    The Carboxyl-Terminal Segment of Apolipoprotein A-V Undergoes a Lipid-Induced Conformational Change

    Apolipoprotein (apo) A-V is a 343-residue, multidomain protein that plays an important role in regulation of plasma triglyceride homeostasis. Primary sequence analysis revealed a unique tetraproline sequence (Pro293-Pro296) near the carboxyl terminus of the protein. A peptide corresponding to the 48-residue segment beyond the tetraproline motif was generated from a recombinant apoA-V precursor wherein Pro295 was replaced by Met. Cyanogen bromide cleavage of the precursor protein, followed by negative affinity chromatography, yielded a purified peptide. Nondenaturing polyacrylamide gel electrophoresis verified that apoA-V(296-343) solubilizes phospholipid vesicles, forming a relatively heterogeneous population of reconstituted high-density lipoprotein with Stokes’ diameters\u3e17 nm. At the same time, apoA-V(296-343) failed to bind a spherical lipoprotein substrate in vitro. Far-UV circular dichroism spectroscopy revealed the peptide is unstructured in buffer yet adopts significant R-helical secondary structure in the presence of the lipid mimetic solvent trifluoroethanol (TFE; 50% v/v). Heteronuclear multidemensional NMR spectroscopy experiments were conducted with uniformly 15N- and 15N/13C-labeled peptide in 50% TFE. Peptide backbone assignment and secondary structure prediction using TALOSþ reveal the peptide adopts R-helix secondary structure from residues 309 to 334. In TFE, apoA-V(296-343) adopts an extended amphipathic R-helix, consistent with a role in lipoprotein binding as a component of full-length apoA-V

    Very long baseline interferometry presents an opportunity to measure at the centimeter level such geodetic parameters as baseline length and instantaneous pole position. In order to achieve such precision, the geophysical and astronomical models used in data analysis must be as accurate as possible. The Mark-3 interactive data analysis system includes a number of refinements beyond conventional practice in modeling precession, nutation, diurnal polar motion, UT1, solid Earth tides, relativistic light deflection, and reduction to solar system barycentric coordinates. The algorithms and their effects on the recovered geodetic, geophysical, and astrometric parameters are discussed

    The Mark 3 data base handler

    A data base handler which would act to tie Mark 3 system programs together is discussed. The data base handler is written in FORTRAN and is implemented on the Hewlett-Packard 21MX and the IBM 360/91. The system design objectives were to (1) provide for an easily specified method of data interchange among programs, (2) provide for a high level of data integrity, (3) accommodate changing requirments, (4) promote program accountability, (5) provide a single source of program constants, and (6) provide a central point for data archiving. The system consists of two distinct parts: a set of files existing on disk packs and tapes; and a set of utility subroutines which allow users to access the information in these files. Users never directly read or write the files and need not know the details of how the data are formatted in the files. To the users, the storage medium is format free. A user does need to know something about the sequencing of his data in the files but nothing about data in which he has no interest

    Telecommunications and data acquisition support for the Pioneer Venus Project: Pioneers 12 and 13, prelaunch through March 1984

    The support provided by the Telecommunications and Data Acquisition organization of the Jet Propulsion Laboratory (JPL) to the Pioneer Venus missions is described. The missions were the responsibility of the Ames Research Center (ARC). The Pioneer 13 mission and its spacecraft design presented one of the greatest challenges to the Deep Space Network (DSN) in the implementation and operation of new capabilities. The four probes that were to enter the atmosphere of Venus were turned on shortly before arrival at Venus, and the DSN had to acquire each of these probes in order to recover the telemetry being transmitted. Furthermore, a science experiment involving these probes descending through the atmosphere required a completed new data type to be generated at the ground stations. This new data type is known as the differential very long baseline interferometry. Discussions between ARC and JPL of the implementation requirements involved trade-offs in spacecraft design and led to a very successful return of science data. Specific implementation and operational techniques are discussed, not only for the prime mission, but also for the extended support to the Pioneer 12 spacecraft (in orbit around Venus) with its science instruments including that for radar observations of the planet
