    An Attempt to Probe the Radio Jet Collimation Regions in NGC 4278, NGC 4374 (M84), and NGC 6166

    NRAO Very Long Baseline Array (VLBA) observations of NGC 4278, NGC 4374 (M84), NGC 6166, and M87 (NGC 4486) have been made at 43 GHz in an effort to image the jet collimation region. This is the first attempt to image the first three sources at 43 GHz using Very Long Baseline Interferometry (VLBI) techniques. These three sources were chosen because their estimated black hole mass and distance implied a Schwarzschild radius with large angular size, giving hope that the jet collimation regions could be studied. Phase referencing was utilize for the three sources because of their expected low flux densities. M87 was chosen as the calibrator for NGC 4374 because it satisfied the phase referencing requirements: nearby to the source and sufficiently strong. Having observed M87 for a long integration time, we have detected its sub-parsec jet, allowing us to confirm previous high resolution observations made by Junor, Biretta & Livio, who have indicated that a wide opening angle was seen near the base of the jet. Phase referencing successfully improved our image sensitivity, yielding detections and providing accurate positions for NGC 4278, NGC 4374 and NGC 6166. These sources are point dominated, but show suggestions of extended structure in the direction of the large-scale jets. However, higher sensitivity will be required to study their sub-parsec jet structure

    TARGET: Rapid Capture of Process Knowledge

    TARGET (Task Analysis/Rule Generation Tool) represents a new breed of tool that blends graphical process flow modeling capabilities with the function of a top-down reporting facility. Since NASA personnel frequently perform tasks that are primarily procedural in nature, TARGET models mission or task procedures and generates hierarchical reports as part of the process capture and analysis effort. Historically, capturing knowledge has proven to be one of the greatest barriers to the development of intelligent systems. Current practice generally requires lengthy interactions between the expert whose knowledge is to be captured and the knowledge engineer whose responsibility is to acquire and represent the expert's knowledge in a useful form. Although much research has been devoted to the development of methodologies and computer software to aid in the capture and representation of some types of knowledge, procedural knowledge has received relatively little attention. In essence, TARGET is one of the first tools of its kind, commercial or institutional, that is designed to support this type of knowledge capture undertaking. This paper will describe the design and development of TARGET for the acquisition and representation of procedural knowledge. The strategies employed by TARGET to support use by knowledge engineers, subject matter experts, programmers and managers will be discussed. This discussion includes the method by which the tool employs its graphical user interface to generate a task hierarchy report. Next, the approach to generate production rules for incorporation in and development of a CLIPS based expert system will be elaborated. TARGET also permits experts to visually describe procedural tasks as a common medium for knowledge refinement by the expert community and knowledge engineer making knowledge consensus possible. The paper briefly touches on the verification and validation issues facing the CLIPS rule generation aspects of TARGET. A description of efforts to support TARGET's interoperability issues on PCs, Macintoshes and UNIX workstations concludes the paper

    Observations of the Structure and Dynamics of the Inner M87 Jet

    M87 is the best source in which to study a jet at high resolution in gravitational units because it has a very high mass black hole and is nearby. The angular size of the black hole is second only to Sgr A*, which does not have a strong jet. The jet structure is edge brightened with a wide opening angle base and a weak counterjet. We have roughly annual observations for 17 years plus intensive monitoring at three week intervals for a year and five day intervals for 2.5 months made with the Very Long Baseline Array (VLBA) at 43 GHz. The inner jet shows very complex dynamics, with apparent motions both along and across the jet. Speeds from zero to over 2c are seen, with acceleration observed over the first 3 milli-arcseconds. The counterjet decreases in brightness much more rapidly than the main jet, as is expected from relativistic beaming in an accelerating jet oriented near the line-of-sight. Details of the structure and dynamics are discussed. The roughly annual observations show side-to-side motion of the whole jet with a characteristic time scale of about 9 years.Comment: 11 pages, 7 figures.     Transcriptome analysis of bacteriophage communities in periodontal health and disease.

    BackgroundThe role of viruses as members of the human microbiome has gained broader attention with the discovery that human body surfaces are inhabited by sizeable viral communities. The majority of the viruses identified in these communities have been bacteriophages that predate upon cellular microbiota rather than the human host. Phages have the capacity to lyse their hosts or provide them with selective advantages through lysogenic conversion, which could help determine the structure of co-existing bacterial communities. Because conditions such as periodontitis are associated with altered bacterial biota, phage mediated perturbations of bacterial communities have been hypothesized to play a role in promoting periodontal disease. Oral phage communities also differ significantly between periodontal health and disease, but the gene expression of oral phage communities has not been previously examined.ResultsHere, we provide the first report of gene expression profiles from the oral bacteriophage community using RNA sequencing, and find that oral phages are more highly expressed in subjects with relative periodontal health. While lysins were highly expressed, the high proportion of integrases expressed suggests that prophages may account for a considerable proportion of oral phage gene expression. Many of the transcriptome reads matched phages found in the oral cavities of the subjects studied, indicating that phages may account for a substantial proportion of oral gene expression. Reads homologous to siphoviruses that infect Firmicutes were amongst the most prevalent transcriptome reads identified in both periodontal health and disease. Some genes from the phage lytic module were significantly more highly expressed in subjects with periodontal disease, suggesting that periodontitis may favor the expression of some lytic phages.ConclusionsAs we explore the contributions of viruses to the human microbiome, the data presented here suggest varying expression of bacteriophage communities in oral health and disease

    Overproduction of immunoglobulin light chains leads to systemic amyloidosis, a lethal disease characterized by the formation of amyloid fibrils in patients' tissues. Excess light chains are in equilibrium between dimers and less stable monomers which can undergo irreversible aggregation to the amyloid state. The dimers therefore must disassociate into monomers prior to forming amyloid fibrils. Here we identify ligands that inhibit amyloid formation by stabilizing the Mcg light chain variable domain dimer and shifting the equilibrium away from the amyloid-prone monomer

    Optometrists' attitudes toward using OCT angiography lag behind other retinal imaging types

    Purpose: While optometrists' attitudes toward established retinal imaging types are generally positive, they are unknown for optical coherence tomography angiography (OCTA). We performed a cross-sectional survey to estimate attitudes toward OCTA and identify clinician and/or practice characteristics that influence them. Methods: A paper-based survey was mailed to 252 randomly selected optometrists in Australia. Five-point Likert-scale items from a previous survey assessing attitudes toward new technology were included to probe respondent characteristics and attitudes toward retinal imaging. Performance expectancy attitudes toward OCTA were elicited by the statement ‘I believe OCTA is useful in daily practice’. Mean scores out of five (mean [SD]) were rounded and mapped to appropriate descriptive statements. Results: The response rate was 47% (118/252). The mean (SD) age of respondents was 44.0 (13.8) years and 50.8% (60/118) were female. Optometrists had 19.9 (14.0) years of clinical experience and 66.9% (79/118) worked at independent practices. In total, 8.5% (10/118) of respondents used OCTA to provide clinical care. Optometrists agreed that optical coherence tomography (OCT), colour fundus imaging, ultra-wide field imaging and fundus autofluorescence (mean scores 3.6–4.7 out of 5) were useful in daily practice but felt neutral about whether OCTA was useful (3.4 [0.8]). Optometrists believed that OCTA was less enjoyable to use (p < 0.0001), less endorsed by peers (p < 0.0001) and felt less confident that they had the knowledge to interpret OCTA (p < 0.0001) compared to other retinal imaging types. Conclusions: Optometrists are undecided on whether OCTA is useful in daily practice and had lower expectations that using OCTA would confer job performance benefits compared to other retinal imaging types. Further work is needed to advocate the benefits of using OCTA across the profession
