Multi-confocal fluorescence correlation spectroscopy in living cells using a complementary metal oxide semiconductor-single photon avalanche diode array
International audienceLiving cells are heterogeneous and rapidly changing biological samples. It is thus desirable to measure molecular concentration and dynamics in many locations at the same time. In this note, we present a multi-confocal setup capable of performing simultaneous fluorescence correlation spectroscopy measurements, by focusing the spots with a spatial light modulator and acquiring data with a monolithic 32 Ă— 32 single-photon avalanche photodiode array. A post-processing method is proposed to correct cross-talk effects between neighboring spots. We demonstrate the applicability of our system by simultaneously measuring the diffusion of free enhanced Green Fluorescent Protein (eGFP) molecules at nine different points in living cells
Comparing modelled fire dynamics with charcoal records for the Holocene
An earth system model of intermediate complexity (CLIMate and BiosphERe – CLIMBER-2) and a land surface model (JSBACH), which dynamically represent vegetation, are used to simulate natural fire dynamics through the last 8000 yr. Output variables of the fire model (burned area and fire carbon emissions) are used to compare model results with sediment-based charcoal reconstructions. Several approaches for processing model output are also tested. Charcoal data are reported in Z-scores with a base period of 8000–200 BP in order to exclude the strong anthropogenic forcing of fire during the last two centuries. The model–data comparison reveals a robust correspondence in fire activity for most regions considered, while for a few regions, such as Europe, simulated and observed fire histories show different trends. The difference between modelled and observed fire activity may be due to the absence of anthropogenic forcing (e.g. human ignitions and suppression) in the model simulations, and also due to limitations inherent to modelling fire dynamics. The use of spatial averaging (or Z-score processing) of model output did not change the directions of the trends. However, Z-score-transformed model output resulted in higher rank correlations with the charcoal Z-scores in most regions. Therefore, while both metrics are useful, processing model output as Z-scores is preferable to areal averaging when comparing model results to transformed charcoal records
Black hole entropy for the general area spectrum
We consider the possibility that the horizon area is expressed by the general
area spectrum in loop quantum gravity and calculate the black hole entropy by
counting the degrees of freedom in spin-network states related to its area.
Although the general area spectrum has a complex expression, we succeeded in
obtaining the result that the black hole entropy is proportional to its area as
in previous works where the simplified area formula has been used. This gives
new values for the Barbero-Immirzi parameter () which are larger than that of previous works.Comment: 5 page
Design of super-paramagnetic bilayer films based on chitosan and sodium alginate
Bilayer films that combine chitosan and alginate, two natural polysaccharides, and magnetic iron oxide nanoparticles (MNPs) were obtained. Physical and microscopic observations revealed that chitosan and alginate interact strongly through their surfaces, which is attributed to the formation of a polyelectrolyte complex at the interface. The diameter of the individual MNPs was about 10 nm, although in the films they appear arranged in clusters with sizes ranging from 23 nm to several times larger that are formed by collapsed individual particles. All bilayers containing MNP behave as super-paramagnetic materials, exhibiting magnetic synergic effects in comparison to single carbohydrate films. Thus, obtained films could find novel and interesting applications as pH responsiveness systems, pad dressings including hyperthermia, adsorption of positive and negative charged pollutants, etc.Fil: Kloster, Gianina Andrea. Dynamic critical behavior of failure and plastic deformation in the random fiber bundle model
The random fiber bundle (RFB) model, with the strength of the fibers
distributed uniformly within a finite interval, is studied under the assumption
of global load sharing among all unbroken fibers of the bundle. At any fixed
value of the applied stress (load per fiber initially present in the bundle),
the fraction of fibers that remain unbroken at successive time steps is shown
to follow simple recurrence relations. The model is found to have stable fixed
point for applied stress in the range 0 and 1; beyond which total failure of
the bundle takes place discontinuously. The dynamic critical behavior near this
failure point has been studied for this model analysing the recurrence
relations. We also investigated the finite size scaling behavior. At the
critical point one finds strict power law decay (with time t) of the fraction
of unbroken fibers. The avalanche size distribution for this mean-field
dynamics of failure has been studied. The elastic response of the RFB model has
also been studied analytically for a specific probability distribution of fiber
strengths, where the bundle shows plastic behavior before complete failure,

Large Scale Structures, Symmetry, and Universality in Sandpiles
We introduce a sandpile model where, at each unstable site, all grains are
transferred randomly to downstream neighbors. The model is local and
conservative, but not Abelian. This does not appear to change the universality
class for the avalanches in the self-organized critical state. It does,
however, introduce long-range spatial correlations within the metastable
states. We find large scale networks of occupied sites whose density vanishes

Generic isolated horizons in loop quantum gravity
Isolated horizons model equilibrium states of classical black holes. A
detailed quantization, starting from a classical phase space restricted to
spherically symmetric horizons, exists in the literature and has since been
extended to axisymmetry. This paper extends the quantum theory to horizons of
arbitrary shape. Surprisingly, the Hilbert space obtained by quantizing the
full phase space of \textit{all} generic horizons with a fixed area is
identical to that originally found in spherical symmetry. The entropy of a
large horizon remains one quarter its area, with the Barbero-Immirzi parameter
retaining its value from symmetric analyses. These results suggest a
reinterpretation of the intrinsic quantum geometry of the horizon surface.Comment: 13 page
Logarithmic corrections to black hole and black ring entropy in tunneling approach
The tunneling approach beyond semiclassical approximation has been used to
calculate the corrected Hawking temperature and entropy for various black holes
and FRW universe model. We examine their derivations, and prove that the
quantity in the corrected temperature is the explicit function of the only
free parameter (which is an auxiliary parameter defined by
). Our analysis improves previous calculations, and
indicates that the leading order logarithmic correction to entropy is a natural
result of the corrected temperature and the first law of thermodynamics.
Additionally, we apply the tunneling approach beyond semiclassical
approximation to neutral black rings. Based on the analysis, we show that the
entropy of neutral black rings also has a logarithmic leading order correction.Comment: 13 pages, rewritte
Directed avalanche processes with underlying interface dynamics
We describe a directed avalanche model; a slowly unloading sandbox driven by
lowering a retaining wall. The directness of the dynamics allows us to
interpret the stable sand surfaces as world sheets of fluctuating interfaces in
one lower dimension. In our specific case, the interface growth dynamics
belongs to the Kardar-Parisi-Zhang (KPZ) universality class. We formulate
relations between the critical exponents of the various avalanche distributions
and those of the roughness of the growing interface. The nonlinear nature of
the underlying KPZ dynamics provides a nontrivial test of such generic exponent
relations. The numerical values of the avalanche exponents are close to the
conventional KPZ values, but differ sufficiently to warrant a detailed study of
whether avalanche correlated Monte Carlo sampling changes the scaling exponents
of KPZ interfaces. We demonstrate that the exponents remain unchanged, but that
the traces left on the surface by previous avalanches give rise to unusually
strong finite-size corrections to scaling. This type of slow convergence seems
