    Contribution of nuclear texture analysis for the differential diagnosis of follicular lesions of the thyroid: comparison to immunohistochemical markers

    Objective and methods: To investigate the utility of nuclear chromatin texture assessment in the differential diagnosis of follicular patterned lesions, by means of examining 76 samples previously submitted to the immunohistochemical protein analysis of HBME-1, CK-19 and galectina-3. Results: HBME-1 confirmed to be the most sensitive marker of malignancy. A series of morphometric, densitometric and texture variables were useful in the discrimination of the different types of follicular lesions. Among these variables, r(2), a parameter related to the granularity of the nucleus presented the best accuracy, sensitivity, specificity and positive and negative predictive values, distinguishing benign from malignant lesions. Conclusion: The morphometric analysis of nuclear chromatin images may add accuracy to the differential diagnosis of follicular patterned lesions.     Rosiglitazone decreases intra- to extramyocellular fat ratio in obese non-diabetic adults with metabolic syndrome

    Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)Background Insulin resistance is intrinsically related to intramyocellular (IMCL) rather than extramyocellular (EMCL) triglyceride content. Conflicting results have been reported on the ability of insulin sensitizer agents, such as thiazolidinediones, to modify muscle fat distribution. The aim of this study was to investigate the role of rosiglitazone on muscle fat compartment distribution in an adult population of obese non-diabetic metabolic syndrome patients. Patients and methods Fifteen obese, non-diabetic, metabolic syndrome patients were studied by means of proton nuclear magnetic resonance ((1)H-NMR) spectroscopy before and after treatment with rosiglitazone 8 mg/day for 6 months. Anthropometrical and metabolic variables were assessed. Results After rosiglitazone, body weight and hip circumference increased [100.9 (91.12-138.7) vs. 107.0 (79.6-142.8) kg and 118 (107-126) vs. 122 (110-131) cm]; while waist-hip ratio (WHR) decreased from 0.93 (0.87-1.00) to 0.89 (0.82-0.97) (P < 0.001 for all). Additionally, fasting plasma glucose, insulin and homeostatis model assessment of insulin resistance (HOMA-IR) significantly decreased while adiponectin increased over threefold [9.7 (3.7-17.7) vs. 38.0 (19.3-42.4) mu g/ml] without any changes in resistin. Finally, the IMCL did not change [267.54 (213.94-297.94) vs. 305.75 (230.80-424.75) arbitrary units (AU), P = 0.15] while the EMCL increased [275.53 (210.39-436.66) vs. 411.39 (279.92-556.59) AU; P < 0.01] therefore decreasing the IMCL-to-EMCL (IMCL/EMCL) ratio [1.07 (0.78-1.23) vs. 0.71 (0.53-0.96); P < 0.01]. Conclusion Rosiglitazone treatment increased body weight and hip circumference and decreased WHR.     Phage inducible islands in the gram-positive cocci

    The SaPIs are a cohesive subfamily of extremely common phage-inducible chromosomal islands (PICIs) that reside quiescently at specific att sites in the staphylococcal chromosome and are induced by helper phages to excise and replicate. They are usually packaged in small capsids composed of phage virion proteins, giving rise to very high transfer frequencies, which they enhance by interfering with helper phage reproduction. As the SaPIs represent a highly successful biological strategy, with many natural Staphylococcus aureus strains containing two or more, we assumed that similar elements would be widespread in the Gram-positive cocci. On the basis of resemblance to the paradigmatic SaPI genome, we have readily identified large cohesive families of similar elements in the lactococci and pneumococci/streptococci plus a few such elements in Enterococcus faecalis. Based on extensive ortholog analyses, we found that the PICI elements in the four different genera all represent distinct but parallel lineages, suggesting that they represent convergent evolution towards a highly successful lifestyle. We have characterized in depth the enterococcal element, EfCIV583, and have shown that it very closely resembles the SaPIs in functionality as well as in genome organization, setting the stage for expansion of the study of elements of this type.     EVH Black Holes, AdS3 Throats and EVH/CFT Proposal

    Within class of generic black holes there are extremal black holes (with vanishing Hawking temperature T) and vanishing horizon area Ah, but with finite Ah/T ratio,the Extremal Vanishing Horizon (EVH) black holes. We study the near horizon limit of a four dimensional EVH black hole solution to a generic (gauged) Einstein-Maxwell dilaton theory and show that in the near horizon limit they develop a throat which is a pinching orbifold limit of AdS3. This is an extension of the well known result for extremal black holes the near horizon limit of which contains an AdS2 throat. We show that in the near EVH near horizon limit the pinching AdS3 factor turns to a pinching BTZ black hole and that this near horizon limit is indeed a decoupling limit. We argue that the pinching AdS3 or BTZ orbifold is resolved if the near horizon limit is accompanied by taking the 4d Newton constant G4 to zero such that the Bekenstein-Hawking entropy S = Ah/(4G4) remains finite. We propose that in this limit the near horizon EVH black hole is dual to a 2d CFT. We provide pieces of evidence in support of the EVH/CFT correspondence and comment on its connection to the Kerr/CFT proposal and speculations how the EVH/CFT may be used to study generic e.g. Schwarzchild-type black holes.Comment: 31 pages, 3 figures, JHEP styl
