Los comités de ética de investigación en la Ciudad Autónoma de Buenos Aires: a catorce años de la implementación de la Ley 3301
This article analyzes the implementation of research ethics committees in the Autonomous City of Buenos Aires, a process that took place in the context of a historical process of implementing health research policies at both the national and jurisdictional levels. Employing a qualitative, observational, and cross-sectional approach, we gathered secondary sources of public information and conducted semi-structured, in-depth interviews with members of the committees in the Autonomous City of Buenos Aires. The results are presented in relation to three aspects: 1) the composition of the research ethics committees; 2) work processes, emphasizing the impact of the COVID-19 pandemic; and 3) obstacles and proposals, that focuses on improvements identified by committee members.El presente artículo se propone analizar el proceso de implementación de los comités de ética en investigación en la Ciudad Autónoma de Buenos Aires, cuya conformación se observa en el marco de un proceso histórico de implementación de políticas de investigación en salud a nivel nacional y jurisdiccional. Desde un enfoque de investigación cualitativo, observacional y de corte transversal, se realizó un relevamiento de fuentes secundarias de información pública, y entrevistas semiestructuradas y en profundidad a integrantes de los comités de la Ciudad Autónoma de Buenos Aires. Los resultados se presentan agrupados en tres ejes: 1) la conformación de los comités de ética en investigación; 2) procesos de trabajo, haciendo hincapié en el impacto de la pandemia de covid-19; y 3) obstáculos y propuestas, que se focaliza en las mejoras identificadas por sus integrantes

Strategie terapeutiche innovative basate sulla duplice attivit\ue0, antibatterica e probiotica, del predatore batterico Bdellovibrio bacteriovorus
Introduzione: Bdellovibrio bacteriovorus preda batteri Gram-negativi ed \ue8 ubiquitario in vari
ambienti, dove svolge un ruolo di equilibratore delle specie batteriche presenti. Molte sono le
infezioni causate da biofilm microbici misti, di solito resistenti alle cure antibiotiche: ne \ue8 un
esempio la Fibrosi Cistica (FC). Il primo obiettivo del presente studio \ue8 stato valutare
l\u2019attivit\ue0 antibatterica di B. bacteriovorus verso specie batteriche isolate da pazienti FC. Il
secondo obiettivo \ue8 stato valutare l\u2019utilizzo di B. bacteriovorus come probiotico, valutandone
la presenza nell\u2019ecosistema polimicrobico intestinale umano, in soggetti sani e affetti da
Celiachia, Morbo di Crohn, Colite Ulcerosa, e FC.
Metodi: L\u2019attivit\ue0 contro biofilm preformati di Pseudomonas aeruginosa e Staphylococcus
aureus \ue8 stata misurata sia in sistemi statici (piastre) che dinamici (bioflux). Con lo
zymogramma \ue8 stata inoltre valutata l\u2019attivit\ue0 litica contro tali ceppi. Al fine di valutare la
presenza del B. bacteriovorus nell\u2019ecosistema intestinale umano, il DNA totale di campioni
fecali e bioptici collezionati da pazienti sani e malati \ue8 stato utilizzato per effettuare Real-time
PCR con primers B. bacteriovorus-specifici. Risultati: I risultati ottenuti indicano una
predazione di B. bacteriovorus non solo nei confronti del Gram-negativo P. aeruginosa, ma
anche, e per la prima volta osservata, del Gram-positivo S. aureus. Le prove in statico hanno
evidenziato una significativa diminuzione del 70% del biofilm dopo 24 ore di incubazione,
mentre le prove in dinamico una diminuzione del 35% dopo 15 ore di incubazione. Inoltre,
per la prima volta, abbiamo dimostrato la presenza di B. bacteriovorus a livello della mucosa
intestinale e la sua significativa diminuzione nei soggetti malati, rispetto ai controlli.
Conclusioni: I risultati ottenuti incoraggiano una pi\uf9 approfondita ricerca sul Bdellovibrio,
per un suo eventuale utilizzo in strategie terapeutiche innovative volte sia a combattere
infezioni sostenute da biofilm batterici (antibatterico), che al ripristino di ecosistemi microbici

Bdellovibrio bacteriovorus directly attacks Pseudomonas aeruginosa and Staphylococcus aureus Cystic fibrosis isolates
Bdellovibrio bacteriovorus is a predator bacterial species found in the environment and within the human gut, able to attack Gram-negative prey. Cystic fibrosis (CF) is a genetic disease which usually presents lung colonization by Pseudomonas aeruginosa or Staphylococcus aureus biofilms. Here, we investigated the predatory behavior of B. bactenovorus against these two pathogenic species with: (1) broth culture; (2) "static" biofilms; (3) field emission scanning electron microscope (FESEM); (4) "flow" biofilms; (5) zymographic technique. We had the first evidence of B. bacteriovorus survival with a Gram-positive prey, revealing a direct cell-to-cell contact with S. aureus and a new "epibiotic" foraging strategy imaged with FESEM. Mean attaching time of HD100 to S. aureus cells was 185s, while "static" and "flow" S. aureus biofilms were reduced by 74 (at 24h) and 46% (at 20h), respectively. Furthermore, zymograms showed a differential bacteriolytic activity exerted by the B. bacteriovorus lysates on P aeruginosa and S. aureus. The dual foraging system against Gram-negative (periplasmic) and Gram-positive (epibiotic) prey could suggest the use of B. bacteriovorus as a "living antibiotic" in CF, even if further studies are required to simulate its in vivo predatory behavior

Abstracts from the 23rd Italian congress of Cystic Fibrosis and the 13th National congress of Cystic Fibrosis Italian Society
Cystic Fibrosis (CF) occurs most frequently in caucasian populations. Although less common, this disorder have been reported in all the ethnicities. Currently, there are more than 2000 described sequence variations in CFTR gene, uniformly distributed and including variants pathogenic and benign (CFTR1:www.genet.sickkids.on.ca/). To date,only a subset have been firmily established as variants annotated as disease-causing (CFTR2: www.cftr2.org). The spectrum and the frequency of individual CFTR variants, however, vary among specific ethnic groups and geographic areas. Genetic screening for CF with standard panels of CFTR mutations is widely used for the diagnosis of CF in newborns and symptomatic patients, and to diagnose CF carrier status. These screening panels have an high diagnostic sensitivity (around 85%) for CFTR mutations in caucasians populations but very low for non caucasians. Developed in the last decade, Next-Generation Sequencing (NGS) has been the last breakthrough technology in genetic studies with a substantial reduction in cost per sequenced base and a considerable enhancement of the sequence generation capabilities. Extended CFTR gene sequencing in NGS includes all the coding regions, the splicing sites and their flankig intronic regions, deep intronic regions where are localized known mutations,the promoter and the 5'-3' UTR regions. NGS allows the analysis of many samples concurrently in a shorter period of time compared to Sanger method . Moreover, NGS platforms are able to identify CFTR copy number variation (CNVs), not detected by Sanger sequencing.
This technology has provided new and reliable approaches to molecular diagnosis of CF and CFTR-Related Disorders. It also allows to improve the diagnostic sensitivity of newborn and carrier screeningmolecular tests. In fact, bioinformatics tools suitable for all the NGS platforms can filter data generated from the gene sequencing, and
analyze only mutations with well-established disease liability. This approach allows the development of targeted mutations panels with a higher number of frequent CF mutations for the target populationcompared to the standard panels and a consequent enhancement of the diagnostic sensitivity. Moreover, in the emerging challenge of diagnosing CF in non caucasians patients, the possibility of customize a NGS targeted mutations panel should increase the diagnostic sensitivity when the target
population has different ethnicities
