Study of cosmogenic activation above ground for the DarkSide-20k experiment
The activation of materials due to exposure to cosmic rays may become an important background source for experiments investigating rare event phenomena. DarkSide-20k, currently under construction at the Laboratori Nazionali del Gran Sasso, is a direct detection experiment for galactic dark matter particles, using a two-phase liquid-argon Time Projection Chamber (TPC) filled with 49.7 tonnes (active mass) of Underground Argon (UAr) depleted in 39Ar. Here, the cosmogenic activity of relevant long-lived radioisotopes induced in the experiment has been estimated to set requirements and procedures during preparation of the experiment and to check that it is not dominant over primordial radioactivity; particular attention has been paid to the activation of the 120 t of UAr used in DarkSide-20k. Expected exposures above ground and production rates, either measured or calculated, have been considered in detail. From the simulated counting rates in the detector due to cosmogenic isotopes, it is concluded that activation in copper and stainless steel is not problematic. The activity of 39Ar induced during extraction, purification and transport on surface is evaluated to be 2.8% of the activity measured in UAr by DarkSide-50 experiment, which used the same underground source, and thus considered acceptable. Other isotopes in the UAr such as 37Ar and 3H are shown not to be relevant due to short half-life and assumed purification methods
Implications of miRNA expression pattern in bovine oocytes and follicular fluids for developmental competence
Developmental competence determines the oocyte capacity to support initial embryo growth, but the molecular mechanisms underlying this phenomenon are still ill-defined. Changes in microRNA (miRNA) expression pattern have been described during follicular growth in several species. Therefore, aim of this study was to investigate whether miRNA expression pattern in cow oocyte and follicular fluid (FF) is associated with the acquisition of developmental competence. Samples were collected from ovaries with more than, or fewer than, 10 mid-antral follicles (H- and L-ovaries) because previous studies demonstrated that this parameter is a reliable predictor of oocyte competence. After miRNA deep sequencing and bioinformatic data analysis, we identified 58 miRNAs in FF and 6 in the oocyte that were differentially expressed between H- and L-ovaries. Overall, our results indicate that miRNA levels both in FF and in the ooplasm must remain within specific thresholds and that changes in either direction compromising oocyte competence. Some of the miRNAs found in FF (miR-769, miR-1343, miR-450a, miR-204, miR-1271 and miR-451) where already known to regulate follicle growth and their expression pattern indicate that they are also involved in the acquisition of developmental competence. Some miRNAs were differentially expressed in both compartments but with opposite patterns, suggesting that miRNAs do not flow freely between FF and oocyte. Gene Ontology analysis showed that the predicted gene targets of most differentially expressed miRNAs are part of a few signalling pathways. Regulation of maternal mRNA storage and mitochondrial activity seem to be the processes more functionally relevant in determining oocyte quality. In conclusion, our data identified a few miRNAs in the follicular fluid and in the ooplasm that modulate the oocyte developmental competence. This provides new insights that could help with the management of cattle reproductive efficiency
Diagnostic potential of simplified methods for measuring glomerular filtration rate to detect chronic kidney disease in dogs
Background: Glomerular filtration rate (GFR) is the most sensitive indicator of initial renal function decline during chronic kidney disease (CKD), but conventional protocols for measuring GFR are labor-intensive and stressful for the dog.
Objectives: To assess the diagnostic potential for detecting CKD with simplified GFR protocols based on iohexol plasma clearance.
Animals: Seventeen CKD positive and 23 CKD negative dogs of different breeds and sex.
Methods: Prospective nonrandomised study. Plasma iohexol was measured 5, 15, 60, 90, 180 min after injection. GFR was calculated using five samples (GFR5) or simplified protocols based on one, two, or three samples. GFR5 and simplified GFR were compared by Bland-Altmann and concordance correlation coefficient (CCC) analysis, and diagnostic accuracy for CKD by receiver operating characteristic curves. A grey zone for each protocol was bounded by the fourth quartile of the CKD positive population (lower cut-off) and the first quartile of the CKD negative population (upper cut-off).
Results: All simplified protocols gave reliable GFR measurements, comparable to reference GFR5 (CCC > 0.92). Simplified protocols which included the 180-min sampling granted the best GFR measure (CCC: 0.98), with strong diagnostic potential for CKD (area under the receiver operating characteristic curve \ub1 SE: 0.98 \ub1 0.01). A double cut-off including a zone of CKD uncertainty guaranteed reliable diagnosis outside the grey area, and identified borderline dogs inside it.
Conclusions: The simplified GFR protocols offer an accurate, hands-on tool for CKD diagnosis in dogs. The grey zone might help decision-making in the management of early kidney dysfunction
A microfabricated sensor for thin dielectric layers
We describe a sensor for the measurement of thin dielectric layers capable of
operation in a variety of environments. The sensor is obtained by
microfabricating a capacitor with interleaved aluminum fingers, exposed to the
dielectric to be measured. In particular, the device can measure thin layers of
solid frozen from a liquid or gaseous medium. Sensitivity to single atomic
layers is achievable in many configurations and, by utilizing fast, high
sensitivity capacitance read out in a feedback system onto environmental
parameters, coatings of few layers can be dynamically maintained. We discuss
the design, read out and calibration of several versions of the device
optimized in different ways. We specifically dwell on the case in which
atomically thin solid xenon layers are grown and stabilized, in cryogenic
conditions, from a liquid xenon bath
Maternal exposure to di(2-ethylhexyl)phthalate (DEHP) promotes the transgenerational inheritance of adult-onset reproductive dysfunctions through the female germline in mice
Endocrine disruptors (EDS) are compounds known to promote transgenerational inheritance of adult-onset disease in subsequent generations after maternal exposure during fetal gonadal development. This study was designed to establish whether gestational and lactational exposure to the plasticizer di(2-ethylhexyl)phthalate (DEHP) at environmental doses promotes transgenerational effects on reproductive health in female offspring, as adults, over three generations in the mouse.
Gestating F0 mouse dams were exposed to 0, 0.05, 5 mg/kg/day DEHP in the diet from gestational day 0.5 until the end of lactation. The incidence of adult-onset disease in reproductive function was recorded in F1, F2 and F3 female offspring.
In adult Fl females, DEHP exposure induced reproductive adverse effects with: i) altered ovarian follicular dynamics with reduced primordial follicular reserve and a larger growing pre-antral follicle population, suggesting accelerated follicular recruitment; ii) reduced oocyte quality and embryonic developmental competence; iii) dysregulation of the expression profile of a panel of selected ovarian and pre-implantation embryonic genes. F2 and F3 female offspring displayed the same altered reproductive morphological phenotype and gene expression profiles as Fl, thus showing transgenerational transmission of reproductive adverse effects along the female lineage. These findings indicate that in mice exposure to DEHP at doses relevant to human exposure during gonadal sex determination significantly perturbs the reproductive indices of female adult offspring and subsequent generations. Evidence of transgenerational transmission has important implications for the reproductive health and fertility of animals and humans, significantly increasing the potential biohazards of this toxicant
Cosmogenic 11C production and sensitivity of organic scintillator detectors to pep and CNO neutrinos
Several possible background sources determine the detectability of pep and
CNO solar neutrinos in organic liquid scintillator detectors. Among such
sources, the cosmogenic 11C nuclide plays a central role. 11C is produced
underground in reactions induced by the residual cosmic muon flux. Experimental
data available for the effective cross section for 11C by muons indicate that
11C will be the dominant source of background for the observation of pep and
CNO neutrinos. 11C decays are expected to total a rate 2.5 (20) times higher
than the combined rate of pep and CNO neutrinos in Borexino (KamLAND) in the
energy window preferred for the pep measurement, between 0.8 and 1.3 MeV.
This study examines the production mechanism of 11C by muon-induced showers
in organic liquid scintillators with a novel approach: for the first time, we
perform a detailed ab initio calculation of the production of a cosmogenic
nuclide, 11C, taking into consideration all relevant production channels.
Results of the calculation are compared with the effective cross sections
measured by target experiments in muon beams.
This paper also discusses a technique for reduction of background from 11C in
organic liquid scintillator detectors, which allows to identify on a one-by-one
basis and remove from the data set a large fraction of 11C decays. The
background reduction technique hinges on an idea proposed by Martin Deutsch,
who suggested that a neutron must be ejected in every interaction producing a
11C nuclide from 12C. 11C events are tagged by a three-fold coincidence with

Early surgical anteroseptal ventricular endocardial restoration after acute myocardial infarction : pathophysiology and surgical considerations
Background. The efficacy and safety of surgical anteroseptal ventricular endocardial restoration (a procedure that excludes non-contracting scarred segments) in the left ventricle with chronic dilation and remodeling secondary to an anterior myocardial infarction are well established. We present a small series and discuss the indication for early left ventricular restoration in the setting of complicated acute myocardial infarction. Methods. Early ventricular restoration was performed in 8 patients (4 males, 4 females, mean age 70 \ub1 8 years). A postinfarction ventricular septal defect was diagnosed in 3 cases. All patients were operated, on an urgent or emergent basis, between 1 and 16 days following the onset of infarction. Surgical coronary revascularization was associated in 7 patients. Results. There was one operative death. At discharge, echocardiographic morphofunctional assessment revealed: a reduction of the left ventricular end-diastolic and end-systolic volume indexes, an increase of the ejection fraction, and, most importantly, an improvement of remote myocardial shortening fraction. At a mean follow-up of 15.6 months (range 2-21 months), there were no late deaths and all survivors are in NYHA functional class I or II. Conclusions. Left ventricular restoration may represent an effective adjunct to the surgical management of patients with an acute extensive anterior myocardial infarction complicated by severe heart failure, with or without septal rupture
Solar neutrino detection in a large volume double-phase liquid argon experiment
Precision measurements of solar neutrinos emitted by specific nuclear
reaction chains in the Sun are of great interest for developing an improved
understanding of star formation and evolution. Given the expected neutrino
fluxes and known detection reactions, such measurements require detectors
capable of collecting neutrino-electron scattering data in exposures on the
order of 1 ktonne yr, with good energy resolution and extremely low background.
Two-phase liquid argon time projection chambers (LAr TPCs) are under
development for direct Dark Matter WIMP searches, which possess very large
sensitive mass, high scintillation light yield, good energy resolution, and
good spatial resolution in all three cartesian directions. While enabling Dark
Matter searches with sensitivity extending to the "neutrino floor" (given by
the rate of nuclear recoil events from solar neutrino coherent scattering),
such detectors could also enable precision measurements of solar neutrino
fluxes using the neutrino-electron elastic scattering events. Modeling results
are presented for the cosmogenic and radiogenic backgrounds affecting solar
neutrino detection in a 300 tonne (100 tonne fiducial) LAr TPC operating at
LNGS depth (3,800 meters of water equivalent). The results show that such a
detector could measure the CNO neutrino rate with ~15% precision, and
significantly improve the precision of the 7Be and pep neutrino rates compared
to the currently available results from the Borexino organic liquid
