    Variability of morphological descriptors in Sicilian oat (Avena sativa L.) populations

    Due to its rusticity and feasibility of use, oat (Avena sativa L.) represents a crucial agronomic and economic resource for many semiarid environments. Presently, the recourse to new commercial varieties has caused a dramatic lowering of areas covered with the traditional local genotypes, and a severe risk of genetic erosion is emerging. To deepen the knowledge about the autochthonous oat populations, an activity of collection and cataloging across semiarid cropping areas was carried out. Sixteen oat populations were collected from different areas of Sicily and put in a field study for two consecutive years (2014 and 2015) in the experimental farm “Sparacia” (Cammarata, Italy). In both years and all populations, 21 morphological characters, related to different aspects of the whole plant or plant parts, were measured as described in the guidelines Community Plant Variety Office—Office Communautaire des Varietes Vegetales (CPVO-OCVV) (rif. CPVO-TP/020/2). Multivariate analysis (MA) was applied to assess the similarity/dissimilarity level among populations, also evaluating the relative discriminatory importance of each selected plant character. Although a strong variability between years did not allow perfect discrimination among genotypes, an association between oat groups emerged based on their prevalent utilization form. Among categorical characters, measurements on glumes and grain provided the best characterization of the populations in both years

    Milk Thistle (Silybum Marianum L.) as a Novel Multipurpose Crop for Agriculture in Marginal Environments: A Review

    Milk thistle (Silybum marianum (L.) Gaertn.) is a versatile crop that has adapted to the broadly different soil and environmental conditions throughout all continents. To date, the fruits (\u201cseeds\u201d) of the plant are the only reliable source of silymarin, which, given its recognized therapeutic effects and its many present and potential uses, has led to a significant re-discovery and enhancement of the crop in recent years. Overall, although many studies have been carried out globally on the bioactivity, phytochemistry, and genetics of milk thistle, few and discontinuous research activity has been conducted on its basic agronomy as well as on the farm opportunities offered by the cultivation of this species. However, the multiple potential uses of the plant and its reduced need for external inputs suggest that milk thistle can perfectly fit among the most interesting alternative crops, even for marginal environments. The growing interest in natural medicine, the increasing popularity of herbal dietary supplements, and the multiple possibilities for livestock feeding are all arguments supporting the idea that in many rural areas, this crop could represent a significant tool for enhancing and stabilizing farm income. However, several issues still have to be addressed. The species retains some morphological and physiological traits belonging to non-domesticated plants, which make the application of some common agronomic practices challenging. Furthermore, the lack of reliable field data devoted to the definition of suitable cropping protocols represents a major constraint on the spread of this crop among farmers. This review has therefore focused on updating information on the main morphological and phytochemical traits of the crop and its agronomic characteristics and novel uses. Several gaps in technical knowledge have been addressed, and further goals for experimental activity have been outlined in order to guide farmers eager to cope with the cultivation of such a challenging and resource-rich crop

    Therapeutic sequences in patients with grade 1−2 neuroendocrine tumors (NET): an observational multicenter study from the ELIOS group

    Purpose: Many different treatments are suggested by guidelines to treat grade 1−2 (G1−G2) neuroendocrine tumors (NET). However, a precise therapeutic algorithm has not yet been established. This study aims at identifying and comparing the main therapeutic sequences in G1−G2 NET. Methods: A retrospective observational Italian multicenter study was designed to collect data on therapeutic sequences in NET. Median progression-free survival (PFS) was compared between therapeutic sequences, as well as the number and grade of side effects and the rate of dose reduction/treatment discontinuation. Results: Among 1182 patients with neuroendocrine neoplasia included in the ELIOS database, 131 G1–G2 gastroenteropancreatic, lung and unknown primary NET, unresectable or persistent/relapsing after surgery, treated with ≥2 systemic treatments, were included. Four main therapeutic sequences were identified in 99 patients: (A) somatostatin analogs (SSA) standard dose to SSA high dose (n = 36), (B) SSA to everolimus (n = 31), (C) SSA to chemotherapy (n = 17), (D) SSA to peptide receptor radionuclide therapy (PRRT) (n = 15). Median PFS of the second-line treatment was not reached in sequence A, 33 months in sequence B, 20 months in sequence C, 30 months in sequence D (p = 0.16). Both total number and severity of side effects were significantly higher in sequences B and C than A and D (p = 0.04), as well as the rate of dose reduction/discontinuation (p = 0.03). Conclusions: SSA followed by SSA high dose, everolimus, chemotherapy or PRRT represent the main therapeutic sequences in G1−G2 NET. Median PFS was not significantly different between sequences. However, the sequences with SSA high dose or PRRT seem to be better tolerated than sequences with everolimus or chemotherapy

    Assessment of existing steel frames: Numerical study, pseudo-dynamic testing and influence of masonry infills

    Most of existing steel multi-storey frames in Europe have been designed before the introduction of modern seismic design provisions, hence they often exhibit low performance under earthquake loads due to their low lateral resistance and energy dissipation capacity. In addition, such structures often include rigid and brittle masonry infill walls that highly influence their lateral response and distribution of damage pattern. However, current procedures for the assessment of existing steel buildings in Europe, included in the Eurocode 8 – Part 3 (EC8–3), do not provide adequate guidance for the assessment of ‘weak’ steel frame with masonry infill walls. Moreover, most of available modelling approaches of masonry infills formerly developed for reinforced concrete (RC) structures do not properly represent the behaviour of infill walls in steel frames. An improved numerical has to be provided to satisfactorily mimic infill walls' behaviour in steel moment frames. To this end, an experimental and theoretical study was carried out within the framework of HITFRAMES (i.e., HybrId Testing of an Existing Steel Frame with Infills under Multiple EarthquakeS) SERA project. This paper firstly presents the limitations of current EC8–3 by conducting a code-based assessment on a case study steel moment frame using pushover analysis. Three different single strut models, widely used for simulating the presence of masonry infills in RC structures, are considered for the numerical analyses. The paper also presents the results of pseudo-dynamic (PsD) tests performed on a large-scale 3D steel frame with masonry infills. The capability of the different masonry infill models is successively evaluated by comparisons between numerical and experimental results. On the basis of the obtained results, recommendations on how to potentially improve the single strut model for masonry infills surrounded by steel frames are also provided

    Seismic analysis and retrofitting of an existing R.C. highway bridge: investigation through pseudo-dynamic

    The “Retro” TA project funded by the European commission within the Series-project aims at studying numerically and experimentally the seismic behaviour of an old existing reinforced concrete bridge with portal frame piers and the effectiveness of different isolation systems. In particular, an experimental test campaign will be performed at ELSA Laboratory of JRC (Ispra, Italy). Two piers (scale 1:2.5) will be built and tested using the PsD technique with sub-structuring; the modelling of he entire viaduct is considered along with the non-linear behaviour of each pier, due to bending, shear on the transverse beams and strain penetration effect at the column bases. The comprehensive numerical investigations have shown the high vulnerability of the sample bridge. Consequently two isolation systems (yielding-based and friction-based bearings) have been currently designed and characterized.     A Root in Synapsis and the Other One in the Gut Microbiome-Brain Axis: Are the Two Poles of Ketogenic Diet Enough to Challenge Glioblastoma?

    Glioblastoma is the most frequent and aggressive brain cancer in adults. While precision medicine in oncology has produced remarkable progress in several malignancies, treatment of glioblastoma has still limited available options and a dismal prognosis. After first-line treatment with surgery followed by radiochemotherapy based on the 2005 STUPP trial, no significant therapeutic advancements have been registered. While waiting that genomic characterization moves from a prognostic/predictive value into therapeutic applications, practical and easy-to-use approaches are eagerly awaited. Medical reports on the role of the ketogenic diet in adult neurological disorders and in glioblastoma suggest that nutritional interventions may condition outcomes and be associated with standard therapies. The acceptable macronutrient distribution of daily calories in a regular diet are 45–65% of daily calories from carbohydrates, 20–35% from fats, and 10–35% from protein. Basically, the ketogenic diet follows an approach based on low carbohydrates/high fat intake. In carbohydrates starvation, body energy derives from fat storage which is used to produce ketones and act as glucose surrogates. The ketogenic diet has several effects: metabolic interference with glucose and insulin and IGF-1 pathways, influence on neurotransmission, reduction of oxidative stress and inflammation, direct effect on gene expression through epigenetic mechanisms. Apart from these central effects working at the synapsis level, recent evidence also suggests a role for microbiome and gut-brain axis induced by a ketogenic diet. This review focuses on rationales supporting the ketogenic diet and clinical studies will be reported, looking at future possible perspectives

    Nobiletin and xanthohumol sensitize colorectal cancer stem cells to standard chemotherapy

    Colorectal cancer (CRC) mortality is mainly caused by patient refractoriness to common anti-cancer therapies and consequent metastasis formation. Besides, the notorious toxic side effects of chemotherapy are a concurrent obstacle to be tackled. Thus, new treatment approaches are needed to effectively improve patient outcomes. Compelling evidence demonstrated that cancer stem cells (CSCs) are responsible for treatment failure and relapse. New natural treatment approaches showed capabilities to selectively target the CSC subpopulation by rendering them targetable by standard cytotoxic compounds. Herein we show the anti-cancer properties of the polymethoxyflavones and prenylflavonoids extracted from Citrus sinensis and Humulus lupulus, respectively. The natural biofunctional fractions, singularly and in combination, reduced the cell viability of CRC stem cells (CR-CSCs) and synergized with 5-fluorouracil and oxaliplatin (FOX) chemotherapy. These phenomena were accompanied by a reduced S and G2/M phase of the cell cycle and upregulation of cell death-related genes. Notably, both phytoextracts in combination with FOX thwarted stemness features in CR-CSCs as demonstrated by the impaired clonogenic potential and decreased Wnt pathway activation. Extracts lowered the expression of CD44v6 and affected the expansion of metastatic CR-CSCs in patients refractory to chemotherapy. Together, this study highlights the importance of polymethoxyflavones and prenylflavonoids as natural remedies to aid oncological therapies
