    Of bandits and saints: JesĆŗs Malverde and the struggle for place in Sinaloa, Mexico

    JesĆŗs Malverde, a bandit who was assassinated in 1909, crystallizes the struggle for place-understood both literally and metaphorically-in northern Mexico. The socially and economically marginal people who revered him in the nineteenth century adore him as a lay saint today. Contention over building a chapel to Malverde in CuliacĆ”n, the capital city of the northern Mexican state of Sinaloa, distils broader tensions over the Mexican stateā€™s persistent deferral of the poor from inclusion in the official landscape of the nation. Malverdeā€™s appropriation by Sinaloaā€™s narcotraffickers as their patron saint extends this symbolic and material claim to legitimacy to include those who exist outside the official boundaries. The border between the sacred and the profane is often a site of social struggle, and the case of Malverde is no exception. While the legend of Malverde may well have been invented, its negotiation has proven remarkably long-lived and powerful in shaping and reshaping the iconographic and material landscapes of social inclusion and exclusion. Malverde thus offers an empty signifier whose multiple interpretations yield a surplus of symbolic meanings and material production based on the circulation, negotiation, appropriation, and reinterpretation of those meanings

    Elder Financial Exploitation: Implications for Future Policy and Research in Elder Mistreatment

    Recent advances in the understanding of elder mistreatment have demonstrated that financial exploitation tends to be one of the most common forms of mistreatment affecting older populations. Agencies such as the World Bank and World Health Organization show significant concern regarding financial exploitation and its connection to physical and emotional injury to victims. The World Bank uses the term ā€œfinancial violenceā€ as a means of generally describing the harm caused to an individual as a result of financial exploitation or abuse. The proportion of financial exploitation in relation to other forms of elder mistreatment is defined in our research. We discuss the potential impact of elder financial exploitation on victims as well as explore the implications for future research and policy development focused on financial aspects of elder mistreatment and call for further study in the concept of financial exploitation as a violent act

    Crystal Structure and Twisted Aggregates of Oxcarbazepine Form III

    Polymorphism and crystal habit play vital roles in dictating the properties of crystalline materials. Here, the structure and properties of oxcarbazepine (OXCBZ) form III are reported along with the occurrence of twisted crystalline aggregates of this metastable polymorph. OXCBZ III can be produced by crystallization from the vapor phase and by recrystallization from solution. The crystallization process used to obtain OXCBZ III is found to affect the pitch, with the most prominent effect observed from the sublimation-grown OXCBZ III material where the pitch increases as the length of aggregates increases. Sublimation-grown OXCBZ III follows an unconventional mechanism of formation with condensed droplet formation and coalescence preceding nucleation and growth of aggregates. A crystal structure determination of OXCBZ III from powder X-ray diffraction methods, assisted by crystal structure prediction (CSP), reveals that OXCBZ III, similar to carbamazepine form II, contains void channels in its structure with the channels, aligned along the c crystallographic axis, oriented parallel to the twist axis of the aggregates. The likely role of structural misalignment at the lattice or nanoscale is explored by considering the role of molecular and closely related structural impurities informed by crystal structure prediction

    Novel rat Alzheimer's disease models based on AAV-mediated gene transfer to selectively increase hippocampal AĪ² levels

    <p>Abstract</p> <p>Background</p> <p>Alzheimer's disease (AD) is characterized by a decline in cognitive function and accumulation of amyloid-Ī² peptide (AĪ²) in extracellular plaques. Mutations in amyloid precursor protein (APP) and presenilins alter APP metabolism resulting in accumulation of AĪ²42, a peptide essential for the formation of amyloid deposits and proposed to initiate the cascade leading to AD. However, the role of AĪ²40, the more prevalent AĪ² peptide secreted by cells and a major component of cerebral AĪ² deposits, is less clear. In this study, virally-mediated gene transfer was used to selectively increase hippocampal levels of human AĪ²42 and AĪ²40 in adult Wistar rats, allowing examination of the contribution of each to the cognitive deficits and pathology seen in AD.</p> <p>Results</p> <p>Adeno-associated viral (AAV) vectors encoding BRI-AĪ² cDNAs were generated resulting in high-level hippocampal expression and secretion of the specific encoded AĪ² peptide. As a comparison the effect of AAV-mediated overexpression of APPsw was also examined. Animals were tested for development of learning and memory deficits (open field, Morris water maze, passive avoidance, novel object recognition) three months after infusion of AAV. A range of impairments was found, with the most pronounced deficits observed in animals co-injected with both AAV-BRI-AĪ²40 and AAV-BRI-AĪ²42. Brain tissue was analyzed by ELISA and immunohistochemistry to quantify levels of detergent soluble and insoluble AĪ² peptides. BRI-AĪ²42 and the combination of BRI-AĪ²40+42 overexpression resulted in elevated levels of detergent-insoluble AĪ². No significant increase in detergent-insoluble AĪ² was seen in the rats expressing APPsw or BRI-AĪ²40. No pathological features were noted in any rats, except the AAV-BRI-AĪ²42 rats which showed focal, amorphous, Thioflavin-negative AĪ²42 deposits.</p> <p>Conclusion</p> <p>The results show that AAV-mediated gene transfer is a valuable tool to model aspects of AD pathology <it>in vivo</it>, and demonstrate that whilst expression of AĪ²42 alone is sufficient to initiate AĪ² deposition, both AĪ²40 and AĪ²42 may contribute to cognitive deficits.</p

    TNF-block genotypes influence susceptibility to HIV-associated sensory neuropathy in Indonesians and South Africans

    HIV-associated sensory neuropathy (HIV-SN) is a disabling complication of HIV disease and antiretroviral therapies (ART). Since stavudine was removed from recommended treatment schedules, the prevalence of HIV-SN has declined and associated risk factors have changed. With stavudine, rs1799964*C (TNF-1031) associated with HIV-SN in Caucasians and Indonesians but not in South Africans. Here, we investigate associations between HIV-SN and rs1799964*C and 12 other polymorphisms spanning TNF and seven neighboring genes (the TNF-block) in Indonesians (n = 202; 34/168 cases) and South Africans (n = 75; 29/75 cases) treated without stavudine. Haplotypes were derived using fastPHASE and haplotype networks built with PopART. There were no associations with rs1799964*C in either population. However, rs9281523*C in intron 10 of BAT1 (alternatively DDX39B) independently associated with HIV-SN in Indonesians after correcting for lower CD4 T-cell counts and \u3e500 copies of HIV RNA/mL (model p = 0.0011, Pseudo R2 = 0.09). rs4947324*T (between NFKBIL1 and LTA) independently associated with reduced risk of HIV-SN and shared haplotype 1 (containing no minor alleles) associated with increased risk of HIV-SN after correcting for greater body weight, a history of tuberculosis and nadir CD4 T-cell counts (model: p = 0.0003, Pseudo R2 = 0.22). These results confirm TNF-block genotypes influence susceptibility of HIV-SN. However, critical genotypes differ between ethnicities and with stavudine use

    ACCORD (ACcurate COnsensus Reporting Document): a reporting guideline for consensus methods in biomedicine developed via a modified Delphi

    Background:Ā In biomedical research, it is often desirable to seek consensus among individuals who have differing perspectives and experience. This is important when evidence is emerging, inconsistent, limited, or absent. Even when research evidence is abundant, clinical recommendations, policy decisions, and priority-setting may still require agreement from multiple, sometimes ideologically opposed parties. Despite their prominence and influence on key decisions, consensus methods are often poorly reported. Our aim was to develop the first reporting guideline dedicated to and applicable to all consensus methods used in biomedical research regardless of the objective of the consensus process, called ACCORD (ACcurate COnsensus Reporting Document). Methods and findings:Ā We followed methodology recommended by the EQUATOR Network for the development of reporting guidelines: a systematic review was followed by a Delphi process and meetings to finalize the ACCORD checklist. The preliminary checklist was drawn from the systematic review of existing literature on the quality of reporting of consensus methods and suggestions from the Steering Committee. A Delphi panel (nĀ = 72) was recruited with representation from 6 continents and a broad range of experience, including clinical, research, policy, and patient perspectives. The 3 rounds of the Delphi process were completed by 58, 54, and 51 panelists. The preliminary checklist of 56 items was refined to a final checklist of 35 items relating to the article title (nĀ = 1), introduction (nĀ = 3), methods (nĀ = 21), results (nĀ = 5), discussion (nĀ = 2), and other information (nĀ = 3). Conclusions:Ā The ACCORD checklist is the first reporting guideline applicable to all consensus-based studies. It will support authors in writing accurate, detailed manuscripts, thereby improving the completeness and transparency of reporting and providing readers with clarity regarding the methods used to reach agreement. Furthermore, the checklist will make the rigor of the consensus methods used to guide the recommendations clear for readers. Reporting consensus studies with greater clarity and transparency may enhance trust in the recommendations made by consensus panels

    CD161 expression defines new human Ī³Ī“ T cell subsets

    Ī³Ī“ T cells are a highly versatile immune lineage involved in host defense and homeostasis, but questions remain around their heterogeneity, precise function and role during health and disease. We used multiāˆ’parametric flow cytometry, dimensionality reduction, unsupervised clustering, and self-organizing maps (SOM) to identify novel Ī³Ī“ T cell naĆÆve/memory subsets chiefly defined by CD161 expression levels, a surface membrane receptor that can be activating or suppressive. We used middle-to-old age individuals given immune blockade is commonly used in this population. Whilst most VĪ“1+subset cells exhibited a terminal differentiation phenotype, VĪ“1āˆ’ subset cells showed an early memory phenotype. Dimensionality reduction revealed eight Ī³Ī“ T cell clusters chiefly diverging through CD161 expression with CD4 and CD8 expression limited to specific subpopulations. Comparison of matched healthy elderly individuals to bronchiectasis patients revealed elevated VĪ“1+ terminally differentiated effector memory cells in patients potentially linking this population with chronic proinflammatory disease
