    Detection and distribution of Craspedacusta sowerbii: Observations of medusae are not enough

    The freshwater cnidarian Craspedacusta sowerbii, native to the Yangtze valley, has invaded lakes and ponds throughout the world. Most distribution records have to date been based on observations of the medusa (jellyfish) stage, including numerous recent publications. We aimed to determine whether polyps are widespread in lakes, and geographical areas, outside of where medusae have been observed, and whether constructed waters are more easily invaded than natural waters. Our results show that C. sowerbii is more common and widespread than is apparent from observations of medusae. We argue that observed occurrences of medusae provide little useful information regarding the distribution of this species, and that published records of new jellyfish occurrences provide unreliable estimates of the timing of introduction, establishment or spread of C. sowerbii in new regions. We found no evidence that constructed waters were more readily invaded than natural waters. Overall, accurate determination of Craspedacusta occurrence and distribution requires systematic surveys of the polyp stages

    How Do Students Use Self-Regulation in the Learning of Science in an Alternative School Setting?

    In my classroom, I am able to work with each student on an individual basis. The students are able to work at their own pace without any other student knowing what lessons they are on. The students have a prescribed way to go through each lesson. They also have a specific way to take notes for the class. They have to put the title of the lesson, then the objectives (essential questions), vocabulary and then notes from the lessons. At the end of each lesson they have about ten questions that they have to answer. They get a grade for each lesson. Which is great so they can see how they are understanding the material. One of the best parts of the class is when they do not understand something in the material, I can go over and help them with the material. They get their own personal lesson. This is a great bonus for the students, since in a regular classroom, a teacher just cannot stop teaching the class to make sure a student comprehends the information. The student would have to come back after school and talk with the teacher on an individual basis

    Environmental governance for whom? Examining the political, institutional, fiscal, and legal determinants of state environmental agency budget policy in the US

    Budgets are a prospective tool of governance, and appropriations are a planning vehicle reflecting: bureaucracies’ values, complex interactions, collective preferences, political influences, and available resources. Research spanning 30 years finds that environmental pollution is a key determinant of environmental budgets in the US, though myriad factors, actors, and subsystems are important to consider. Due to federalism and devolution of responsibilities and authorities, environmental governance falls largely to the states. While the dynamics that shape state environmental budget policy have received scholarly interest, theoretically-driven examinations of environmental appropriations remain limited within the public budgeting and environmental policy literature. Using panel data from 2010 to 2015, this dissertation examines the legal, political, institutional, and fiscal factors that influence state own-source environmental funding drawing from several theories. Given the key relationship between environmental pollution and environmental budgets—as stressed by previous scholarship—findings reveal the effects are conditioned on business interests from polluting sectors. While the interaction effect holds across funding sources, the negative budgetary influence depends on the type of air pollution modeled. Fiscal capacity is found to increase appropriations from state general funds but not appropriations from fees and other sources. Mandatory climate policies have a positive influence on budgets, though the evidence is inconsistent between models. Given cuts to federal environmental funding, flat trends in state funding, what factors influence the financing of environmental protection are of critical importance for civil society, practitioners, and public officials; therefore, this dissertation concludes with policy implications and avenues for future research

    Increasing prevalence of asthma diagnosis and symptoms in children is confined to mild symptoms

    BACKGROUND: The prevalence of childhood asthma is increasing but few studies have investigated trends in asthma severity. We investigated trends in asthma diagnosis and symptom morbidity between an eight year time period in a paired prevalence study. METHODS: All children in one single school year aged 8-9 years in the city of Sheffield were given a parent respondent questionnaire in 1991 and 1999 based on questions from the International Survey of Asthma and Allergy in Children (ISAAC). Data were obtained regarding the prevalence of asthma and wheeze and current (12 month) prevalences of wheeze attacks, speech limiting wheeze, nocturnal cough and wheeze, and exertional symptoms. RESULTS: The response rates in 1991 and 1999 were 4580/5321 (85.3%) and 5011/6021 (83.2%), respectively. There were significant increases between the two surveys in the prevalence of asthma ever (19.9% v 29.7%, mean difference 11.9%, 95% confidence interval (CI) 10.16 to 13.57, p<0.001), current asthma (10.3% v 13.0%, mean difference 2.7%, 95% CI 1.44 to 4.03, p<0.001), wheeze ever (30.3% v 35.8%, mean difference 5.7%, 95% CI 3.76 to 7.56, p<0.001), wheeze in the previous 12 months (17.0% v 19.4%, mean difference 2.5, 95% CI 0.95 to 4.07, p<0.01), and reporting of medication use (16.9% v 20%, mean difference 3.0%, 95% CI 1.46 to 4.62, p<0.001). There were also significant increases in reported hayfever and eczema diagnoses. CONCLUSIONS: Diagnostic labelling of asthma and lifetime prevalence of wheeze has increased. The current 12 month point prevalence of wheeze has increased but this is confined to occasional symptoms. The increased medication rate may be responsible for the static prevalence of severe asthma symptoms. The significant proportion of children receiving medication but reporting no asthma symptoms identified from our 1999 survey suggests that some children are being inappropriately treated or overtreated

    Identification of a 3-Alkylpyridinium Compound from the Red Sea Sponge Amphimedon chloros with In Vitro Inhibitory Activity against the West Nile Virus NS3 Protease.

    Viruses are underrepresented as targets in pharmacological screening efforts, given the difficulties of devising suitable cell-based and biochemical assays. In this study we found that a pre-fractionated organic extract of the Red Sea sponge Amphimedon chloros was able to inhibit the West Nile Virus NS3 protease (WNV NS3). Using liquid chromatography⁻mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR) spectroscopy, the identity of the bioactive compound was determined as a 3-alkylpyridinium with m/z = 190.16. Diffusion Ordered Spectroscopy (DOSY) NMR and NMR relaxation rate analysis suggest that the bioactive compound forms oligomers of up to 35 kDa. We observed that at 9.4 μg/mL there was up to 40⁻70% inhibitory activity on WNV NS3 protease in orthogonal biochemical assays for solid phase extracts (SPE) of A. chloros. However, the LC-MS purified fragment was effective at inhibiting the protease up to 95% at an approximate amount of 2 µg/mL with negligible cytotoxicity to HeLa cells based on a High-Content Screening (HCS) cytological profiling strategy. To date, 3-alkylpyridinium type natural products have not been reported to show antiviral activity since the first characterization of halitoxin, or 3-alkylpyridinium, in 1978. This study provides the first account of a 3-alkylpyridinium complex that exhibits a proposed antiviral activity by inhibiting the NS3 protease. We suggest that the here-described compound can be further modified to increase its stability and tested in a cell-based assay to explore its full potential as a potential novel antiviral capable of inhibiting WNV replication

    Q fever through consumption of unpasteurised milk and milk products – a risk profile and exposure assessment

    Q fever is a zoonotic disease caused by the bacterium Coxiella burnetii which is endemic in cattle, sheep and goats in much of the world, including the United Kingdom (UK). There is some epidemiological evidence that a small proportion of cases in the developed world may arise from consumption of unpasteurised milk with less evidence for milk products such as cheese. Long maturation at low pH may give some inactivation in hard cheese, and viable C. burnetii are rarely detected in unpasteurised cheese compared to unpasteurised milk. Simulations presented here predict that the probability of exposure per person to one or more C. burnetii through the daily cumulative consumption of raw milk in the UK is 04203. For those positive exposures, the average level of exposure predicted is high at 1266 guinea pig intraperitoneal infectious dose 50% units (GP_IP_ID50) per person per day. However, in the absence of human dose–response data, the case is made that the GP_IP_ID50 unit represents a very low risk through the oral route. The available evidence suggests that the risks from C. burnetii through consumption of unpasteurised milk and milk products (including cheese) are not negligible but they are lower in comparison to transmission via inhalation of aerosols from parturient products and livestock contact
