    Role of miR-155 and miR-146a in Mast Cell Function

    Mast cells are resident immune cells abundantly found in the tissue at the host-environment interface, where they play a critical role in inflammatory allergic responses. Mast cell responses may be regulated by the cytokine milieu at the site of inflammation. Recent studies have revealed microRNAs to be important in altering cytokine signaling in immune cells. Here, we demonstrate for the first time that IL-10 and IL-33 induce miR-155 and miR-146a, respectively, to alter mast cell functions. We report that IL-10 enhanced IgE induced activation of mast cells. IL-10 effects are dependent on Stat3 activation, which elicits miR-155 expression, resulting in a loss of suppressor of cytokine signaling-1 (SOCS-1). The importance of miR-155 was demonstrated by the inability of IL-10 to enhance anaphylaxis in miR-155–deficient mice. Additionally, we show that IL-33 treatment greatly enhances miR-146a expression in mast cells and in mast cell derived exosomes. miR-146a induction is dependent on MyD88 and NFκB and seems to negatively regulate ST2 signaling, which is demonstrated by the hyperresponsiveness of miR-146a knockout BMMC in response to IL-33. Our preliminary data suggest that miR-146a serves as a feedback negative regulator of IL-33 signaling by targeting IRAK proteins. miR-155 and miR-146a are key microRNAs that regulate a range of immune functions. Taken together, our results reveal two novel microRNA pathways that regulate mast cell IgE and IL-33 induced responses

    Peace Conceptual Metaphor Analysis in the Afghanistan Peacemaking Process - Case Study: Afghanistan Peace Map

    In this research, we have used the descriptive-analytic method. The main purpose of this paper is to show the reflection and perception of the peace conceptual metaphor in Afghanistan's discourse space on the basis of "conceptual metaphor theory." Humans typically reflect on concrete or more physical things, according to this linguistic theory. The findings of this study show that the country's peace-making operatives reflected the abstract and nonphysical concept of peace on a variety of concrete and physical things. For example, on the basis of the "Afghanistan Peace Map" context, the peace abstract concept is reflected on the experimental and objective concepts, like: "journey", "structure", and "live objects." Therefore, the conceptual metaphors of "peace is journey", "peace is structure" and "peace is live objects" are the three more prevalent concepts that are reflected in this context. These metaphors are not only more prevalent and user-friendly in the context of the "Afghanistan peace map," but research into published contexts in the Afghanistan peace-making process reveals that these metaphors flowed in the substructure of all peace contexts

    Selection of potent bacterial strain for over-production of PHB by using low cost carbon source for eco-friendly bioplastics

    Background: The microbial PHB production is a promising tool for the plastic industry for the synthesis of environmental friendly, biodegradable plastic in contrast to the conventional petro-chemical based non-degradable plastics. The selection of potent bacterial strains, inexpensive carbon source, efficient fermentation and recovery processes are important aspects that were taken into account during this study.Methods: Different bacterial strains i.e. Bacillus Spp, P. putida and P. fluorescens were screened for maximum PHB production. Under media optimization, various carbon and nitrogen sources (alone or in combination) were used to achieve the maximum PHB production. Finally the degradation tests of the PHB sheet were also performed to test its biodegradability potential.Results: Shake flask studies have shown the PHB concentrations upto 7.02, 4.50 and 34.4 mg/g of dry cell mass of P. putida, P. fluorescens and Bacillus Spp. respectively. Almost same results were observed at laboratory scale production of PHB in 10 L fermenter i.e. 6.28, 6.23 and 39.5 mg/g of dry cell mass by P. putida, P. fluorescens and Bacillus Spp. respectively. On the basis of these observations, Bacillus Spp. was chosen for laboratory scale PHB production. Corn steep liquor (4%) was chosen as the best medium to achieve the highest PHB contents. Isolated PHB has shown biodegradation in soil up to 86.7% at 37oC.Conclusion: The Bacillus Spp. Proved to be the best strain for PHB production on only 4% CSL which is cheapest and easily available

    Association of Anterior Alveolar Dimensions with Different Sagittal Jaw Relationships

Methodology: This Cross-sectional study took place in Orthodontic Department, Institute of Dentistry, Liaquat University of Medical and Health Sciences, Jamshoro, from August 2018 to January 2019. Patients of both genders ranging in age from 18 to 30 years were included. All the subjects, as per ANB angle, were grouped into three categories as (Class I = value between 1° and 4° for ANB angle), (Class II = value > 5° for ANB angle) and (Class III= value <1° for ANB angle). All the data was recorded in the Performa for the purpose of analysis. Results: A total of 90 patients were studied; their average age was 21.12+3.47 years and 52.2% were females. Mean upper posterior alveolus width was significantly higher in sagittal class II as 12.69 ± 5.52 than in sagittal class I and III p-value 0.058. The mean upper anterior alveolus height was found to be significantly greater in class I and III in contrast to class II p-value 0.028. Mean lower anterior alveolus width was insignificantly related with sagittal classification, p-value 0.343. Mean upper anterior alveolus width and lower posterior alveolus width were insignificantly related to sagittal classification, and the p-value were quite insignificant. Sagittal class II and III were significantly associated with female gender 0.021, while class I was linked to male gender p-value 0.021. Conclusion:  There was a significant association between dimensions of anterior alveolar among different vertical and sagittal jaw association

    Factors Affecting Employer-Employee Relationship in Police Sector of Pakistan: The Mediating Role of Working Condition

    The study examines the factors i.e. (collective bargaining, recruitment, remuneration, communication) affecting employer- employee relationship with inclusion of mediator i.e. workplace condition. Primary data was collected from police departments working in four provinces of Pakistan namely, Khyber Pakhtunkhwa, Sindh, Punjab and Baluchistan. Questionnaires were administered among the three tiers of leadership i.e. top, middle and the bottom. For validation of data collection instrument the confirmatory factor analysis via structure equation model was conducted and all the models were found with significant loadings. Liseral and SPSS software were used for data analyses. For measuring direct and mediating effect study used hierarchal linear regression model. The findings of the study indicate that collective bargaining, recruitment, communication and remuneration have significant positive effect on employer-employee relationship whereas, workplace condition partially mediates the relationship amid factor affecting employee relation i.e. (collective bargaining, recruitment, communication and remuneration) and employer- employee relationship. Moreover, study reveals that inclusion of collective bargaining, recruitment, communication and remuneration inside organization will result in better performance of the police department and lead to build trust and better image of police department. Theoretical implications and future area suggestions are also incorporated in the study
