    Delineation of Cavity in Downstream Surge Chamber at Punatsangchhu-II Hydroelectric Project, Bhutan

    On 3rd March, 2016, there occurred a massive roof fall incidence in one of the major caverns of Punatsangchhu-II Hydroelectric Project (PHEP-II), Bhutan which halted the operations in the major caverns and required additional strengthening measures. Prior to concluding anything on the treatment measures for the muck flown into the cavern and the cavity formed over the crown of Downstream Surge Chamber, there was a strong need to determine the extents of cavity. Even to understand the influence of cavity formation on adjacent caverns, the cavity needs to be delineated. Therefore, the management of PHEP-II adopted several techniques to decipher the shape and dimensions of the cavity like, surveying, geophysical, cross-hole seismic surveys, borehole scanning, and exploratory drilling. The tentative shape of the cavity could finally be established. The findings from each method are explained in this paper

    Antagonism of Tetherin Restriction of HIV-1 Release by Vpu Involves Binding and Sequestration of the Restriction Factor in a Perinuclear Compartment

    The Vpu accessory protein promotes HIV-1 release by counteracting Tetherin/BST-2, an interferon-regulated restriction factor, which retains virions at the cell-surface. Recent reports proposed β-TrCP-dependent proteasomal and/or endo-lysosomal degradation of Tetherin as potential mechanisms by which Vpu could down-regulate Tetherin cell-surface expression and antagonize this restriction. In all of these studies, Tetherin degradation did not, however, entirely account for Vpu anti-Tetherin activity. Here, we show that Vpu can promote HIV-1 release without detectably affecting Tetherin steady-state levels or turnover, suggesting that Tetherin degradation may not be necessary and/or sufficient for Vpu anti-Tetherin activity. Even though Vpu did not enhance Tetherin internalization from the plasma membrane (PM), it did significantly slow-down the overall transport of the protein towards the cell-surface. Accordingly, Vpu expression caused a specific removal of cell-surface Tetherin and a re-localization of the residual pool of Tetherin in a perinuclear compartment that co-stained with the TGN marker TGN46 and Vpu itself. This re-localization of Tetherin was also observed with a Vpu mutant unable to recruit β-TrCP, suggesting that this activity is taking place independently from β-TrCP-mediated trafficking and/or degradation processes. We also show that Vpu co-immunoprecipitates with Tetherin and that this interaction involves the transmembrane domains of the two proteins. Importantly, this association was found to be critical for reducing cell-surface Tetherin expression, re-localizing the restriction factor in the TGN and promoting HIV-1 release. Overall, our results suggest that association of Vpu to Tetherin affects the outward trafficking and/or recycling of the restriction factor from the TGN and as a result promotes its sequestration away from the PM where productive HIV-1 assembly takes place. This mechanism of antagonism that results in TGN trapping is likely to be augmented by β-TrCP-dependent degradation, underlining the need for complementary and perhaps synergistic strategies to effectively counteract the powerful restrictive effects of human Tetherin

    The Singhbhum craton, eastern India records multiple stages of emplacement of Precambrian dyke swarms with contrasting petrogenetic models proposed for their formation. In this study, we document elemental and Sr-Nd isotopic data for three major dyke swarms in the southern part of the craton, including the ca. 2.7 Ga Ghatgaon dyke swarm, the Early Proterozoic Keonjhar dyke swarm and the ca. 1.76 Ga Pipilia dyke swarm. Dyke compositions are dominated by basalt and basaltic andesite with minor andesite, showing trace element signatures typical of continental crustal rocks. Age-corrected Nd isotopic data for Ghatgaon (εNdt = −4.8 to + 4.6), Keonjhar (εNdt = −11.9 to + 3.8), and Pipilia (a single sample with εNdt = −8.8) dyke swarms display substantial variations. The lack of magma compositions that could indicate the presence of elevated mantle potential temperature among the rocks suggests melting regime was likely similar to the ambient mantle. The Dy/Yb and Dy/Dy* systematics of the rocks indicates melting occurred between spinel-stable depths and the spinel-garnet transition zone. The dominantly mafic compositions of the rocks and ubiquitous continental crustal trace element signature are best explained by peridotite source with recycled crustal components, probably in the form of pyroxenites. Our new Nd isotopic data, which argue against any simple secular evolution trend invoked in previous studies, indicate that crustal recycling was likely an episodic phenomenon rather than a discrete, single-stage process since the Archean. Geochemical modelling indicates that a sublithospheric mantle source with (10% or less) recycled crustal components satisfactorily explains the trace element variations of the dyke swarms.ISSN:2296-646

    Not AvailableIn a tropical country like India, thermal stress is one of the major factors which significantly affects the productivity of dairy cattle. The present study was aimed to identify the effect of heat and cold stress on cell viability, mitogen stimulation indices, nitric oxide production and HSP70 expression in Sahiwal and Holstein crossbred (Frieswal) population in India. The results indicated that the Sahiwal breed can better withstand the effect of heat and cold stress significantly (P<0.05) when compared to the crossbred cattle due to the higher survivability of the Peripheral Blood Mononuclear Cells (PBMCs) and Phytohemagglutinin (PHA-P) mitogen based stimulation indices. The study also revealed the significant differences (P<0.05) in the level of nitric oxide (µM) production amongst the pre and post thermal stressed samples of Sahiwal and Frieswal crossbred samples. Further, the expression of HSP70 was significantly (P<0.05) higher in Sahiwal compared to Frieswal immediately after heat/cold shock to 6h of recovery as indirect ELISA analysis showed gradual rise in the Hsp70 protein concentration (ng/ml) immediately after heat and cold stress (0h) and reached the peak at 6h of recovery. Western blot and immune fluorescent assay results were also corroborated with the findings of indirect ELISA. In Sahiwal cattle the mRNA expression of HSP70 and its protein concentration were higher (P<0.05) during peak summer (44°C) and winter (10°C) as compared to Frieswal cattle. This investigation supports the earlier information on the higher adaptability of indigenous cattle breeds to hot and humid conditions compared to the crossbreds of temperate cattle breeds.Not Availabl

The variation in the immune status of the animals may alter the level of tolerance of animals exposed to heat or cold stress. The present study was conducted to investigate the expression profile of two important nucleotide binding and oligomerization domain receptors (NLRs) (NOD1 and NOD2) and their central signalling molecule RIP2 gene during in vitro thermal-stressed bovine peripheral blood mononuclear cells (PBMCs) of native (Sahiwal) and crossbred (Sahiwal X HF) cattle. We also examined the differential expression profile of certain acute inflammatory cytokines in in vitro thermal-stressed PBMC culture among native and its crossbred counterparts. Results revealed that the expression profile of NOD1/2 positively correlates with the thermal stress, signalling molecule and cytokines. Present findings also highlighted that the expression patterns during thermal stress were comparatively superior among indigenous compared to crossbred cattle which may add references regarding the better immune adaptability of Zebu cattleNot Availabl
