    Polarization in the ELAIS-N1 LOFAR deep field

    The physical characteristics of cosmic magnetic fields are encoded in the po- larization properties of the extragalactic radio sources. Linearly polarized radiation undergoes Faraday rotation as it crosses a magneto-ionic medium; the rotation of the polarization angle of the signal is equal to its wavelength squared times the Rotation Measure (RM), that is proportional to the line- of-sight integral of the product of the magnetic field component along the line of sight and the density of thermal electrons. Mapping the values of the RM across the sky provides a mean to constrain cosmic magnetic fields. For this purpose, statistical studies of the properties of polarized radio sources spread over cosmological distances are essential.The advantage of low radio-frequency observations over higher-frequency ones is the better precision on the inferred RM values. On the other hand, at low radio-frequency observations are more affected by depolarization, which affect the detection rate. Indeed, the population of faint polarized extragalac- tic sources at low radio frequencies is still mostly unknown.In this context, the LOw Frequency ARray (LOFAR) plays an important role because of sensitivity, angular resolution and precision on the inferred RM values that can be achieved through low-frequency broad-band polarimetry, allowing us to study the polarized radio emission at frequencies around 150 MHz.In our work, we developed a new method to combine polarimetric obser- vations with slightly different frequency configurations, and we applied this method to the European Large Area ISO Survey-North 1 (ELAIS-N1) deep field, one of the deepest of the LOFAR Two-Meter Sky Survey (LoTSS) Deep Fields so far, at 114.9–177.4 MHz. We imaged an area of 25 deg2 at 6-arcsec of resolution in which, through stacking of 19 8-hour-long epochs, detected 1.28 sources per square degrees, the highest number density of polarized sources ever found at low radio frequencies. We compared our results with other RM catalogs and we quantified the depolarization properties of sources detected also at 1.4 GHz. We also modeled the source counts in polarization from the source counts in total flux density.This work dealt with technical and theoretical challenges inherent to the observation and interpretation of polarimetric data and represents a step in solving complex issues that modern radio astronomy is facing due the large amounts of data generated by new-generation radio-interferometers

    Unusual skin metastasis due to adenocarcinoma of the stomach: a case report

    Clinical Findings A 68- year-old man presented with a skin thickening of the anterior neck started few months earlier. Upper respiratory and digestive tract had already been examined which proved normal. On clinical examination a hyper pigmented massively indurated leathery plaque was seen on the anterior and lateral aspect of the neck from the submandibular region down to the jugular fossae. The plaque was firm, well demarcated and measured 15 by 12 cm in diameter. It had a cobblestone like appearance with exaggerated folds, and was non painful. Patient complained of a mild discomfort while swallowing saliva of recent onset, but was otherwise asymptomatic. All his routine blood test was normal. The full skin examination was otherwise unremarkable. Our main differential diagnosis at that stage included a reactive process such as a sclerodermatous process, a neoplastic reaction or a lymph proliferative disease.</br

    Investigation into Cryptosporidium and Giardia in bivalve mollusks farmed in Sardinia region and destined for human consumption

    Cryptosporidium and Giardia are protozoan parasites transmitted by fecal-oral ingestion of (oo)cysts, and are responsible for enteritis in several animal species and humans worldwide. These (oo)cysts can survive for over a year in aquatic environments and can accumulate in bivalve mollusks, which filter large volumes of water. The aim of this study is to evaluate the natural occurrence of Cryptosporidium and Giardia contamination in different specimens of edible bivalves mollusks from farming sites of the western and north-eastern coasts of Sardinia. From April 2011 to February 2012, 1095 specimens of Mytilus galloprovincialis and 240 of Crassostrea gigas were sampled from Olbia and Oristano gulf and San Teodoro pond. Hepatopancreas and gills, including the labial palp, were examined for oocysts and cysts after pooling and homogenisation using different techniques: i) staining for light and fluorescence microscopy; ii) direct immunofluorescence (IF) MerifluorÂź test Cryptosporidium/ Giardia (Meridian Bioscience Inc., Cincinnati, OH, USA); and iii) molecular procedures. However, in the context under study, all mollusks examined with the three main diagnostic techniques were negative for both parasites pointing out the hypothetically low zoonotic risk related to Cryptosporidium and Giardia in bivalves, especially Mytilus galloprovincialis and Crassostrea gigas

    Anterior segment mesenchymal dysgenesis in a large Australian family is associated with the recurrent 17 bp duplication in PITX3

    Purpose: A recurrent 17 bp duplication (c.657ins17bp) of a segment of the paired-like homeodomain transcription factor 3 (PITX3) gene on human chromosome 10 has been reported in seven families with autosomal dominant posterior polar cataracts with or without anterior segment mesenchymal dysgenesis (ASMD). ASMD can include Peters anomaly with corneal clouding, iridolenticular corneal adhesions, displaced Schwalbe's line, and cataract as described previously in a large Australian family. This study reports the examination of PITX3 in this Australian family. Methods: Clinical examinations of the proband and her relatives were performed as part of routine follow up. A polymerase chain reaction (PCR) based test for the duplication in PITX3 was developed, and DNA from 21 members of the proband's family was tested. Results: All clinically affected members of the family had the same 17 bp duplication of PITX3. There was no difference in the size of the duplication between the severely affected individuals and the more mildly affected individuals. Prenatal diagnosis was performed for two offspring of one affected person. In the first pregnancy, the fetus was shown to carry the duplication while in the second pregnancy, the fetus was shown to be homozygous for the normal allele. Conclusions: The results show that in some individuals within one family, duplication of this segment of PITX3 can result in severe symptoms leading to functional blindness while in other individuals in the same family or in other families, the same duplication leads to treatable cataract with minimal visual impairment

    Fatty Acid Profile and Antimicrobial Activity of Rubus ulmifolius Schott Extracts Against Cariogenic Bacterium Streptococcus mutans

    Abstract: The wild edible species Rubus ulmifolius is normally known as a source of several functional- natural compounds used in the traditional diet in several parts of the world. At present, few data are available in the literature about the biological property of its leaves, normally rich in phenolic acids, fatty acids, and other organic compounds with potential antimicrobial activity. Following this hypothesis, we have investigated the antibacterial activity of different dried leaved extracts against the main cariogenic bacterium, Streptococcus mutans. Standard antimicrobial-antibiofilm methods (MIC, MBC, MBIC) were performed to evaluate each extract's antimicrobial profile. In addition, the fatty acids (FA) quali-quantitative profile of R. ulmifolius leave extracts was assessed by reversed-phase HPLC-DAD/ELSD analysis. The results showed that the behavior of this bacterium with different extracts was strictly related to extraction method type, even though it was not related to fatty acid amount and composition, in fact, all the extracts showed similar, qualitative FA patterns, characterized by a concentration in the range from (25 to 36) % of saturated compounds. The methanolic extract showed the better result as antibacterial MIC 6.25 %. These preliminary results encourage further studies for the use of R. ulmifolius in mouthwashes or toothpaste with great anticaries activity

    Treatment of an aneurysmal bone cyst in a young dog: A case report

    BACKGROUND: An aneurysmal bone cyst (ABC) is a rare benign lytic lesion affecting the medullary canal of long bones. It has been widely reported in human medicine, but rarely described in domestic animals. OBJECTIVE: To report the surgical treatment and long term follow‐up of a dog affected by ABC. METHODS: An 8‐month‐old, intact female Weimaraner was presented with lameness affecting the left front limb and progressive swelling of the mid‐distal radius. Survey radiographs revealed a mid‐distal diaphyseal radial lesion. Fine needle aspirates, biopsy, CT scan and histopathology results supported the diagnosis of ABC. Treatment consisted of partial corticotomy of the affected radius, filling of the cystic cavity with demineralised bone matrix and autologous bone graft and stabilisation using lag screws and a neutralisation plate. RESULTS: The long‐term follow‐up, at 36 post‐operative months, showed no recurrence of the cyst and bone modelling. Comparing preoperative radiographs with those at 36 months, bone modelling reduced the radial area by 23.3% in the craniocaudal radiographic view and 30% in the mediolateral projection. CONCLUSIONS: This treatment was sucessful in the case here described, with a 3 years follow‐up

    36. Genome-Wide Insight Into the Transcriptional Modulations Triggered By Lentiviral Transduction in Human Hematopoietic Stem Cells

    Recent studies suggest that hematopoietic stem cells (HSC) can sense foreign nucleic acids and pathogen-associated molecular patterns (PAMPs). Exposure to lentiviral vectors (LV) upon gene transfer may thus trigger acute host responses in HSC that could potentially impact on their biological properties, although no comprehensive studies are available to date. We have performed a high throughput RNA-Seq analysis on human cord-blood (CB)-derived CD34+ hematopoietic stem and progenitor cells (HSPC) exposed to research- or clinical-grade VSV-g pseudotyped (SIN) LV at a high multiplicity of infection, matching current clinical vector dose requirements. As controls, cells were exposed to non-transducing Env-less, genome-less or heat inactivated control vectors or kept in culture untreated. RNA was extracted at different times early after transduction, processed and ran in Illumina HiSeq2000. Analysis of Differential Expression in Time Course was performed using LIMMA R/BioConductor library. Key pathways were assessed by Term Enrichment Analysis considering KEGG pathways and Gene Ontology Biological processes. Transduction with both research-and clinical-grade LV significantly triggered DNA damage and apoptosis-related responses. In particular, p53 signaling was among the most significantly altered pathways (p<3.47×10−14) and induction of several key players, including a 8-fold increase in p21 mRNA, was further confirmed by Taqman. This signaling occurred also in bone-marrow-derived CD34+ cells and was integration-independent as Integrase-Defective LV (IDLV) induced p21 to a similar extent as LV. Furthermore, equal induction was observed in all CD34+ subpopulations, including in the most primitive CD38-CD133+ fraction. Finally, LV/IDLV exposure lead to a slight but significant increase in the percentage of apoptotic HSPC in culture (p<0.001) as compared to control vector exposed cells and untreated controls. Experiments are ongoing to further investigate the potential short and long-term consequences of this signaling on the biological properties of HSPC in vitro and in vivo. Overall, our results suggest for the first time that LV transduction triggers transcriptional changes in HSPC involving pathways pivotal for their biology. Better understanding of the potential functional consequences this may have will be important for the development of improved gene therapy protocols

    Dietary triacylglycerols with palmitic acid in the sn-2 position modulate levels of N-acylethanolamides in rat tissues

    BACKGROUND: Several evidences suggest that the position of palmitic acid (PA) in dietary triacylglycerol (TAG) influences different biological functions. We aimed at evaluating whether dietary fat with highly enriched (87%) PA in sn-2 position (Hsn-2 PA), by increasing PA incorporation into tissue phospholipids (PL), modifies fatty acid profile and biosynthesis of fatty acid-derived bioactive lipids, such as endocannabinoids and their congeners. STUDY DESIGN: Rats were fed for 5 weeks diets containing Hsn-2 PA or fat with PA randomly distributed in TAG with 18.8% PA in sn-2 position (Lsn-2 PA), and similar total PA concentration. Fatty acid profile in different lipid fractions, endocannabinoids and congeners were measured in intestine, liver, visceral adipose tissue, muscle and brain. RESULTS: Rats on Hsn-2 PA diet had lower levels of anandamide with concomitant increase of its congener palmitoylethanolamide and its precursor PA into visceral adipose tissue phospholipids. In addition, we found an increase of oleoylethanolamide, an avid PPAR alpha ligand, in liver, muscle and brain, associated to higher levels of its precursor oleic acid in liver and muscle, probably derived by elongation and further delta 9 desaturation of PA. Changes in endocannabinoids and congeners were associated to a decrease of circulating TNF alpha after LPS challenge, and to an improved feed efficiency. CONCLUSIONS: Dietary Hsn-2 PA, by modifying endocannabinoids and congeners biosynthesis in different tissues may potentially concur in the physiological regulation of energy metabolism, brain function and body fat distribution

    Usefulness of salivary sampling for the molecular detection of a genetic variant associated with bipolar disorders

    Under certain conditions, the hyperthymic temperament traits associated with an increased risk of developing bipolar disorders may in fact produce adaptive responses. The purpose of this study is to see if the type of biological material used for genetic analysis (saliva or blood) affects the detection of mutations in the CACNA1C (RS1006737) gene. The first experimental group consisted of Sardinian migrants (“volunteers”) in South American and European megacities. The second experimental group consisted of older healthy subjects with hyperactivity and novelty-seeking characteristics from Cagliari, Italy. The genetic procedure included DNA extraction, real-time PCR, and the Sanger method. Nonetheless, the authors believe that saliva is the most appropriate biological material, given its many advantages. In contrast to blood, saliva can be collected by any type of healthcare provider after following a few simple instructions
