    Fluid intake patterns of children and adolescents: results of six Liq.In7 national cross-sectional surveys

    Purpose: This study aimed to identify and characterize patterns of fluid intake in children and adolescents from six countries: Argentina, Brazil, China, Indonesia, Mexico and Uruguay. Methods: Data on fluid intake volume and type amongst children (4–9 years; N = 1400) and adolescents (10–17 years; N = 1781) were collected using the validated 7-day fluid-specific record (Liq.In7 record). To identify relatively distinct clusters of subjects based on eight fluid types (water, milk and its derivatives, hot beverages, sugar-sweetened beverages (SSB), 100% fruit juices, artificial/non-nutritive sweetened beverages, alcoholic beverages, other beverages), a cluster analysis (partitioning around k-medoids algorithm) was used. Clusters were then characterized according to their socio-demographics and lifestyle indicators. Results: The six interpretable clusters identified were: low drinkers–SSB (n 523), low drinkers–water and milk (n 615), medium mixed drinkers (n 914), high drinkers–SSB (n 513), high drinkers–water (n 352) and very high drinkers–water (n 264). Country of residence was the dominant characteristic, followed by socioeconomic level, in all six patterns. Conclusions: This analysis showed that consumption of water and SSB were the primary drivers of the clusters. In addition to country, socio-demographic and lifestyle factors played a role in determining the characteristics of each cluster. This information highlights the need to target interventions in particular populations aimed at changing fluid intake behavior and improving health in children and adolescents

    An assessment of the high-entropy alloy system VCrMnFeAlx

    A major consideration when choosing materials for nuclear reactors is the future radioactive waste produced by the irradiation of their component parts. Only a handful of structural metals can be considered ‘low activation’ in a fusion environment. In recent work, we showed that the low-activation multicomponent equiatomic alloy VCrMnFe comprises a single BCC (A2) phase at 1200°C. Here, we examine its stability on ageing at lower temperatures, and the effect of Al additions (to create VCrMnFeAlx alloys) to destabilise the sigma phase and form strengthening superlattice structures. It is found that substantial volume fractions of sigma phase form after ageing VrCrMnFe at 600°C and 800°C for 1000 h. The addition of Al was found to destabilise the sigma phase, as predicted using thermodynamic modelling, with it being eliminated at all temperatures with additions of 6.6 at% Al. Increasing Al additions also led to the formation of superlattice structures: B2 and L21 (Heusler). Higher Al content had a slight increasing effect on the alloys’ hardness, but also embrittled the alloys (at room temperature). Significant hardening was produced by nano-segregation induced in the higher Al x = 0.25, 0.5 and 1.0 alloys after aging at 600 °C. This alloy system presents an attractive opportunity to fine-tune the composition to obtain a balance of ductility and high-temperature strength and stability. Of particular interest was the formation a two-phase basket weave cube-on-cube orientated, coherent, microstructure in VCrMnFeAl1.0 after aging at 800 °C

    Techniques to Facilitate the Acquisition of Accurate Spectral Measurements and Multispectral Imagery of Plant Foliage Under Artificial Lighting Conditions

    Spectral measurements obtained in the laboratory under artificial lighting sources have been used for many years to develop spectral ‘libraries’ for various soil types, rocks and minerals, and other inanimate features occurring on or near the earth’s surface. Quartz halogen lamps have been shown to emit all of the electromagnetic (EMR) wavelengths required for the acquisition of quality multispectral imagery, and various techniques have been developed to facilitate the acquisition of accurate spectral measurements using such artificial lighting sources. Our objectives in this study were to evaluate the several factors of critical importance in obtaining accurate spectral measurements for plant foliage in the laboratory, including the effects of leaf orientation, excision, and background reflectance on reflectance of visible and NIR wavelengths. Results demonstrated that accurate spectral measurements and imagery of various types of foliage may be obtained using quartz halogen lighting provided that excised leaf samples are placed on an NIR-absorbent background and samples are maintained in containers designed to minimize desiccation

    Upregulation of mucin glycoprotein MUC1 in the progression to esophageal adenocarcinoma and therapeutic potential with a targeted photoactive antibody-drug conjugate

    BACKGROUND: Mucin glycoprotein 1 (MUC1) is a glycosylated transmembrane protein on epithelial cells. We investigate MUC1 as a therapeutic target in Barrett's epithelium (BE) and esophageal adenocarcinoma (EA) and provide proof of concept for a light based therapy targeting MUC1. RESULTS: MUC1 was present in 21% and 30% of significantly enriched pathways comparing BE and EA to squamous epithelium respectively. MUC1 gene expression was x2.3 and x2.2 higher in BE (p=<0.001) and EA (p=0.03). MUC1 immunohistochemical expression increased during progression to EA and followed tumor invasion. HuHMFG1 based photosensitive antibody drug conjugates (ADC) showed cell internalization, MUC1 selective and light-dependent cytotoxicity (p=0.0006) and superior toxicity over photosensitizer alone (p=0.0022). METHODS: Gene set enrichment analysis (GSEA) evaluated pathways during BE and EA development and quantified MUC1 gene expression. Immunohistochemistry and flow cytometry evaluated the anti-MUC1 antibody HuHMFG1 in esophageal cells of varying pathological grade. Confocal microscopy examined HuHMFG1 internalization and HuHMFG1 ADCs were created to deliver a MUC1 targeted phototoxic payload. CONCLUSIONS: MUC1 is a promising target in EA. Molecular and light based targeting of MUC1 with a photosensitive ADC is effective in vitro and after development may enable treatment of locoregional tumors endoscopically

    Tensor completion in hierarchical tensor representations

    Compressed sensing extends from the recovery of sparse vectors from undersampled measurements via efficient algorithms to the recovery of matrices of low rank from incomplete information. Here we consider a further extension to the reconstruction of tensors of low multi-linear rank in recently introduced hierarchical tensor formats from a small number of measurements. Hierarchical tensors are a flexible generalization of the well-known Tucker representation, which have the advantage that the number of degrees of freedom of a low rank tensor does not scale exponentially with the order of the tensor. While corresponding tensor decompositions can be computed efficiently via successive applications of (matrix) singular value decompositions, some important properties of the singular value decomposition do not extend from the matrix to the tensor case. This results in major computational and theoretical difficulties in designing and analyzing algorithms for low rank tensor recovery. For instance, a canonical analogue of the tensor nuclear norm is NP-hard to compute in general, which is in stark contrast to the matrix case. In this book chapter we consider versions of iterative hard thresholding schemes adapted to hierarchical tensor formats. A variant builds on methods from Riemannian optimization and uses a retraction mapping from the tangent space of the manifold of low rank tensors back to this manifold. We provide first partial convergence results based on a tensor version of the restricted isometry property (TRIP) of the measurement map. Moreover, an estimate of the number of measurements is provided that ensures the TRIP of a given tensor rank with high probability for Gaussian measurement maps.Comment: revised version, to be published in Compressed Sensing and Its Applications (edited by H.     BACKGROUND: Recent reports suggest that latrepirdine (Dimebon, dimebolin), a retired Russian antihistamine, improves cognitive function in aged rodents and in patients with mild to moderate Alzheimer's disease (AD). However, the mechanism(s) underlying this benefit remain elusive. AD is characterized by extracellular accumulation of the amyloid-beta (Abeta) peptide in the brain, and Abeta-lowering drugs are currently among the most popular anti-amyloid agents under development for the treatment of AD. In the current study, we assessed the effect of acute dosing of latrepirdine on levels of extracellular Abeta using in vitro and in vivo experimental systems. RESULTS: We evaluated extracellular levels of Abeta in three experimental systems, under basal conditions and after treatment with latrepirdine. Mouse N2a neuroblastoma cells overexpressing Swedish APP were incubated for 6 hr in the presence of either vehicle or vehicle + latrepirdine (500pM-5 muM). Synaptoneurosomes were isolated from TgCRND8 mutant APP-overexpressing transgenic mice and incubated for 0 to 10 min in the absence or presence of latrepirdine (1 muM or 10 muM). Drug-naĂŻve Tg2576 Swedish mutant APP overexpressing transgenic mice received a single intraperitoneal injection of either vehicle or vehicle + latrepirdine (3.5 mg/kg). Picomolar to nanomolar concentrations of acutely administered latrepirdine increased the extracellular concentration of Abeta in the conditioned media from Swedish mutant APP-overexpressing N2a cells by up to 64% (p = 0.01), while a clinically relevant acute dose of latrepirdine administered i.p. led to an increase in the interstitial fluid of freely moving APP transgenic mice by up to 40% (p = 0.01). Reconstitution of membrane protein trafficking and processing is frequently inefficient, and, consistent with this interpretation, latrepirdine treatment of isolated TgCRND8 synaptoneurosomes involved higher concentrations of drug (1-10 muM) and led to more modest increases in extracellular Abeta(x-42 )levels (+10%; p = 0.001); of note, however, was the observation that extracellular Abeta(x-40 )levels did not change. CONCLUSIONS: Here, we report the surprising association of acute latrepirdine dosing with elevated levels of extracellular Abeta as measured in three independent neuron-related or neuron-derived systems, including the hippocampus of freely moving Tg2576 mice.     Recent Advances in ÎŁ-definability over Continuous Data Types

    The purpose of this paper is to survey our recent research in computability and definability over continuous data types such as the real numbers, real-valued functions and functionals. We investigate the expressive power and algorithmic properties of the language of Sigma-formulas intended to represent computability over the real numbers. In order to adequately represent computability we extend the reals by the structure of hereditarily finite sets. In this setting it is crucial to consider the real numbers without equality since the equality test is undecidable over the reals. We prove Engeler's Lemma for Sigma-definability over the reals without the equality test which relates Sigma-definability with definability in the constructive infinitary language L_{omega_1 omega}. Thus, a relation over the real numbers is Sigma-definable if and only if it is definable by a disjunction of a recursively enumerable set of quantifier free formulas. This result reveals computational aspects of Sigma-definability and also gives topological characterisation of Sigma-definable relations over the reals without the equality test. We also illustrate how computability over the real numbers can be expressed in the language of Sigma-formulas
