    Identification of onosma visianii roots extract and purified shikonin derivatives as potential acaricidal agents against tetranychus urticae

    There is an increasing need for the discovery of reliable and eco-friendly pesticides and natural plant-derived products may play a crucial role as source of new active compounds. In this research, a lipophilic extract of Onosma visianii roots extract containing 12% of shikonin derivatives demonstrated significant toxicity and inhibition of oviposition against Tetranychus urticae mites. Extensive chromatographic separation allowed the isolation of 11 naphthoquinone derivatives that were identified by spectral techniques and were tested against Tetranychus urticae. All the isolated compounds presented effects against the considered mite and isobutylshikonin (1) and isovalerylshikonin (2) were the most active, being valuable model compounds for the study of new anti-mite agents

    Beta-tricalcium phosphate ceramic triggers fast and robust bone formation by human mesenchymal stem cells

    Due to their osteoconductive and inductive properties, a variety of calcium phosphate (CaP) scaffolds are commonly used in orthopaedics as graft material to heal bone defects. In this study, we have used two CaP scaffolds with different hydroxyapatite (HA) and \u3b2-tricalcium phosphate (\u3b2-TCP) ratios (MBCP\uae; 60/40 and MBCP+\uae; 20/80) to investigate their intrinsic capacity to favour human bone marrow stem cells (hBMSCs) osteogenic differentiation capacity. We report that MBCP+\uae showed in in vitro culture model a higher rate of calcium ion release in comparison with MBCP\uae. In two defined coculture systems, the hBMSC seeded onto MBCP+\uae presented an increased amount of VEGF secretion, resulting in an enhanced endothelial cell proliferation and capillary formation compared with hBMSC seeded onto MBCP\uae. When both ceramics combined with hBMSC were implanted in a nude mouse model, we observed a faster osteogenic differentiation and enhancement mature bone deposition sustained by the presence of a vast host vasculature within the MBCP+\uae ceramics. Bone formation was observed in samples highly positive to the activation of calcium sensing receptor protein (CaSr) on the surface of seeded hBMSC that also shown higher BMP-2 protein expression. With these data we provide valuable insights in the possible mechanisms of ossification and angiogenesis by hBMSC that we believe to be primed by calcium ions released from CaP scaffolds.     A mortalidade embrionária precoce é a maior causa de perdas reprodutivas em bovinos e ela relaciona-se, principalmente com os distúrbios do reconhecimento materno da prenhez originados em disfunções do corpo lúteo ovariano. Recentemente, foi desenvolvido na Embrapa Pecuária Sudeste, um protocolo hormonal que melhora a função luteínica e permite aumentar a oportunidade para um reconhecimento materno da prenhez bem sucedido. O protocolo baseia-se na administração de hormônios reprodutivos (GnRH e hCG) em ocasiões estratégicas após a ovulação. Foram realizados dois experimentos, um com vacas leiteiras e outro com receptoras de embrião, para testar e validar o uso desse protocolo. Após a aplicação do tratamento houve aumento nas taxas de prenhez. Os resultados detalhados do uso dessa prática estão descritos no presente Boletim de Pesquisa e Desenvolvimento. O protocolo testado traduz-se numa prática pecuária de controle das funções ovariana e uterina que otimiza a eficiência reprodutiva de vacas leiteiras e de receptoras de embrião.bitstream/item/43513/1/Boletim27.pd

    TA treatment of depression : a hermeneutic single-case efficacy design study - ‘Caterina’

    This study is the second of a series of seven, and belongs to the second Italian systematic replication of findings from two previous series (Widdowson 2012a, 2012b, 2012c, 2013; Benelli, 2016a, 2016b, 2016c) that investigated the effectiveness of a manualised transactional analysis treatment for depression through Hermeneutic Single-Case Efficacy Design. The therapist was a white Italian woman with 10 years of clinical experience and the client, Caterina, was a 28-year old white Italian woman who attended 16 sessions of transactional analysis psychotherapy. Caterina satisfied DSM-5 criteria for major depressive disorder with generalized anxiety disorder. The conclusion of the judges was that this was an outstanding good-outcome case: the depressive symptoms showed an early clinical and reliable improvement, maintained till the 6 months follow-up, accompanied by reductions in anxiety symptoms, global distress and severity of personal problems. Adherence to the manualised treatment for depression appears good to excellent. In this case study, transactional analysis treatment for depression has proven its efficacy in treating major depressive disorder in comorbidity with anxiety disorder

    Leishmania infantum and Dirofilaria immitis infections in Italy, 2009-2019: Changing distribution patterns

    Background: For long time, canine leishmaniosis (CanL) was considered endemic in the southern, central, and insular regions of Italy, whereas heartworm disease (HW) caused by Dirofilaria immitis was considered endemic in the northern region and in the swampy Po Valley. Following the reports of new foci of both diseases, in this study we update the distribution patterns and occurrence of new foci of CanL and HW discussing the main drivers for the changes in the epidemiology of these two important zoonotic canine vector-borne diseases. Methods: Based on the statistical analyses of serological assays (n = 90,633) on L. infantum exposure and D. immitis infection performed by two reference diagnostic centres in Italy over a ten-year period (2009-2019) irrespective of the anamnesis of dogs. The distribution patterns of both parasites are herein presented along with the occurrence of new foci. Results: Results highlighted the changing distribution patterns of L. infantum vs D. immitis infection in Italy. CanL is endemic in some areas of northern regions and HW has endemic foci in central and southern regions and islands. Significant differences in L. infantum exposure and HW infection prevalence among the study macroareas were detected. The overall results of the positive tested samples were 28.2% in southern Italy and islands, 29.6% in central Italy and 21.6% in northern Italy for L. infantum and 2.83% in northern Italy, 7.75% in central Italy and 4.97% in southern Italy and islands for HW. HW positivity significantly varied over years (χ 2 = 108.401, df = 10, P < 0.0001), gradually increasing from 0.77% in 2009 to 8.47% in 2016-2017. Conclusions: New potential epidemiological scenarios are discussed according to a range of factors (e.g. environmental modifications, occurrence of competent insect vectors, transportation of infected animals to non-endemic areas, chemoprophylaxis or vector preventative measures), which may affect the current distribution. Overall, the results advocate for epidemiological surveillance programmes, more focussed preventative and control measures even in areas where few or no cases of both diseases have been diagnosed.[Figure not available: see fulltext.

    Chemical composition of Cinnamosma madagascariensis (Cannelaceae) essential oil and its larvicidal potential against the filariasis vector Culex quinquefasciatus Say

    Madagascar flora is diverse and unique. Cinnamosma madagascariensis is an endemic species widely present in the forests of Madagascar. This plant has important traditional uses ranging from management of dementia, epilepsy, headache to malaria. Fewdata have been reported about the chemical composition of the essential oil, and no studies have been published on its bioactivity againstmosquitoes. Here,we focus on the chemical composition of essential oils extracted from C. madagascariensis stem bark and leaves, and their larvicidal potential against the filariasis vector Culex quinquefasciatus. GC-MS analysis revealed differences between the chemical volatile profiles of leaves and bark oils. In the former, linalool (30.1%), limonene (12.0%), myrcene (8.9%) and α-pinene (8.4%) were the major constituents, while in the latter β-pinene (33.3%), α-pinene (19.3%) and limonene (12.0%) were the most representative compounds. Acute toxicity experiments conducted on larvae of the filariasis vector C. quinquefasciatus led to LC50 of 61.6 μL L−1 and 80.1 μL L−1 for the bark and the leaf essential oils, respectively. Overall, the chance to use compounds from the C. madagascariensis bark and leaf essential oils against filariasis vectors seems promising, since they are effective at moderate doses and could be an advantageous alternative to build newer and safer mosquito control tools.     Insecticidal Activity of Four Essential Oils Extracted from Chilean Patagonian Plants as Potential Organic Pesticide

    Patagonia is a geographical area characterized by a wide plant biodiversity. Several native plant species are traditionally used in medicine by the local population and demonstrated to be sources of biologically active compounds. Due to the massive need for green and sustainable pesticides, this study was conducted to evaluate the insecticidal activity of essential oils (EOs) from understudied plants growing in this propitious area. Ciprés (Pilgerodendron uviferum), tepa (Laureliopsis philippiana), canelo (Drimys winteri), and paramela (Adesmia boronioides) EOs were extracted through steam distillation, and their compositions were analyzed through GC–MS analysis. EO contact toxicity against Musca domestica L., Spodoptera littoralis (Boisd.), and Culex quinquefasciatus Say was then evaluated. As a general trend, EOs performed better on housefly males over females. Ciprés EO showed the highest insecticidal efficacy. The LD50(90) values were 68.6 (183.7) and 11.3 (75.1) µg adult−1 on housefly females and males, respectively. All EOs were effective against S. littoralis larvae; LD50 values were 33.2–66.7 µg larva−1, and tepa EO was the most effective in terms of LD90 (i.e., <100 µg larva−1). Canelo, tepa, and paramela EOs were highly effective on C. quinquefasciatus larvae, with LC50 values < 100 µL L−1. Again, tepa EO achieved LD90 < 100 µL L−1. This EO was characterized by safrole (43.1%), linalool (27.9%), and methyl eugenol (6.9%) as major constituents. Overall, Patagonian native plant EOs can represent a valid resource for local stakeholders, to develop effective insecticides for pest and vector management, pending a proper focus on their formulation and nontarget effects
