De lâInstrumentarium au MusĂ©um. Le cabinet de Jean-François SĂ©guier (1703-1784)
Le cabinet de Jean-François SĂ©guier est probablement un de ceux qui, dans les annĂ©es 1760-1770, jouirent de la renommĂ©e la plus flatteuse dans la partie mĂ©ridionale du Royaume, bien quâil ne se caractĂ©risĂąt guĂšre par une mĂ©ridionalitĂ© affirmĂ©e dans sa composition. Dâabord cabinet dâĂ©tude, alimentant la production intellectuelle lors du sĂ©jour vĂ©ronais de SĂ©guier auprĂšs de Scipione Maffei, il revĂȘtit par la suite une dimension didactique et dĂ©monstrative lors du retour Ă NĂźmes en 1755. La donation des collections Ă lâAcadĂ©mie royale de NĂźmes en 1778 ne fit quâentĂ©riner la constitution dâun musĂ©um Ă part entiĂšre, façonnĂ©e par une longue expĂ©rience de la collection nourrie par lâĂ©change Ă©pistolaire et par une connaissance peu commune et directe des grands cabinets europĂ©ens. Rassemblant, selon un schĂ©ma hĂ©ritĂ© de lâhumanisme savant, naturalia et antiqua, en un mĂȘme lieu, lâĂ©tude ce cabinet permet de fait dâinterroger les ressorts et les finalitĂ©s de la collection Ă travers la figure complexe dâune personnalitĂ© incontournable de lâĂ©rudition provinciale.Jean-François SĂ©guierâs cabinet is probably one of those which, in the years 1760-1770, enjoyed the most flattering fame in the Southern part of the Kingdom, although it was not characterised by a real French southern anchoring. Cabinet of study, feeding the intellectual production during the Veronese stay of SĂ©guier with Scipione Maffei, it took then a didactic and demonstrative dimension after Seguier went back to NĂźmes in 1755. In 1778 the donation of collections to the Royal Academy of NĂźmes confirmed the constitution of a true museum, shaped by a long experiment of the collection and fed by the correspondence and by the direct knowledge of great European cabinets. Rooted in a plan inherited from a tradition of learned humanism, the collections gathered naturalia and antiqua in the same place. The study of this cabinet brings to understand the mechanisms and the purpose of collection through the complex and prominent character of one of the most outstanding figures of provincial erudition

Lâorientation sexuelle, facteur de suicide et de conduites Ă risque chez les adolescents et les jeunes adultes ? : lâinfluence de lâhomophobie et de la victimation homophobe en milieu scolaire
Several studies (mostly North-American) put into light a significant link between sexual orientation and suicidal behaviors among teenagers and/or young adults. Homophobia to which the young homosexuals are exposed is considered the most serious hypothesis to explain this link. However quantitative studies have never been conducted on this hypothesis in France. We took it into account while carrying out a survey based on a self-questionnaire built up from scales from âAdolescent Risk Inventoryâ (Lescano & al., 2007), Suicidal Behaviors Questionnaire â revisedâ (Linehan, 1989), and Homophobia Scaleâ (Wright, Adams & Bernat, 1999). Said survey was answered by 475 young women, and 426 young men aged 18-24.Results confirmed oversuicidality among young homo/bisexuals. However, in the analysis conducted on male participants only, sexual orientation ceases to appear a significant predictor of suicide attempts or risk behavior in explanatory models in which variables « Homophobic Verbal Bullying in school » and « Bullying based on Gender nonconformity » are introduced. Said variables turn out to be the strongest predictors of suicidal behaviors among male adolescents and young adults. Young men have mean scores on the homophobia/lesbophobia scale that are significantly higher than young women's, and homophobic verbal bullying seems a lot more common than lesbophobic verbal bullying. Besides, lesbophobic verbal bullying does not have a significant effect on suicidal attempts and risk behaviors among young women. However, internalized lesbophobia has a significant effect on suicidal behaviors of the sub-sample of young women sexually attracted to same-sex individuals or wondering (ASMSQ), whereas internalized homophobia has no effect on ASMSQ young men. This last result is based on a lower population, and should be confirmed by subsequent studies, whereas analysis on all participants highlight the extent of homophobic bullying in school, and its impact on male teenagers and young adults.De nombreuses recherches, principalement nord-amĂ©ricaines, ont mis en Ă©vidence un lien significatif entre orientation sexuelle et comportements suicidaires des adolescent e s / jeunes adultes (Beck & al., 2010). Les rĂ©sultats confirment la sursuicidalitĂ© des jeunes homo/bi-sexuel-le-s. Cependant, dans les analyses menĂ©es auprĂšs des participants de sexe masculin, lâorientation sexuelle nâapparait plus comme un prĂ©dicteur significatif des tentatives de suicide et des conduites Ă risque dans les modĂšles explicatifs oĂč lâon introduit les variables «Victimation Verbale Homophobe en milieu scolaire» et «Victimation basĂ©e sur la Non-conformitĂ© de genre». Ces variables sâavĂšrent dâailleurs les plus forts prĂ©dicteurs des comportements suicidaires chez les garçons. Cette derniĂšre ne prĂ©sente par ailleurs pas dâeffet significatif sur les tentatives de suicide et les conduites Ă risque des filles. En revanche, la lesbophobie intĂ©riorisĂ©e a un effet significatif sur les comportements suicidaires dans le sous-Ă©chantillon des filles attirĂ©es sexuellement par le mĂȘme sexe ou en questionnement (ASMSQ), alors que lâhomophobie intĂ©riorisĂ©e nâen prĂ©sente pas chez les garçons ASMSQ. Systematic mapping of regions of human cardiac troponin I involved in binding to cardiac troponin C: N- and C-terminal low affinity contributing regions
AbstractThe Spot method of multiple peptide synthesis was used to map in a systematic manner regions of the human cardiac troponin I sequence (hcTnI) involved in interactions with its physiological partner, troponin C (cTnC). Ninety-six 20-mer peptides describing the entire hcTnI sequence were chemically assembled; their reactivity with [125I]cTnC, in the presence of 3 mM Ca2+, enabled the assignment of six sites of interaction (residues 19â32, 45â54, 129â138, 145â164, 161â178 and 191â210). For several sites, a good correlation with literature data was obtained, thus validating this methodological approach. Synthetic peptides, each containing in their sequence an interaction site, were prepared. As assessed by BIACORE, all of them exhibited an affinity for cTnC in the range of 10â6â10â7 M, except for hcTnI [39â58] which showed a nanomolar affinity. This peptide was also able to block the interaction between hcTnI and cTnC. We therefore postulate that despite the existence of multiple cTnC interaction sites on the hcTnI molecule, only that region of hcTnI allows a stabilization of the complex. Residues 19â32 from the N-terminal cardio-specific extension of hcTnI were also found to be involved in interaction with cTnC; residues 19â32 may correspond to the minimal sequence of the extension which could switch between the N- and C-terminal TnC domains, depending on its phosphorylation state. Finally, two Ca2+-dependent cTnC binding domains within the C-terminal part of hcTnI (residues 164â178 and 191â210) were also mapped. The latter site may be linked with the cardiac dysfunction observed in stunned myocardium
PIP30/FAM192A is a novel regulator of the nuclear proteasome activator PA28Îł
PA28Îł is a nuclear activator of the 20S proteasome involved in the regulation of several essential cellular processes, such as cell proliferation, apoptosis, nuclear dynamics, and cellular stress response. Unlike the 19S regulator of the proteasome, which specifically recognizes ubiquitylated proteins, PA28Îł promotes the degradation of several substrates by the proteasome in an ATP- and ubiquitin-independent manner. However, its exact mechanisms of action are unclear and likely involve additional partners that remain to be identified. Here we report the identification of a cofactor of PA28Îł, PIP30/FAM192A. PIP30 binds directly and specifically via its C-terminal end and in an interaction stabilized by casein kinase 2 phosphorylation to both free and 20S proteasome-associated PA28Îł. Its recruitment to proteasome-containing complexes depends on PA28Îł and its expression increases the association of PA28Îł with the 20S proteasome in cells. Further dissection of its possible roles shows that PIP30 alters PA28Îł-dependent activation of peptide degradation by the 20S proteasome in vitro and negatively controls in cells the presence of PA28Îł in Cajal bodies by inhibition of its association with the key Cajal body component coilin. Taken together, our data show that PIP30 deeply affects PA28Îł interactions with cellular proteins, including the 20S proteasome, demonstrating that it is an important regulator of PA28Îł in cells and thus a new player in the control of the multiple functions of the proteasome within the nucleus
