Bulletin No. 345 - Effectiveness of Gravity Drains and Experimental Pumping for Drainage Delta Area, Utah
Cooperatie investigations on drainage and reclamation of salted soils in the Delta Area, Utah, were undertaken by the Utah Agricultural Experiment Station, the U. S. Regional Salinity Laboratory, and Millard County Drainage Districts under a memorandum of understanding effective January 1, 1946.
This investigation comprises project 250 of the Utah Agricultural Experiment Station, R. H. Walker, director; and project 50-46-1 of the U. S. Regional Salinity Laboratory, H. E. Hayward, director.
The objectives of the investigations covered by the memorandum of understanding were:
1. To study the effectiveness of present methods of drainage; tile and open drains.
2. To determine the possibilities of pumping from wells as a method of drainage in the Delta Area and the possibility of using the pumped water for irrigation.
3. To determine practical and economical methods of reclaiming unproductive saline and alkali soils of the area by leaching.
The leaching phase of these investigations has been completed and the results are reported in Experiment Station Bulletin 335 entitled Reclamation of saline-alkali soils by leaching. The first and second \u27phases concerned with the effectiveness of present drainage methods and the possibilities of pumping for drainage are included in this report. Drainage and leaching are inseparable and must be considered as integral parts of an overall reclamation program for areas in which salinity is a problem
Economic Impacts of Farm Program Payment Limits
The high levels of government payments to farmers resulting from the 1985 farm bill have once again led the Congress to examine the payment limit issue. Payment limits were initially established in 1970 and have since been revised several times. In this report, policy and farm management economists analyze the consequences of alternative payment limits on economic efficiency, economic viability of family-size farms, international competitiveness, and consumer food costs. Effective payment limits encourage reduced farm size and in the presence of economies of size, tend to increase production costs for program crops. The Agricultural and Food Policy Center is charged with evaluating economic impacts of policy alternatives -- not recommending, advocating, or opposing particular policies. The Center's orientation is toward Texas agriculture -- evaluating policy impacts on its producers and consumers. Farm prices and income, however, are determined in world markets that are influenced by national economic policy and farm programs. Multicenter prevalence of anaphylaxis in clinic-based oral food challenges
Background
Although previous single-center studies report the rate of anaphylaxis for oral food challenges (OFCs) as 9% to 11%, little is known regarding the epidemiology of clinical OFCs across multiple centers in the United States.
Objective
To examine the epidemiology, symptoms, and treatment of clinical low-risk OFCs in the nonresearch setting.
Methods
Data were obtained from 2008 to 2013 through a physician survey in 5 food allergy centers geographically distributed across the United States. Allergic reaction rates and the association of reaction rates with year, hospital, and demographics were determined using a linear mixed model. Meta-analysis was used to pool the proportion of reactions and anaphylaxis with inverse-variance weights using a random-effects model with exact confidence intervals (CIs).
Results
A total of 6,377 OFCs were performed, and the pooled estimate of anaphylaxis was 2% (95% CI, 1%-3%). The rate of allergic reactions was 14% (95% CI, 13%-16%) and was consistent during the study period (P = .40). Reaction rates ranged from 13% to 33%. Males reacted 16% more frequently than females (95% CI, 4%-37.5%; P = .04). Foods challenged in 2013 varied geographically, with peanut as the most challenged food in the Northeast, Midwest, and West and egg as the most challenged in the South.
Conclusion
As the largest national survey of allergic reactions of clinical open OFCs in a nonresearch setting in the United States, this study found that performing clinical nonresearch open low-risk OFCs results in few allergic reactions, with 86% of challenges resulting in no reactions and 98% without anaphylaxis
PARP14 promotes the warburg effect in hepatocellular carcinoma by inhibiting JNK1-dependent PKM2 phosphorylation and activation
Most tumour cells use aerobic glycolysis (the Warburg effect) to support anabolic growth and evade apoptosis. Intriguingly, the molecular mechanisms that link the Warburg effect with the suppression of apoptosis are not well understood. In this study, using loss-of-function studies in vitro and in vivo, we show that the anti-apoptotic protein poly(ADP-ribose) polymerase (PARP)14 promotes aerobic glycolysis in human hepatocellular carcinoma (HCC) by maintaining low activity of the pyruvate kinase M2 isoform (PKM2), a key regulator of the Warburg effect. Notably, PARP14 is highly expressed in HCC primary tumours and associated with poor patient prognosis. Mechanistically, PARP14 inhibits the pro-apoptotic kinase JNK1, which results in the activation of PKM2 through phosphorylation of Thr365. Moreover, targeting PARP14 enhances the sensitization of HCC cells to anti-HCC agents. Our findings indicate that the PARP14-JNK1-PKM2 regulatory axis is an important determinant for the Warburg effect in tumour cells and provide a mechanistic link between apoptosis and metabolism

Pyruvate kinase M2 activators promote tetramer formation and suppress tumorigenesis
Cancer cells engage in a metabolic program to enhance biosynthesis and support cell proliferation. The regulatory properties of pyruvate kinase M2 (PKM2) influence altered glucose metabolism in cancer. PKM2 interaction with phosphotyrosine-containing proteins inhibits enzyme activity and increases availability of glycolytic metabolites to support cell proliferation. This suggests that high pyruvate kinase activity may suppress tumor growth. We show that expression of PKM1, the pyruvate kinase isoform with high constitutive activity, or exposure to published small molecule PKM2 activators inhibit growth of xenograft tumors. Structural studies reveal that small molecule activators bind PKM2 at the subunit interaction interface, a site distinct from that of the endogenous activator fructose-1,6-bisphosphate (FBP). However, unlike FBP, binding of activators to PKM2 promotes a constitutively active enzyme state that is resistant to inhibition by tyrosine-phosphorylated proteins. These data support the notion that small molecule activation of PKM2 can interfere with anabolic metabolism
