. The Lao PDR is fortunate to have substantial bat guano deposits in limestone caves. Extensive experience is available on straw and husk management for lowland systems and green manure species for upland production system

    Sugar-sweetened beverage consumption, correlates and interventions among Australian Aboriginal and Torres Strait Islander communities: a scoping review protocol

    Introduction: Aboriginal and Torres Strait Islander communities of Australia experience poorer health outcomes in the areas of overweight and obesity, diabetes and cardiovascular disease. Contributing to this burden of disease in the Australian community generally and in Aboriginal and Torres Strait Islander communities, is the consumption of sugar-sweetened beverages (SSBs). We have described a protocol for a review to systematically scope articles that document use of SSBs and interventions to reduce their consumption with Aboriginal and Torres Strait Islander people. These results will inform future work that investigates interventions aimed at reducing harm associated with SSB consumption. Methods and analysis: This scoping review draws on a methodology that uses a six-step approach to search databases including PubMed, SCOPUS, CINAHL, Informit (including Informit: Indigenous Peoples), Joanna Briggs Institute EBP Database and Mura, between January 1980 and February 2017. Two reviewers will be engaged to search for and screen studies independently, using formulated selection criteria, for inclusion in our review. We will include primary research studies, systematic reviews including meta-analysis or meta-synthesis, reports and unpublished grey literature. Results will be entered into a table identifying study details and characteristics, summarised using a Preferred Reporting Items for Systematic Reviews and Meta-Analysis chart and then critically analysed. Ethics and dissemination: This review will not require ethics committee review.     Evidence for softening of first-order transition in 3D by quenched disorder

    We study by extensive Monte Carlo simulations the effect of random bond dilution on the phase transition of the three-dimensional 4-state Potts model which is known to exhibit a strong first-order transition in the pure case. The phase diagram in the dilution-temperature plane is determined from the peaks of the susceptibility for sufficiently large system sizes. In the strongly disordered regime, numerical evidence for softening to a second-order transition induced by randomness is given. Here a large-scale finite-size scaling analysis, made difficult due to strong crossover effects presumably caused by the percolation fixed point, is performed.Comment: LaTeX file with Revtex, 4 pages, 4 eps figure
