    A Two-stage Polynomial Method for Spectrum Emissivity Modeling

    Spectral emissivity is a key in the temperature measurement by radiation methods, but not easy to determine in a combustion environment, due to the interrelated influence of temperature and wave length of the radiation. In multi-wavelength radiation thermometry, knowing the spectral emissivity of the material is a prerequisite. However in many circumstances such a property is a complex function of temperature and wavelength and reliable models are yet to be sought. In this study, a two stages partition low order polynomial fitting is proposed for multi-wavelength radiation thermometry. In the first stage a spectral emissivity model is established as a function of temperature; in the second stage a mathematical model is established to describe the dependence of the coefficients corresponding to the wavelength of the radiation. The new model is tested against the spectral emissivity data of tungsten, and good agreement was found with a maximum error of 0.64

    Real-time study of rapid spread of antibiotic resistance plasmid in biofilm using microfluidics

    Gene transfer in biofilms is known to play an important role in antibiotic resistance dissemination. However, the process remains poorly understood. In this study, microfluidics with time-lapse imaging was used for real-time monitoring of plasmid-mediated horizontal gene transfer (HGT) in biofilms. Pseudomonas putida KT2440 harboring an antibiotic resistance plasmid RP4 was chosen as the donor while Escherichia coli and activated sludge bacteria were used as the recipient cells. Dynamic features of the transfer process, including the transfer rate, cell growth rate and kinetic changes of the transfer frequency, were determined. It was found that the routes for gene transfer strongly depend on the structure and composition of a biofilm. While intraspecies HGT is essential to initiate a transfer event, the secondary retransfer from transconjugants to the same species is more efficient and can cause cascading gene spread in single-strain biofilms. For the activated sludge biofilm, only small and scattered colonies formed and vertical gene transfer appears to be the dominant route after initial intraspecies transfer. Furthermore, more than 46% of genera in the activated sludge were permissive to plasmid RP4, many of which are associated with human pathogens. These phenomena imply early prevention and interruptions to biofilm structure could provide an effect way to inhibit rapid antibiotic resistance gene spread and reduce the likelihood of catastrophic events associated with antibiotic resistance

    Erythromycin Enhances CD4+Foxp3+ Regulatory T-Cell Responses in a Rat Model of Smoke-Induced Lung Inflammation

    Heavy smoking can induce airway inflammation and emphysema. Macrolides can modulate inflammation and effector T-cell response in the lungs. However, there is no information on whether erythromycin can modulate regulatory T-cell (Treg) response. This study is aimed at examining the impact of erythromycin on Treg response in the lungs in a rat model of smoking-induced emphysema. Male Wistar rats were exposed to normal air or cigarette smoking daily for 12 weeks and treated by gavage with 100 mg/kg of erythromycin or saline daily beginning at the forth week for nine weeks. The lung inflammation and the numbers of inflammatory infiltrates in bronchoalveolar lavage fluid (BALF) were characterized. The frequency, the number of Tregs, and the levels of Foxp3 expression in the lungs and IL-8, IL-35, and TNF-α in BALF were determined by flow cytometry, RT-PCR and ELISA, respectively. Treatment with erythromycin reduced smoking-induced inflammatory infiltrates, the levels of IL-8 and TNF-α in the BALF and lung damages but increased the numbers of CD4+Foxp3+ Tregs and the levels of Foxp3 transcription in the lungs, accompanied by increased levels of IL-35 in the BALF of rats. Our novel data indicated that erythromycin enhanced Treg responses, associated with the inhibition of smoking-induced inflammation in the lungs of rats

    Protective effect of low-dose risedronate against osteocyte apoptosis and bone loss in ovariectomized rats

    Osteocyte apoptosis is the first reaction to estrogen depletion, thereby stimulating osteoclastic bone resorption resulting in bone loss. We investigated the effects of two different risedronate (RIS) doses (high and low) on osteocyte apoptosis, osteoclast activity and bone loss in ovariectomized rats. Forty rats with ovariectomy (OVX) and sham ovariectomy (SHAM) were divided into 4 groups: 1) SHAM rats treated with saline (SHAM); 2) OVX rats treated with saline (OVX); 3) OVX rats treated with low-dose RIS (OVX-LR, 0.08 μg/kg/day); 4) OVX rats treated with high-dose RIS (OVX-HR, 0.8 μg/kg/day). All animals were sacrificed 90 days after surgery for the examinations of osteocyte apoptosis by caspase-3 staining, osteoclast activity by TRAP staining and bone volume by micro-CT scanning in lumbar vertebral cancellous bone. Both low and high dose RIS significantly reduced caspase-3 positive osteocytes, empty lacunae and TRAP positive osteoclasts in OVX rats. Although the difference in caspase-3 positive osteocytes was not significant between the OVX-LR and OVX-HR groups, numerically these cells were significantly more prevalent in OVX-HR (not OVX-LR) group than in SHAM group. TRAP positive osteoclasts were significantly higher in OVX-LR group than in SHAM or OVX-HR group. There was no significant difference in bone volume among the OVX-LR, OVX-HR and SHAM groups, but lower in OVX group alone. However, significant increase in trabecular thickness only occurred in OVX-LR group. We conclude that both low and high dose RIS significantly inhibit osteocyte apoptosis and osteoclast activity in OVX rats, but the low-dose RIS has weaker effect on osteoclast activity. However, low-dose RIS preserves cancellous bone mass and microarchitecture as well as high-dose RIS after estrogen depletion

    Reducing Bidirectional Pedestrian Conflict Based on Lane Formation Phenomenon in Subway Corridors

    With the rapid increase of the subway passenger volume, the conflict among passengers emerges as a significant issue which affects subway serviceability, especially in the bidirectional flow. The aim of this study is to explore the characteristics of the bidirectional flow of pedestrians in a subway corridor. Pedestrian experiments were conducted to investigate microscopic characteristics of the pedestrian flow. It was found that the microscopic characteristics, including the walking speed and turning angle, were time-dependent and had a generalized trend with time. It was also found that different pedestrian volumes affected the microscopic characteristics. Based on the trend of the microscopic characteristics, the lane formation phenomenon was observed and quantitatively studied, identifying three phases: conflict phase, lane formation phase, and steady lane phase. To alleviate the bidirectional pedestrian conflict, additional pedestrian experiments for the countermeasure of adding separating strap in the corridor, which was based on the lane formation analysis, was conducted. The effectiveness of the countermeasure was demonstrated through a before-and-after comparison. The results showed that adding the separation between the adjacent lanes had the best performance in reducing the conflicts. The results would provide a rationale for subway managers in optimizing the corridor bidirectional pedestrian flow.</p
