    Penicillin-resistant isolates of Neisseria-lactamica produce altered forms of penicillin-binding protein-2 that arose by interspecies horizontal gene-transfer

    Isolates of Neisseria lactamica that have increased resistance to penicillin have emerged in recent years. Resistance to penicillin was shown to be due to the production of altered forms of penicillin-binding protein 2 (PBP 2) that have reduced affinity for the antibiotic. The sequences of the PBP 2 genes (penA) from two penicillin-resistant isolates were almost identical (less than or equal to 1% sequence divergence) to that of a penicillin-susceptible isolate, except in a 175-bp region where the resistant and susceptible isolates differed by 27%. The nucleotide sequences of these divergent regions were identical (or almost identical) to the sequence of the corresponding region of the penA gene of N. flavescens NCTC 8263. Altered forms of PBP 2 with decreased affinity for penicillin in the two penicillin-resistant isolates of N. lactamica appear, therefore, to have arisen by the replacement of part of the N. lactamica penA gene with the corresponding region from the penA gene of N. flavescens

    Non-beta-lactamase-producing, penicillin-resistant strains of Neisseria meningitidis produce altered forms of penicillin-binding protein 2 that have decreased affinity for penicillin. The sequence of the penicillin-binding protein 2 gene (penA) from a penicillin-resistant strain of N. meningitidis was compared to the sequence of the same gene from penicillin-sensitive strains and from penicillin-sensitive and penicillin-resistant strains of Neisseria gonorrhoeae. The penA genes from penicillin-sensitive strains of N. gonorrhoeae and N. meningitidis were 98% identical. The gene from the penicillin-resistant strain of N. meningitidis consisted of regions that were almost identical to the corresponding regions in the penicillin-sensitive strains (less than 0.2% divergence) and two regions that were very different from them (approximately 22% divergence). The two blocks of altered sequence have arisen by the replacement of meningococcal sequences with the corresponding regions from the penA gene of Neisseria flavescens and result in an altered form of penicillin-binding protein 2 that contains 44 amino acid substitutions and 1 amino acid insertion compared to penicillin-binding protein 2 of penicillin-sensitive strains of N. meningitidis. A similar introduction of part of the penA gene of N. flavescens, or a very similar commensal Neisseria species, appears to have occurred independently during the development of altered penA genes in non-beta-lactamase-producing penicillin-resistant strains of N. gonorrhoeae

    Antioxidative Activities of Natural Hydroxy-Bearing Cinnamaldehydes and Cinnamic Acids: A Comparative Study

    Purpose: To determine the potent antioxidants among six hydroxy-bearing cinnamaldehyde compounds and their corresponding acids.Methods: The antioxidative activities were evaluated by scavenging 2, 2’-diphenyl-1-picrylhydrazyl (DPPH) free radicals and anti-hemolysis of human red blood cells (RBCs). Fourier transform infrared spectrometry(FT-IR, nuclear magnetic resonance (NMR)and mass spectrometer(MS) evaluations were used to determine the synthesized compounds.Results: The scavenging ability of the compounds on DPPH radicals was in the following rank order: caffeic aldehyde (F) ≈ caffeic acid (C) > vitamin C (Vc) > o-coumaraldehyde (D) > p-coumaric acid (B) > p-coumaraldehyde (E) ≈o-coumaric acid. Inhibitory ability against 2′-Azobis(2-methylpropionamidine) dihydrochloride (AAPH)-induced erythrocyte hemolysis was ranked as follows: o-coumaraldehyde (D) > p-coumaraldehyde (E) ≈ caffeic aldehyde (F) > caffeic acid (C) ≈o-coumaric acid (A) > p-coumaric acid (B) > Vitamin C. The corresponding inhibition time (tinh ) of D and Vc was 63.6, and 23.7 min, respectively.Conclusion: Cinnamaldehydes demonstrated superior antioxidative activities to their corresponding acids. DPPH scavenging ability correlated directly with the number of hydroxyl groups on the catechol ring while the degree of lipophilicity of the compounds may be proportional to their anti-hemolytic activity.Keywords: DPPH scavenging, Cinnamaldehyde, Cinnamic acid, Antioxidative, Hemolysis

    Temporal and Spatial Dynamics of Carbon Fixation by Moso Bamboo (Phyllostachys pubescens) in Subtropical China

    To study the temporal and spatial dynamics of carbon fixation by Moso bamboo (Phyllostachys pubescens) in subtropical China, carbon fixation of leaves within the canopy of P. pubescens was measured with a LI-6400 portable photosynthesis system. The results showed that the capability of carbon fixation of P. pubescens leaves had obvious temporal and spatial dynamic variations. It was revealed that there were two peak periods and two low periods in the season variation of carbon fixation capability. Data also revealed that the capability of carbon fixation by five-year-old P. pubescens was more than that of one-year-old and three-year-old. Daily and seasonal carbon fixation showed a negative correlation with the CO2 concentration. The temporal and spatial dynamics of carbon fixation by P. pubescens described above provided a scientific basis for development of technologies in bamboo timber production

    Superconducting receiver coils for sodium magnetic resonance imaging

    Presents the results from sodium magnetic resonance imaging (MRI) experiments using high-temperature superconducting (HTS) receiver coils. Sodium imaging has been shown to have great potential for the assessment of cell integrity but suffers from substantially lower signal-to-noise ratio (SNR) than that of hydrogen imaging. The use of an HTS receiver coil was found to significantly increase the SNR relative to an equivalent copper receiver coil at room temperature. The SNR gains afforded by HTS coils can also be used to decrease the imaging time.published_or_final_versio

    Urban energy consumption and CO2 emissions in Beijing: current and future

    This paper calculates the energy consumption and CO2 emissions of Beijing over 2005–2011 in light of the Beijing’s energy balance table and the carbon emission coefficients of IPCC. Furthermore, based on a series of energy conservation planning program issued in Beijing, the Long-range Energy Alternatives Planning System (LEAP)-BJ model is developed to study the energy consumption and CO2 emissions of Beijing’s six end-use sectors and the energy conversion sector over 2012–2030 under the BAU scenario and POL scenario. Some results are found in this research: (1) During 2005–2011, the energy consumption kept increasing, while the total CO2 emissions fluctuated obviously in 2008 and 2011. The energy structure and the industrial structure have been optimized to a certain extent. (2) If the policies are completely implemented, the POL scenario is projected to save 21.36 and 35.37 % of the total energy consumption and CO2 emissions than the BAU scenario during 2012 and 2030. (3) The POL scenario presents a more optimized energy structure compared with the BAU scenario, with the decrease of coal consumption and the increase of natural gas consumption. (4) The commerce and service sector and the energy conversion sector will become the largest contributor to energy consumption and CO2 emissions, respectively. The transport sector and the industrial sector are the two most potential sectors in energy savings and carbon reduction. In terms of subscenarios, the energy conservation in transport (TEC) is the most effective one. (5) The macroparameters, such as the GDP growth rate and the industrial structure, have great influence on the urban energy consumption and carbon emissions

    Assessment of multiple hormone activities of a UV-filter (octocrylene) in zebrafish (Danio rerio)

    © 2016 Elsevier Ltd In this study, zebrafish (Danio rerio) were exposed to a UV-filter-octocrylene (OCT) with elevated concentrations for 28 d. The total body accumulation of OCT in zebrafish was found to reach 2321.01 (“L” level), 31,234.80 (“M” level), and 70,593.38 ng g−1 (“H” level) when the average OCT exposure concentration was controlled at 28.61, 505.62, and 1248.70 μg L−1, respectively. Gross and histological observations as well as RT-qPCR analysis were conducted to determine the effects of OCT accumulation on zebrafish. After exposure, the gonad-somatic index and percentage of vitellogenic oocytes were found to increase significantly in the ovaries of female zebrafish at the H accumulation level. Significant up-regulation of esr1 and cyp19b were observed in the gonads, as well as vtg1 in the livers for both female and male zebrafish. At M and H accumulation levels, apparent down-regulation of ar was observed in the ovaries and testis of the female and male zebrafish, respectively. Although the extent of the effects on zebrafish differed at different accumulation levels, the induction of vtg1 and histological changes in the ovaries are indications of estrogenic activity and the inhibition of esr1 and ar showed antiestrogenic and antiandrogenic activity, respectively. Thus, as OCT could easily accumulate in aquatic life such as zebrafish, one of its most of concern hazards would be the disturbance of the histological development and its multiple hormonal activities

    A Comparison of Centering Algorithms in the Astrometry of Cassini Imaging Science Subsystem Images and Anthe’s Astrometric Reduction

    In the CAVIAR software package, a standard tool for astrometry of images from the Cassini imaging science subsystem (ISS), Gaussian fitting is used to measure the centre of point-like objects, achieving a typical precision of about 0.2 pixels. In this work, we consider how alternative methods may improve on this. We compare three traditional centroiding methods: two-dimensional Gaussian fitting, median, and modified moment. Results using 56 selected images show that the centroiding precision of the modified moment method is significantly better than the other two methods, with standard deviations for all residuals in sample and line of 0.065 and 0.063 pixels, respectively, representing a factor of over 2 improvement compared to Gaussian fitting. Secondly, a comparison of observations using Cassini ISS images of Anthe is performed. Anthe results show a similar improvement. The modified moment method is then used to reduce all ISS images of Anthe during the period 2008–2017. The observed-minus-calculated residuals relative to the JPL SAT393 ephemeris are calculated. In terms of α × cos(δ) and δ in the Cassini-centred international celestial reference frame, mean values of all residuals are close to 0 km, and their standard deviations are less than 1 km for narrow angle camera images, and about 4 km for wide angle camera images
