    \u3cem\u3eRhizobium japonicum\u3c/em\u3e Mutants Defective in Symbiotic Nitrogen Fixation

    Rhizobium japonicum strains 3I1b110 and 61A76 were mutagenized to obtain 25 independently derived mutants that produced soybean nodules defective in nitrogen fixation, as assayed by acetylene reduction. The proteins of both the bacterial and the plant portions of the nodules were analyzed by two-dimensional polyacrylamide gel electrophoresis. All of the mutants had lower-than-normal levels of the nitrogenase components, and all but four contained a prominent bacteroid protein not observed in wild-type bacteroids. Experiments with bacteria grown ex planta suggested that this protein was derepressed by the absence of ammonia. Nitrogenase component II of one mutant was altered in isoelectric point. The soluble plant fraction of the nodules of seven mutants had very low levels of heme, yet the nodules of five of these seven mutants contained the polypeptide of leghemoglobin. Thus, the synthesis of the globin may not be coupled to the content of available heme in soybean nodules. The nodules of the other two of these seven mutants lacked not only leghemoglobin but most of the other normal plant and bacteroid proteins. Ultrastructural examination of nodules formed by these two mutants indicated normal ramification of infection threads but suggested a problem in subsequent survival of the bacteria and their release from the infection threads

    Child Support Payment and Child Visitation: Perspectives from Nonresident Fathers and Resident Mothers

    The purpose of this qualitative study was to examine the child support and visitation perspectives of nonresidential fathers and custodial mothers. The focus of the study was to present definitions of child support from both noncustodial fathers and custodial mothers, the barriers they experience that prevent child support and visitation, and suggestions the parents have for improvements in the child support system. The data suggest that although nonresidential fathers and custodial mothers have similar definitions of what characteristicsd efine child support, they have vastly different views of what barriers prevent child support and visitation. Interparental hostility appeared to shape their perspectives about child support and visitation. Recommendations targeting the negative perceptions parents have of one another are presented

    Physicochemical Properties and Catalytic Behavior of the Molecular Sieve SSZ-70

    SSZ-70 is synthesized using 1,3-bis(isobutyl)imidazolium, 1,3-bis(cyclohexyl)imidazolium, and 1,3-bis(cycloheptyl)imidazolium structure directing agents (SDAs), and the solids obtained are characterized by powder X-ray diffraction (XRD), ^(29)Si magic angle spinning nuclear magnetic resonance (MAS NMR), electron microscopy, nitrogen and hydrocarbon adsorption, and thermogravimetric analyses. The physicochemical properties of SSZ-70 show that it is a new molecular sieve that has similarities to MWW-type materials. The catalytic behavior of SSZ-70 is evaluated through the use of the constraint index (CI) test. Distinct differences in the reactivity between Al-SSZ-70 and SSZ-25 (MWW) are observed and are the consequences of the structural differences between these two molecular sieves

    Time-Dependent Regulation of Apoptosis by Aen and Bax in Response to 2-Aminoanthracene Dietary Consumption

    Background/Objective: The modulation of the toxic effects of 2-aminoanthracene (2AA) on the liver by apoptosis was investigated. Fisher-344 (F344) rats were exposed to various concentrations of 2AA for 14 and 28 days. The arylamine 2AA is an aromatic hydrocarbon employed in manufacturing chemicals, dyes, inks, and it is also a curing agent in epoxy resins and polyurethanes. 2AA has been detected in tobacco smoke and cooked foods. Methods: Analysis of total messenger ribonucleic acid (mRNA) extracts from liver for apoptosis-related gene expression changes in apoptosis enhancing nuclease (AEN), Bcl2-associated X protein (BAX), CASP3, Jun proto-oncogene (JUN), murine double minute-2 p53 binding protein homolog (MDM2), tumor protein p53 (p53), and GAPDH genes by quantitative real-time polymerase chain reaction (qRT-PCR) was coupled with terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) and caspase-3 (Casp3) activity assays. Results: Specific apoptosis staining result does not seem to show significant difference between control and treated animals. This may be due to freeze-thaw artifacts observed in the liver samples. However, there appears to be a greater level of apoptosis in medium- and high-dose (MD and HD) 2AA treated animals. Analyses of apoptosis-related genes seem to show AEN and BAX as the main targets in the induction of apoptosis in response to 2AA exposure, though p53, MDM2, and JUN may play supporting roles. Conclusion: Dose-dependent increases in mRNA expression were observed in all genes except Casp3. BAX was very highly expressed in the HD rats belonging to the 2-week exposure group. This trend was not observed in the animals treated for 4 weeks. Instead, AEN was rather very highly expressed in the liver of the MD animals that were treated with 2AA for 28 days

    Exchange Bias Induced by the Fe3O4 Verwey transition

    We present a study of the exchange bias in different configurations of V2O3 thin films with ferromagnetic layers. The exchange bias is accompanied by a large vertical shift in the magnetization. These effects are only observed when V2O3 is grown on top of Ni80Fe20 permalloy. The magnitude of the vertical shift is as large as 60% of the total magnetization which has never been reported in any system. X-Ray diffraction studies show that the growth conditions promote the formation of a ferrimagnetic Fe3O4 interlayer. The change in the easy magnetization axis of Fe3O4 across the Verwey transition at 120 K is correlated with the appearance of exchange bias and vertical shift in magnetization. Both phenomena disappear above 120 K, indicating for the first time a direct relationship between the magnetic signature of the Verwey transition and exchange bias.Comment: Accepted for publication Physical Review

    Presence and Localization of Pro-and Mature Forms of Biglycan and Decorin in Human Costal Cartilage Derived from Chest Wall Deformities

    Costal cartilage is a type of hyaline cartilage that forms rod-like structures that connect the ribs to the sternum. The most common chest wall deformities, pectus excavatum and pectus carinatum involved efective costal cartilage resulting in sternal displacement. Costal cartilage is not widely studied leaving little insight into possible factors involved in the pathogenesis of these pectus deformities. This study focused on the presence and distribution of two important regulators of collagen fibrillogenesis and organization, biglycan and decorin. Immunohistochemical analysis of transverse cross sections of normal and deformed costal cartilage revealed that biglycan and decorin mainly localized in the territorial matrix except for prodecorin which was only found within chondrocytes. Western blot analysis of whole protein extracts demonstrated the presence of both pro and mature forms of biglycan and mature decorin in patients and controls. In normal costal cartilage of different ages, the mature form of decorin was absent in a fetal sample whereas mature biglycan was weakly expressed, suggestive that mature biglycan may play a role in early costal cartilage development. Further studies are needed to determine the functional differences between the pro- and mature forms of biglycan and decorin both in age and disease

    Spin properties of dense near-surface ensembles of nitrogen-vacancy centres in diamond

    We present a study of the spin properties of dense layers of near-surface nitrogen-vacancy (NV) centres in diamond created by nitrogen ion implantation. The optically detected magnetic resonance contrast and linewidth, spin coherence time, and spin relaxation time, are measured as a function of implantation energy, dose, annealing temperature and surface treatment. To track the presence of damage and surface-related spin defects, we perform in situ electron spin resonance spectroscopy through both double electron-electron resonance and cross-relaxation spectroscopy on the NV centres. We find that, for the energy (4−304-30~keV) and dose (5×1011−10135\times10^{11}-10^{13}~ions/cm2^2) ranges considered, the NV spin properties are mainly governed by the dose via residual implantation-induced paramagnetic defects, but that the resulting magnetic sensitivity is essentially independent of both dose and energy. We then show that the magnetic sensitivity is significantly improved by high-temperature annealing at ≥1100∘\geq1100^\circC. Moreover, the spin properties are not significantly affected by oxygen annealing, apart from the spin relaxation time, which is dramatically decreased. Finally, the average NV depth is determined by nuclear magnetic resonance measurements, giving ≈10\approx10-17~nm at 4-6 keV implantation energy. This study sheds light on the optimal conditions to create dense layers of near-surface NV centres for high-sensitivity sensing and imaging applications.Comment: 12 pages, 7 figure

    A k-shell decomposition method for weighted networks

    We present a generalized method for calculating the k-shell structure of weighted networks. The method takes into account both the weight and the degree of a network, in such a way that in the absence of weights we resume the shell structure obtained by the classic k-shell decomposition. In the presence of weights, we show that the method is able to partition the network in a more refined way, without the need of any arbitrary threshold on the weight values. Furthermore, by simulating spreading processes using the susceptible-infectious-recovered model in four different weighted real-world networks, we show that the weighted k-shell decomposition method ranks the nodes more accurately, by placing nodes with higher spreading potential into shells closer to the core. In addition, we demonstrate our new method on a real economic network and show that the core calculated using the weighted k-shell method is more meaningful from an economic perspective when compared with the unweighted one.Comment: 17 pages, 6 figure
