    This study discusses competency and motivation for performance in the Public Works Agency of the Highways of South Sumatra Province. The results of this study prove that the dimensions of competence have a significant effect on improving employee performance at the Public Works Agency of the Highways of South Sumatra Province. This can be proven from the results that can be verified between t count = 3.63 and t table = 2.95. Then the motivation dimension to the performance of employees in the Public Works Agency of Highways of South Sumatra Province does not show more dominant variables on employee performance, this is because the motivation variable has a value of tcount = 2.201 <t table = 2.95.Keywords: Competence, Motivation, Performance, Public Works Agenc

    Therapeutic Effectiveness of Nutrition Therapy in Pediatric Patients with Chronic Liver Diseases Awaiting Liver Transplantation

    Abstract It is important to prevent protein/calorie malnutrition in children with end stage liver diseases prior to transplantation. This study involved 34 patients between the ages of 10 and 156 months (mean value 25.69 months 32.2) (13 females and 21 males) on the liver transplant waiting list. Data collected as of three months before transplant and up to ten months after the procedure concerned gender, age, weight, height, Pediatric End Stage Liver Disease Score, baseline pathology, type of nutrition, type of transplant, immunosuppression, pulse steroid therapy, length of stay, and post transplant complications. Linear regression analysis showed that the length of hospital stay was 24.5 days more for females than for males, but also that intensive nutrition therapy shortens this stay for both female patients (P = 0.085) and younger patients (P = 0.023). The study population was divided into two groups according to the different nutritional therapies adopted. The Student’s t-test and Mann-Whitney test evidenced that the group receiving intensive nutrition therapy grew taller compared with the group following an oral diet (mean -1.37 and Prob = 0.043); that females grew taller compared to males (mean -1.65 +/- 0.56); and that there was an increase in height among the children in the group receiving intensive nutrition therapy despite the presence (-1.37 +/- 0.56) or absence (-14.8 +/- 5.44 and Prob = 0.035) of complications, and despite the administration (-1.03 +/- 0.33) or non administration (-1.48 +/- 0.55 and Prob = 0.019) of steroids. Intensive nutrition therapy enhances the velocity of growth in height and shortens the length of hospital stay, thus optimizing the final prognosis of the baseline pathology

    VALUTAZIONE DEL CALCIO INTRACELLULARE IN PIASTINE DI CANI SANI ED AFFETTI DA LEISHMANIOSI

    Le piastrine rappresentano gli elementi cellulari fondamentali per il processo di regolazione dell’emostasi. La loro interazione con l’endotelio vasale riveste un ruolo fondamentale sia nella fisiologia che nella fisiopatologia della coagulazione. fisiologicamente, infatti, le piastrine hanno la capacità di aderire alla parete del vaso solo dopo attivazione da parte dell’endotelio vasale. In condizioni patologiche, come quelle causate dalla rottura di una placca aterosclerotica, le piastrine aderiscono all’endotelio, si attivano e reclutano numerose altre piastrine per la formazione del tappo piastrinico. In medicina veterinaria, diversamente dalla medicina umana, la conoscenza dei meccanismi fisiologici specifici alla base di tali processi è piuttosto scarsa. Le piastrine subiscono fenomeni di attivazione, fra i quali è possibile menzionare, lo shape change, l’attivazione del recettore per il fibrinogeno che causa l’aggregazione delle piastrine, il rilascio del contenuto dei granuli e la produzione di trombossano A2 (Gachet C. et al., 1996, Mills D.C.B., 1996). Tali fenomeni sono mediati da modificazioni del calcio intracellulare che origina dal rilascio di calcio dagli store intracellulari e dall’entrata di calcio dal mezzo extracellulare (Rink T.J. et al., 1985, Rink T.J. et al., 1990). D’altro canto però, sono note numerose patologie nel cane, tra cui forme di Leishmaniosi eo Ehrlichiosi, che comportano seria compromissione della funzione coagulativa (Harrus S. et al., 1996; Weiss D.J. et al., 1995). La leishmaniosi, in particolare, è una zoonosi largamente diffusa nelle zone costiere del Mediterraneo caratterizzata da una alterazione della coagulazione spesso asintomatica che si inserisce in un quadro sintomatologico estremamente vario. In precedenti ricerche abbiamo mostrato che, in corso di leishmaniosi, si verifica un danno piastrinico di entità variabile che, peraltro, non trova completa risoluzione con le terapie normalmente utilizzate in corso di tale patologia. I meccanismi attraverso i quali l’aggregazione piastrinica risulta alterata in corso di leishmaniosi non sono ancora chiari. Per tale motivo, l’obiettivo di questo studio è stato quello di valutare le variazioni di calcio nelle piastrine di cani sani ed affetti da leishmaniosi allo scopo di raggiungere una migliore comprensione dei meccanismi responsabili di alterazioni dell’emostasi in corso di tale patologia

    Amperometric separation-free immunosensor for real-time environmental monitoring

    Immunoanalytical techniques have found widespread use due to the characteristics of specificity and wide applicability for many analytes, from large polymer antigens, to simple haptens, and even single atoms. Electrochemical sensors offer benefits of technical simplicity, speed and convenience via direct transduction to electronic equipment. Together, these two systems offer the possibility of a convenient, ubiquitous assay technique with high selectivity. However, they are still not widely used, mainly due to the complexity of the associated immunoassay methodologies. A separation-free immunoanalytical technique is described here, which has allowed for the analysis of atrazine in real time and in both quasi-equilibrium and stirred batch configurations. It illustrated that determinations as low as 0.13mM (28 ppb) could be made using equilibrium incubation with an analytical range of 0.1–10mM. Measurements could be made between 1 and 10 mM within several minutes using a real-time, stirred batch method. This system offers the potential for fast, simple, cost-effective biosensors for the analysis of many substances of environmental, biomedical and pharmaceutical concern

    Dual-In/Out strategy for genes integration into bacterial chromosome: a novel approach to step-by-step construction of plasmid-less marker-less recombinant E. coli strains with predesigned genome structure

    <p>Abstract</p> <p>Background</p> <p>The development of modern producer strains with metabolically engineered pathways poses special problems that often require manipulating many genes and expressing them individually at different levels or under separate regulatory controls. The construction of plasmid-less marker-less strains has many advantages for the further practical exploitation of these bacteria in industry. Such producer strains are usually constructed by sequential chromosome modifications including deletions and integration of genetic material. For these purposes complex methods based on <it>in vitro </it>and <it>in vivo </it>recombination processes have been developed.</p> <p>Results</p> <p>Here, we describe the new scheme of insertion of the foreign DNA for step-by-step construction of plasmid-less marker-less recombinant <it>E. coli </it>strains with chromosome structure designed in advance. This strategy, entitled as Dual-In/Out, based on the initial Red-driven insertion of artificial φ80-<it>attB </it>sites into desired points of the chromosome followed by two site-specific recombination processes: first, the φ80 system is used for integration of the recombinant DNA based on selective marker-carrier conditionally-replicated plasmid with φ80-<it>attP</it>-site, and second, the λ system is used for excision of inserted vector part, including the plasmid <it>ori</it>-replication and the marker, flanked by λ-<it>attL/R</it>-sites.</p> <p>Conclusion</p> <p>The developed Dual-In/Out strategy is a rather straightforward, but convenient combination of previously developed recombination methods: phages site-specific and general Red/ET-mediated. This new approach allows us to detail the design of future recombinant marker-less strains, carrying, in particular, rather large artificial insertions that could be difficult to introduce by usually used PCR-based Recombineering procedure. The developed strategy is simple and could be particularly useful for construction of strains for the biotechnological industry.</p

    Assessing measurement systems is a necessary task in all industrial contexts. While a great deal has been written about assessing measurement systems that yield continuous outputs, little work addresses binary measurement systems despite their widespread use. This article proposes two new plans for assessing a binary measurement system that are applicable when we can assume the pass rate of the system is known. This assumption is often reasonable when we need to assess a system used for 100% inspection in a production process. The plans provide estimates of the misclassification rates as well as the proportion of conforming items produced. The two methods are compared to existing plans

    Morphological evaluation of fetal membranes and placenta of wild life Mazama gouazoubira deer in the first third of pregnancy

    Mazama gouazoubira, o veado-catingueiro, é uma espécie de cervídeo de porte pequeno, que pode ser encontrado na América do Sul, desde o sul do Uruguai até o norte de Mato Grosso, no Brasil. Este estudo teve como objetivo descrever as membranas fetais e a placenta de M. gouazoubira no terço inicial de gestação. As amostras coletadas foram analisadas macroscopicamente e microscopicamente. O exame do útero demonstrou uma gestação univitelina e um embrião com crown-rump de 13mm. Na análise do embrião pode ser observado o olho pigmentado, as saliências auriculares, o mesonefro e e metanefro em desenvolvimento, o fígado e sua proeminência externa, o estômago, os membros torácicos e os brotos dos membros pélvicos. A placenta apresentou-se oligocotiledonária e no útero puderam ser observadas nove carúnculas. O saco gestacional mediu 15cm de comprimento e, como observado no início da gestação dos ruminantes domésticos, os cotilédones não puderam ser identificados macroscopicamente. Uma fraca adesão foi observada entre as carúnculas e a membrana corioalantóica (cotilédones) que formavam os placentônios. A membrana corioalantóica demonstrou um alantóide bem vascularizado composto por uma fina camada de células de núcleos e citoplasmas alongados. Na outra face da membrana, o cório foi composto por células cúbicas, de citoplasmas escassos e núcleos grandes e arredondados, características de células trofoblásticas. Envolvendo o embrião visualizou-se o saco amniótico constituído de duas camadas passíveis de separação mecânica, que apresentaram morfologia similar, sendo formadas por um epitélio pavimentoso avascular. Não foi observado saco vitelino no estágio gestacional do espécime estudado. Conclui-se que a placenta de M. gouazoubira é oligocotiledonária, como observado em outros cervídeos, e que as membranas fetais apresentam semelhanças com a de outros ruminantes, incluindo as características citológicas. Estudos adicionais são necessários para determinar a presença do saco vitelino e quando ocorre sua regressão.Mazama gouazoubira is a small-sized deer, which can be found in South America, from southern Uruguay to the north of Mato Grosso, Brazil. This study aimed to describe the fetal membranes and placenta of M. gouazoubira in the first third of pregnancy. Samples collected were macroscopically and microscopically analyzed. Uterus examination demonstrated univitelinic gestation with an embryo measuring a crown-rump length of 13mm. In the embryo analysis we could identify the pigmented eye, auricular hillocks, mesonephron and early metanephron, liver and its external prominence, stomach, thoracic members and buds of pelvic members. Placenta was oligocotyledonary and nine caruncles were found in the uterus. Gestational sac measured 15cm in length, and, as observed in the early gestation of domestic ruminants, cotyledons could not be identified macroscopically. A weak adhesion was found between caruncles and chorioallantoic membrane (cotyledons) forming the placentomes. The chorioallantoic membrane showed a well-vascularized allantoid composed by a thin layer of cells with stretched nucleus and cytoplasm. On the other side of the membrane, the chorion was composed by cubic cells with scant cytoplasm and large-rounded nuclei, features of trophoblast cells. Involving the embryo we identified a two-layered amniotic sac. Layers could be mechanically separated and had similar morphology, showing a non-vascularized squamous epithelium. There was no yolk sac in the gestational stage of this specimen. We concluded that the placenta of M. gouazoubira is an oligocotyledonary placenta, as observed in others cervids, and that the fetal membranes showed similarities with those from other domestic ruminants, including cytological characteristics. Further studies are required in order to determine the presence of the yolk sac and its regression time
