    The use of models in planning: the development of a land use planning strategy.

The complete thesis in print form is available from the University of Adelaide Library.With the advent of comprehensive transportation studies over the last several years in Australia, some inadequacies in land use p1anning have been high1ighted. In general there is seen to be a need for a more systematic approach and a greater use of models or analytical techniques. This thesis briefly reviews transportation studies and planning techniques and then discusses the development and current status of urban development mode1s, classifying them into three viz; (i) descriptive (ii) predictive models, and (iii) prescriptive models or planning models. It is noted that the models form on1y a part of the of the planning process and that they are really sub-models total system. Using a systems analysis approach, a planning framework or system model is then developed which forms the basis for the preparation of a planning strategy, within which detailed development and transport plans can be evolved. The framework is designed to facilitate the use of models as befits their operational status and to highlight areas where judgements and intuition are required. In developing the p1anning system model a discussion is given on forecasting techniques, problems relating to the definition of objectives and criteria, difficu1ties encountered in obtaining community goals and rational public participation, considerations relating to the formulation of plans, techniques of plan eva1uation inc1uding sensitivity analysis and flexibility tests, and implementation strategy. Two case studies are given to illustrate the efficacy of the procedures proposed. The first example related to the preparation of a strategic plan for a city with an expected population of 200,000 people, whereas the second case study concerns the hypothetical choice between two alternative forms of development over the next five years for metropolitan Adelaide.     Unsteady flow and particle migration in dense, non-Brown suspensions

    We present experimental results on dense corn-starch suspensions as examples of non-Brownian, nearly-hard particles that undergo continuous and discontinuous shear thickening (CST and DST) at intermediate and high densities respectively. Our results offer strong support for recent theories involving a stress-dependent effective contact friction among particles. We show however that in the DST regime, where theory might lead one to expect steady-state shear bands oriented layerwise along the vorticity axis, the real flow is unsteady. To explain this, we argue that steady-state banding is generically ruled out by the requirement that, for hard non-Brownian particles, the solvent pressure and the normal-normal component of the particle stress must balance separately across the interface between bands.     The role of friction in the yielding of adhesive non-Brownian suspensions

    Yielding behavior is well known in attractive colloidal suspensions. Adhesive non-Brownian suspensions, in which the interparticle bonds are due to finite-size contacts, also show yielding behavior. We use a combination of steady-state, oscillatory and shear-reversal rheology to probe the physical origins of yielding in the latter class of materials, and find that yielding is not simply a matter of breaking adhesive bonds, but involves unjamming from a shear-jammed state in which the micro-structure has adapted to the direction of the applied load. Comparison with a recent constraint-based rheology model shows the importance of friction in determining the yield stress, suggesting novel ways to tune the flow of such suspensions.Comment: 8 pages, 6 figure

    A Subset of Human Bromodomains Recognizes Butyryllysine and Crotonyllysine Histone Peptide Modifications

    SummaryBromodomains are epigenetic readers that are recruited to acetyllysine residues in histone tails. Recent studies have identified non-acetyl acyllysine modifications, raising the possibility that these might be read by bromodomains. Profiling the nearly complete human bromodomain family revealed that while most human bromodomains bind only the shorter acetyl and propionyl marks, the bromodomains of BRD9, CECR2, and the second bromodomain of TAF1 also recognize the longer butyryl mark. In addition, the TAF1 second bromodomain is capable of binding crotonyl marks. None of the human bromodomains tested binds succinyl marks. We characterized structurally and biochemically the binding to different acyl groups, identifying bromodomain residues and structural attributes that contribute to specificity. These studies demonstrate a surprising degree of plasticity in some human bromodomains but no single factor controlling specificity across the family. The identification of candidate butyryl- and crotonyllysine readers supports the idea that these marks could have specific physiological functions

    SORCS1 and SORCS3 control energy balance and orexigenic peptide production

    SORCS1 and SORCS3 are two related sorting receptors expressed in neurons of the arcuate nucleus of the hypothalamus. Using mouse models with individual or dual receptor deficiencies, we document a previously unknown function of these receptors in central control of metabolism. Specifically, SORCS1 and SORCS3 act as intracellular trafficking receptors for tropomyosin-related kinase B to attenuate signaling by brain-derived neurotrophic factor, a potent regulator of energy homeostasis. Loss of the joint action of SORCS1 and SORCS3 in mutant mice results in excessive production of the orexigenic neuropeptide agouti-related peptide and in a state of chronic energy excess characterized by enhanced food intake, decreased locomotor activity, diminished usage of lipids as metabolic fuel, and increased adiposity, albeit at overall reduced body weight. Our findings highlight a novel concept in regulation of the melanocortin system and the role played by trafficking receptors SORCS1 and SORCS3 in this process

    The Impact of Satellite Sea Surface Salinity for Prediction of the Coupled Indo-Pacific System

    We assess the impact of satellite sea surface salinity (SSS) observations on seasonal to interannual variability of tropical Indo-Pacific Ocean dynamics as well as on dynamical ENSO forecasts. Our coupled model is composed of a primitive equation ocean model for the tropical Indo-Pacific region that is coupled with the global SPEEDY atmospheric model (Molteni, 2003). The Ensemble Reduced Order Kalman Filter is used to assimilate observations to constrain dynamics and thermodynamics for initialization of the coupled model. The baseline experiment assimilates satellite sea level, SST, and in situ subsurface temperature and salinity observations. This baseline is then compared with experiments that additionally assimilate Aquarius (version 4.0) and SMAP (version 2.0) SSS. Twelve-month forecasts are initialized for each month from Sep. 2011 to Dec. 2016. We find that including satellite SSS significantly improves NINO3.4 sea surface temperature anomaly validation after 1 out to 12 month forecast lead times. For initialization of the coupled forecast, the positive impact of SSS assimilation is brought about by surface freshening near the eastern edge of the western Pacific warm pool and density changes that lead to shallower mixed layer between 10S-5N. SST differences at initialization force wide-spread downwelling favorable curl over most of the tropical Pacific. Over an average forecast, SST remains warmer with SSS assimilation at the eastern edge of the warm pool. This warm SST propagates into the eastern Pacific and drags westerly wind anomalies eastward into the NINO3.4 region. In addition, salting near the ITCZ leads to a deepening of the mixed layer and thermocline near 8N. These patterns together lead to a funneling effect that provides the background state to amplify equatorial Kelvin waves. We show that the downwelling Kelvin waves are amplified by assimilating satellite SSS and lead to significantly improved forecasts particularly for the 2015 El Nino

    The binary systems that have been detected in gamma rays have proven very useful to study high-energy processes, in particular particle acceleration, emission and radiation reprocessing, and the dynamics of the underlying magnetized flows. Binary systems, either detected or potential gamma-ray emitters, can be grouped in different subclasses depending on the nature of the binary components or the origin of the particle acceleration: the interaction of the winds of either a pulsar and a massive star or two massive stars; accretion onto a compact object and jet formation; and interaction of a relativistic outflow with the external medium. We evaluate the potentialities of an instrument like the Cherenkov telescope array (CTA) to study the non-thermal physics of gamma-ray binaries, which requires the observation of high-energy phenomena at different time and spatial scales. We analyze the capability of CTA, under different configurations, to probe the spectral, temporal and spatial behavior of gamma-ray binaries in the context of the known or expected physics of these sources. CTA will be able to probe with high spectral, temporal and spatial resolution the physical processes behind the gamma-ray emission in binaries, significantly increasing as well the number of known sources. This will allow the derivation of information on the particle acceleration and emission sites qualitatively better than what is currently available

    The last decade has seen significant growth in the field of sensor networks, which are currently collecting large amounts of environmental data. This data needs to be collected, processed, stored and made available for analysis and interpretation in a manner which is meaningful and accessible to end users and stakeholders with a range of requirements, including government agencies, environmental agencies, the research community, industry users and the public. The COMMONSENSE project aims to develop and provide cost-effective, multi-functional innovative sensors to perform reliable in-situ measurements in the marine environment. The sensors will be easily usable across several platforms, and will focus on key parameters including eutrophication, heavy metal contaminants, marine litter (microplastics) and underwater noise descriptors of the MSFD. The aims of Tasks 2.1 and 2.2 which comprise the work of this deliverable are: • To obtain a comprehensive understanding and an up-to-date state of the art of existing sensors. • To provide a working basis on “new generation” technologies in order to develop cost-effective sensors suitable for large-scale production. This deliverable will consist of an analysis of state-of-the-art solutions for the different sensors and data platforms related with COMMONSENSE project. An analysis of relevant technical issues and deficiencies of existing sensors and related initiatives currently set and working in marine environment will be performed. Existing solutions will be studied to determine the main limitations to be considered during novel sensor developments in further WP’s. Objectives & Rationale The objectives of deliverable 2.1 are: • To create a solid and robust basis for finding cheaper and innovative ways of gathering data. This is preparatory for the activities in other WPs: for WP4 (Transversal Sensor development and Sensor Integration), for WP(5-8) (Novel Sensors) to develop cost-effective sensors suitable for large-scale production, reducing costs of data collection (compared to commercially available sensors), increasing data access availability for WP9 (Field testing) when the deployment of new sensors will be drawn and then realized
