Kinetic and Structural Analysis of the Mg2+ -binding Site of the Guanine Nucleotide-binding Protein p21 H-ras.
The coordination and binding of the Mg2+ ion in the nucleotide−binding site of p21 have been investigated using site−directed mutagenesis, kinetic methods, and phosphorous NMR. Mg2+ in the p21.nucleotide.Mg2+ complex appears to be in fast equilibrium with the solvent. The dissociation constant between Mg2+ and the p21.GDP complex was determined to be 2.8 microM. It decreases 30− or 16−fold on substituting Ser−17 or Asp−57 with alanine, respectively, whereas the T35A mutation has no effect. All three mutations influence the dissociation constants and the association and dissociation rate constants of the interaction between guanine nucleotides and p21, but to a different degree. We conclude that Thr−35 is only complexed to Mg2+ in the GTP conformation and both Asp−57 and Ser−17 appear to be critical for both GDP and GTP binding. 31P NMR spectra of the GDP and Gpp(NH)p (guanosine−5'−(beta,gamma−imido)triphosphate) complexes of mutated p21 show a remarkable perturbation of the guanine nucleotide− binding site compared to wild−type protein. The mutant proteins show reduced GTPase rates, which are not stimulated by the GTPase−activating protein GAP. p21(S17A) has been reported to function just as p21(S17N) as a dominant negative inhibitor of normal p21. Acute Polyhydramnios Complicating Twin Pregnancies
Acute polyhydramnios in the second trimestr is a typical complication in monozygous twin pregnancies. It is caused by a feto-fetal transfusion with anemia on the donor and polycytemia on the recipient twin. Contrary to the chronic hydramnios, there is no increase in malformations. In view of the high mortality rate (100%, according to most authors), the clinical management has to be reconsidered. During the years 1979 to 1983, 10 cases of acute polyhydramnios have been observed at the University Hospital in Zurich. This corresponds to an incidence of 9% in our twin population. All cases investigated were MZ twin pregnancies. With the exception of one patient, who underwent an abortion, all women were hospitalized, had bed rest and received recurrent removals of amniotic fluid and prophylactic tocolysis. The mean gestational age at the time of diagnosis was 23 4/7 weeks and at delivery 30 3/7 weeks. In two cases - one of which is presented in detail - with an unintentional puncture of a placental vessel, the recurrence of the hydramnios did not appear. Eight of 18 newborns survived. No malformations were found. Bed rest, tocolysis and recurrent amniocenteses seem to have a positive influence on the prolongation and outcome of the gestation in acute polyhydramnio

Nonlinear Dynamics of Capacitive Charging and Desalination by Porous Electrodes
The rapid and efficient exchange of ions between porous electrodes and
aqueous solutions is important in many applications, such as electrical energy
storage by super-capacitors, water desalination and purification by capacitive
deionization (or desalination), and capacitive extraction of renewable energy
from a salinity difference. Here, we present a unified mean-field theory for
capacitive charging and desalination by ideally polarizable porous electrodes
(without Faradaic reactions or specific adsorption of ions) in the limit of
thin double layers (compared to typical pore dimensions). We illustrate the
theory in the case of a dilute, symmetric, binary electrolyte using the
Gouy-Chapman-Stern (GCS) model of the double layer, for which simple formulae
are available for salt adsorption and capacitive charging of the diffuse part
of the double layer. We solve the full GCS mean-field theory numerically for
realistic parameters in capacitive deionization, and we derive reduced models
for two limiting regimes with different time scales: (i) In the
"super-capacitor regime" of small voltages and/or early times where the porous
electrode acts like a transmission line, governed by a linear diffusion
equation for the electrostatic potential, scaled to the RC time of a single
pore. (ii) In the "desalination regime" of large voltages and long times, the
porous electrode slowly adsorbs neutral salt, governed by coupled, nonlinear
diffusion equations for the pore-averaged potential and salt concentration
Negative phase time for Scattering at Quantum Wells: A Microwave Analogy Experiment
If a quantum mechanical particle is scattered by a potential well, the wave
function of the particle can propagate with negative phase time. Due to the
analogy of the Schr\"odinger and the Helmholtz equation this phenomenon is
expected to be observable for electromagnetic wave propagation. Experimental
data of electromagnetic wells realized by wave guides filled with different

SWI/SNF-like chromatin remodeling factor Fun30 supports point centromere function in S. cerevisiae
Budding yeast centromeres are sequence-defined point centromeres and are, unlike in many other organisms, not
embedded in heterochromatin. Here we show that Fun30, a poorly understood SWI/SNF-like chromatin remodeling factor
conserved in humans, promotes point centromere function through the formation of correct chromatin architecture at
centromeres. Our determination of the genome-wide binding and nucleosome positioning properties of Fun30 shows that
this enzyme is consistently enriched over centromeres and that a majority of CENs show Fun30-dependent changes in
flanking nucleosome position and/or CEN core micrococcal nuclease accessibility. Fun30 deletion leads to defects in histone
variant Htz1 occupancy genome-wide, including at and around most centromeres. FUN30 genetically interacts with CSE4,
coding for the centromere-specific variant of histone H3, and counteracts the detrimental effect of transcription through centromeres on chromosome segregation and suppresses transcriptional noise over centromere CEN3. Previous work has shown a requirement for fission yeast and mammalian homologs of Fun30 in heterochromatin assembly. As centromeres in budding yeast are not embedded in heterochromatin, our findings indicate a direct role of Fun30 in centromere chromatin by promoting correct chromatin architecture
Nonlocal reflection by photonic barriers
The time behaviour of microwaves undergoing partial reflection by photonic
barriers was measured in the time and in the frequency domain. It was observed
that unlike the duration of partial reflection by dielectric layers, the
measured reflection duration of barriers is independent of their length. The
experimental results point to a nonlocal behaviour of evanescent modes at least
over a distance of some ten wavelengths. Evanescent modes correspond to
photonic tunnelling in quantum mechanics.Comment: 8 pages, 5 figure
Negative time delay for wave reflection from a one-dimensional semi-harmonic well
It is reported that the phase time of particles which are reflected by a
one-dimensional semi-harmonic well includes a time delay term which is negative
for definite intervals of the incoming energy. In this interval, the absolute
value of the negative time delay becomes larger as the incident energy becomes
smaller. The model is a rectangular well with zero potential energy at its

Pre-Emptive Endoluminal Negative Pressure Therapy at the Anastomotic Site in Minimally Invasive Transthoracic Esophagectomy (the preSPONGE Trial): Study Protocol for a Multicenter Randomized Controlled Trial
Introduction: Anastomotic leakage (AL) accounts for a significant proportion of morbidity following oesophagectomy. Endoluminal negative pressure (ENP) therapy via a specifically designed polyurethane foam (EsoSponge®, B.Braun Medical, Melsungen, Germany) has become the standard of care for AL in many specialized centres. The prophylactic (pENP) application of this technique aims to reduce postoperative morbidity and is a novel approach which has not yet been investigated in a prospective study. The aim of this study is therefore to assess the effect of pENP at the anastomotic site in high-risk patients undergoing minimally invasive transthoracic Ivor Lewis oesophagectomy.
Methods and analysis: The study design is a prospective, multi-centre, two-arm, parallel-group, randomised controlled trial and will be conducted in two phases. Phase one is a randomised feasibility and safety pilot trial involving 40 consecutive patients. After definitive sample size calculation, additional patients will be included accordingly during phase two. The primary outcome of the study will be the postoperative length of hospitalization until reaching previously defined “fit for discharge criteria”. Secondary outcomes will include postoperative morbidity, mortality and postoperative AL-rates based on 90-day follow-up. A confirmatory analysis based on intention-to-treat will be performed.
Ethics and dissemination: The ethics committee of the University of Zurich approved this study (2019-00562), which has been registered with ClinicalTrials.gov on 14.11.2019 (NCT04162860) and the Swiss National Clinical Trials Portal (SNCTP000003524).
