    Academic and Business Leaders Agree: Six Skills Essential for Effective Management

    This study surveyed 632 business and public education mid-level managers to ascertain both the importance of and their confidence in using six research-based management competencies. Both groups rated the six as equally or highly valuable; however, educational managers valued conflict resolution more than business counterparts. The education group reported less confidence using their skills in problem solving, conflict resolution, and strategic planning. While both groups perceived most of the six competencies as equally valuable, the two groups did not feel similarly confident using the skills. Participants’ ratings of skill-importance did not predict their level of confidence using the skills

    Hyperplasia of pancreatic islets associated with extrapancreatic lymphoma and sarcoma

    A preliminary study revealed pancreatic hyperplasia to be present in 29 per cent of 100 consecutive necropsies. Malignant neoplasms coexisted with hyperplasia of islets in 14 (48.3 per cent) of the 29 cases. On the basis of the results of this study, the islets of Langerhans in microscopic sections of pancreas from 20 cases of accidental death (normal controls), 10 cases of extrapancreatic lymphoma, and 12 cases of extrapancreatic sarcoma were quantitatively studied. Significant hyperplasia of the islets was present in both categories of neoplastic disease when compared with the islets in the control group. The assessment of hyperplasia of pancreatic islets was neither difficult nor complex when the observer assigned the proper significance to the presence of an increased percentage of large islets.     Combustion Stability Verification for the Thrust Chamber Assembly of J-2X Developmental Engines 10001, 10002, and 10003

    The J-2X engine, a liquid oxygen/liquid hydrogen propellant rocket engine available for future use on the upper stage of the Space Launch System vehicle, has completed testing of three developmental engines at NASA Stennis Space Center. Twenty-one tests of engine E10001 were conducted from June 2011 through September 2012, thirteen tests of the engine E10002 were conducted from February 2013 through September 2013, and twelve tests of engine E10003 were conducted from November 2013 to April 2014. Verification of combustion stability of the thrust chamber assembly was conducted by perturbing each of the three developmental engines. The primary mechanism for combustion stability verification was examining the response caused by an artificial perturbation (bomb) in the main combustion chamber, i.e., dynamic combustion stability rating. No dynamic instabilities were observed in the TCA, although a few conditions were not bombed. Additional requirements, included to guard against spontaneous instability or rough combustion, were also investigated. Under certain conditions, discrete responses were observed in the dynamic pressure data. The discrete responses were of low amplitude and posed minimal risk to safe engine operability. Rough combustion analyses showed that all three engines met requirements for broad-banded frequency oscillations. Start and shutdown transient chug oscillations were also examined to assess the overall stability characteristics, with no major issues observed

    Surgery and postoperative radiotherapy a valid treatment for advanced oropharyngeal carcinoma

    Since 1992 we have prospectively included all head and neck cancer patients in our health region in a departmental based register. Our hospital takes care of all head and neck cancer patients in our health region consisting of approximately 1 million people. In 1997, we evaluated the results of the treatment of oropharyngeal cancer in the 1992–1997 period. On the basis of this evaluation, we changed our treatment policy for tonsillar and base of tongue carcinoma. We first changed the treatment for the lesions with worst prognosis, i.e., those with T3–T4 carcinomas, from radiotherapy only, to radical surgery and postoperative radiotherapy. We have since that time increasingly also operated the smaller oropharyngeal carcinomas. The 2 years’ overall survival and disease-specific survival for all patients diagnosed in the 1992–1997 period was 56 and 63%, respectively. The results from a similar group of patients in the 6 years’ period from 2000 to 2005, after the change in treatment, have increased to 83 and 88%. When we looked at the subgroup of patients in the 2000–2005 period treated with surgery and postoperative radiotherapy, 45 out of 69 patients (65%) presenting with an oropharyngeal cancer were fit for operation. With radical surgery and postoperative radiation therapy, the 2 years overall survival is now 91%. The 2-year disease-specific survival is 96% and the locoregional control is 98%. This is a marked improvement as compared to radiotherapy alone and definitely competitive with modern radiochemotherapy

    Contemporary management of primary parapharyngeal space tumors

    The parapharyngeal space is a complex anatomical area. Primary parapharyngeal tumors are rare tumors and 80% of them are benign. A variety of tumor types can develop in this location; most common are salivary gland neoplasm and neurogenic tumors. The management of these tumors has improved greatly owing to the developments in imaging techniques, surgery, and radiotherapy. Most tumors can be removed with a low rate of complications and recurrence. The transcervical approach is the most frequently used. In some cases, minimally invasive approaches may be used alone or in combination with a limited transcervical route, allowing large tumors to be removed by reducing morbidity of expanded approaches. An adequate knowledge of the anatomy and a careful surgical plan is essential to tailor management according to the patient and the tumor. The purpose of the present review was to update current aspects of knowledge related to this more challenging area of tumor occurrence.Peer reviewe

    Recognition of DNA by the Helix-Turn-Helix Global Regulatory Protein Lrp Is Modulated by the Amino Terminus ▿ †

    The AsnC/Lrp family of regulatory proteins links bacterial and archaeal transcription patterns to metabolism. In Escherichia coli, Lrp regulates approximately 400 genes, over 200 of them directly. In earlier studies, lrp genes from Vibrio cholerae, Proteus mirabilis, and E. coli were introduced into the same E. coli background and yielded overlapping but significantly different regulons. These differences were seen despite amino acid sequence identities of 92% (Vibrio) and 98% (Proteus) to E. coli Lrp, including complete conservation of the helix-turn-helix motifs. The N-terminal region contains many of the sequence differences among these Lrp orthologs, which led us to investigate its role in Lrp function. Through the generation of hybrid proteins, we found that the N-terminal diversity is responsible for some of the differences between orthologs in terms of DNA binding (as revealed by mobility shift assays) and multimerization (as revealed by gel filtration, dynamic light scattering, and analytical ultracentrifugation). These observations indicate that the N-terminal tail plays a significant role in modulating Lrp function, similar to what is seen for a number of other regulatory proteins

    Molecular recognition requires dimerization of a VHH antibody

    ABSTRACTCamelid heavy-chain-only antibodies are a unique class of antibody that possesses only a single variable domain (termed VHH) for antigen recognition. Despite their apparent canonical mechanism of target recognition, where a single VHH domain binds a single target, an anti-caffeine VHH has been observed to possess 2:1 stoichiometry. Here, the structure of the anti-caffeine VHH/caffeine complex enabled the generation and biophysical analysis of variants that were used to better understand the role of VHH homodimerization in caffeine recognition. VHH interface mutants and caffeine analogs, which were examined to probe the mechanism of caffeine binding, suggested caffeine recognition is only possible with the VHH dimer species. Correspondingly, in the absence of caffeine, the anti-caffeine VHH was found to form a dimer with a dimerization constant comparable to that observed with VH:VL domains in conventional antibody systems, which was most stable near physiological temperature. While the VHH:VHH dimer structure (at 1.13 Å resolution) is reminiscent of conventional VH:VL heterodimers, the homodimeric VHH possesses a smaller angle of domain interaction, as well as a larger amount of apolar surface area burial. To test the general hypothesis that the short complementarity-determining region-3 (CDR3) may help drive VHH:VHH homodimerization, an anti-picloram VHH domain containing a short CDR3 was generated and characterized, which revealed it also existed as dimer species in solution. These results suggest homodimer-driven recognition may represent a more common method of VHH ligand recognition, opening opportunities for novel VHH homodimer affinity reagents and helping to guide their use in chemically induced dimerization applications
