miR-9 Acts as an OncomiR in Prostate Cancer through Multiple Pathways That Drive Tumour Progression and Metastasis

    Get PDF
Identification of dysregulated microRNAs (miRNAs) in prostate cancer is critical not only for diagnosis, but also differentiation between the aggressive and indolent forms of the disease. miR-9 was identified as an oncomiR through both miRNA panel RT-qPCR as well as high-throughput sequencing analysis of the human P69 prostate cell line as compared to its highly tumorigenic and metastatic subline M12, and found to be consistently upregulated in other prostate cell lines including DU-145 and PC3. While miR-9 has been characterized as dysregulated either as an oncomiR or tumour suppressor in a variety of other cancers including breast, ovarian, and nasopharyngeal carcinomas, it has not been previously evaluated and proven as an oncomiR in prostate cancer. miR-9 was confirmed an oncomiR when found to be overexpressed in tumour tissue as compared to adjacent benign glandular epithelium through laser-capture microdissection of radical prostatectomy biopsies. Inhibition of miR-9 resulted in reduced migratory and invasive potential of the M12 cell line, and reduced tumour growth and metastases in male athymic nude mice. Analysis showed that miR-9 targets e-cadherin and suppressor of cytokine signalling 5 (SOCS5), but not NF-ĸB mRNA. Expression of these proteins was shown to be affected by modulation in expression of miR-9

Extraction of the Axial Nucleon Form Factor from Neutrino Experiments on Deuterium

We present new parameterizations of vector and axial nucleon form factors. We maintain an excellent descriptions of the form factors at low momentum transfers (Q2Q^2), where the spatial structure of the nucleon is important, and use the Nachtman scaling variable ξ\xi to relate elastic and inelastic form factors and impose quark-hadron duality constraints at high Q2Q^2 where the quark structure dominates. We use the new vector form factors to re-extract updated values of the axial form factor from \numu experiments on deuterium. We obtain an updated world average value from \numud, \numubarH and pion electroproduction experiments of MAM_{A} = 1.014±0.014GeV/c21.014 \pm 0.014 GeV/c^2. Our parameterizations are useful in modeling ν\nu interactions at low energies (e.g. for \numu oscillations experiments). The predictions for high Q2Q^2 can be tested in the next generation electron and \numu scattering experiments.Comment: Presented by A. Ambiguities of neutrino(antineutrino) scattering on the nucleon due to the uncertainties of relevant strangeness form factors

Strange quark contributions to neutrino(antineutrino) scattering are investigated on the nucleon level in the quasi-elastic region. The incident energy range between 500 MeV and 1.0 GeV is used for the scattering. All of the physical observable by the scattering are investigated within available experimental and theoretical results for the strangeness form factors of the nucleon. In specific, a newly combined data of parity violating electron scattering and neutrino scattering is exploited. Feasible quantities to be explored for the strangeness contents are discussed for the application to neutrino-nucleus scattering.Comment: 17 pages, 7 figures, submit to J. Phys.

Partitioning of the global fossil CO2 sink using a 19-year trend in atmospheric O2

O2/N2 is measured in the Cape Grim Air Archive (CGAA), a suite of tanks filled with background air at Cape Grim, Tasmania (40.7°S, 144.8°E) between April 1978 and January 1997. Derived trends are compared with published O2/N2 records and assessed against limits on interannual variability of net terrestrial exchanges imposed by trends of δ13C in CO2. Two old samples from 1978 and 1987 and eight from 1996/97 survive critical selection criteria and give a mean 19-year trend in δ(O2/N2) of -16.7 ± 0.5 per meg yr-1, implying net storage of +2.3 ± 0.7 GtC (1015 g carbon) yr-1 of fossil fuel CO2 in the oceans and +0.2 ± 0.9 GtC yr-1 in the terrestrial biosphere. The uptake terms are consistent for both O2/N2 and δ13C tracers if the mean 13C isotopic disequilibrium flux, combining terrestrial and oceanic contributions, is 93 ± 15 GtC ‰ yr-1. Baby Universes Revisited

The behaviour of baby universes has been an important ingredient in understanding and quantifying non-critical string theory or, equivalently, models of two-dimensional Euclidean quantum gravity coupled to matter. Within a regularized description based on dynamical triangulations, we amend an earlier conjecture by Jain and Mathur on the scaling behaviour of genus-gg surfaces containing particular baby universe `necks', and perform a nontrivial numerical check on our improved conjecture.Comment: 10 pages, 1 figur

The archival version is available at the link below.Despite many researches over the last half century, the mechanism of ac powder electroluminescence remains to be fully elucidated and, to this end, a better understanding of the relatively complex structure of alternate current electroluminescence (ACEL) phosphors is required. Consequently, the structure and morphology of ZnS:Cu,Cl phosphor powders have been investigated herein by means of scanning electron microscopy (SEM) on hydrochloric acid-etched samples and X-ray powder diffraction. The latter technique confirmed that, as a result of two-stage firing during their synthesis, the phosphors were converted from the high temperature hexagonal (wurtzite) structure to the low temperature cubic (sphalerite) polymorph having a high density of planar stacking faults. Optical microscopy revealed that the crystal habit of the phosphor had the appearance of the hexagonal polymorph, which can be explained by the sphalerite pseudomorphing of the earlier wurtzite after undergoing the hexagonal to cubic phase transformation during the synthesis. SEM micrographs of the hydrochloric-etched phosphor particles revealed etch pits, a high density of planar stacking faults along the cubic [111] axis, and the pyramids on the (111) face. These observations were consistent with unidirectional crystal growth originating from the face showing the pyramids.EPSRC, DTI, and the Technology Strategy Board-led Technology Program

A revised edition of the readiness to change questionnaire (treatment version)

    Get PDF
The UK Alcohol Treatment Trial provided an opportunity to examine the factor structure of the Readiness to Change Questionnaire-Treatment Version (RCQ[TV]) in a large sample (N = 742) of individuals in treatment for alcohol problems who were given the RCQ[TV] at baseline, 3-months and 12-months follow-up. Confirmatory factor analysis of the previously reported factor structure (5 items for each of Precontemplation, Contemplation and Action scales) resulted in a relatively poor fit to the data. Removal of one item from each of the scales resulted in a 12-item instrument for which goodness-of-fit indices were improved, without loss of internal consistency of the three scales, on all three measurement occasions. Inspection of relationships between stage allocation by the new instrument and negative alcohol outcome expectancies provided evidence of improved construct validity for the revised edition of the RCQ[TV]. There was also a strong relationship between stage allocation at 3-months follow-up and outcome of treatment at 12 months. The revised edition of the RCQ[TV] offers researchers and clinicians a shorter and improved measurement of stage of change in the alcohol treatment population
