    The extracellular small leucine-rich proteoglycan biglycan is a key player in gastric cancer aggressiveness

    Biglycan (BGN gene), an extracellular proteoglycan, has been described to be associated with cancer aggressiveness. The purpose of this study was to clarify the clinical value of biglycan as a biomarker in multiple independent GC cohorts and determine the in vitro and in vivo role of biglycan in GC malignant features. We found that BGN is commonly over-expressed in all analyzed cohorts, being associated with disease relapse and poor prognosis in patients with advanced stages of disease. In vitro and in vivo experiments demonstrated that biglycan knock-out GC cells display major phenotypic changes with a lower cell survival, migration, and angiogenic potential when compared with biglycan expressing cells. Biglycan KO GC cells present increased levels of PARP1 and caspase-3 cleavage and a decreased expression of mesenchymal markers. Importantly, biglycan deficient GC cells that were supplemented with exogenous biglycan were able to restore biological features, such as survival, clonogenic and migratory capacities. Our in vitro and in vivo findings were validated in human GC samples, where BGN expression was associated with several oncogenic gene signatures that were associated with apoptosis, cell migration, invasion, and angiogenesis.     Thromboprophylaxis With Apixaban in Patients Undergoing Major Orthopedic Surgery: Meta-Analysis and Trial-Sequential Analysis

    Background: Venous thromboembolism (VTE) is a potentially fatal complication of orthopedic surgery, and until recently, few antithrombotic compounds were available for postoperative thromboprophylaxis. The introduction of the non–vitamin K antagonists oral anticoagulants (NOAC), including apixaban, has extended the therapeutic armamentarium in this field. Therefore, estimation of NOAC net clinical benefit in comparison with the established treatment is needed to inform clinical decision making. Objectives: Systematic review to assess the efficacy and safety of apixaban 2.5 mg twice a day versus low-molecular-weight heparins (LMWH) for thromboprophylaxis in patients undergoing knee or hip replacement. Data sources: MEDLINE, Embase, and CENTRAL were searched from inception to September 2016, other systematic reviews, reference lists, and experts were consulted. Study eligibility criteria, participants, and intervention: All major orthopedic surgery randomized controlled trials comparing apixaban 2.5 mg twice daily with LMWH, reporting thrombotic and bleeding events. Data extraction: Two independent reviewers, using a predetermined form. Study appraisal and synthesis methods: The Cochrane tool to assess risk bias was used by two independent authors. RevMan software was used to estimate pooled risk ratio (RR) and 95% confidence intervals (95% CI) using random-effects meta-analysis. Trial sequential analysis (TSA) was performed in statistical significant results to evaluate whether cumulative sample size was powered for the obtained effect. Overall confidence in cumulative evidence was assessed using the Grading of Recommendations Assessment, Development, and Evaluation (GRADE) Working Group methodology. Results: Four studies comparing apixaban 2.5 mg twice daily with LMWH were included, with a total of 11.828 patients (55% undergoing knee and 45% hip replacement). The overall risk of bias across studies was low. In comparison with LMWH (all regimens), apixaban showed a significantly lower risk of VTE events and overall mortality combined (RR: 0.63, 95% CI: 0.42-0.95, I2 = 84%, n = 8346), but not of major VTE events (RR: 0.62, 95% CI: 0.32-1.19, I2 = 63%, n = 9493), or of symptomatic VTE events and VTE-related mortality combined (RR: 1.14, 95% CI: 0.68-1.90, I2 = 0%, n = 11 879). Trial sequential analysis showed that the risk reduction obtained for VTE and mortality was based on underpowered cumulative sample size and effect dimension. Subgroup analysis according to LMWH regimens showed that apixaban reduced the risk of VTE events and overall mortality, and major VTE events, when compared with LMWH once daily, without differences between apixaban and LMWH twice daily. Conclusions: There is low to moderate evidence that in patients undergoing knee or hip replacement, apixaban seems equally effective and safe to LMWH twice a day. When compared with LMWH once a day, apixaban seems a superior thromboprophylaxis option. However, the results are underpowered which precludes definite answers regarding the true net clinical benefit of apixaban versus LMWH in this clinical context

Os dados históricos são agrupados em quatro tipos característicos: os dias úteis, os sábados, os domingos e os feriados. Faz-se a simulação da aplicação destes modelos à previsão em tempo real. Este tipo de ferramenta apresenta grande facilidade na sua actualização com a incorporação dos registos entretanto ocorridos e, portanto, adaptam-se às eventuais variações. Recorrendo aos resultados destes modelos é possível enviar mensagens de alerta quando as previsões se afastam para além da tolerância admitida, e assim, contribuir para a redução das perdas de água nos sistemas de abastecimento e distribuição.Fundação para a Ciência e a Tecnologia (FCT

    Incorporation of industrial wastes in wood pellets

    ABSTRACT: The present work evaluates the incorporation of industrial wastes (Refuse Derived Fuel-RDF) into biomass for pellet production. Its influence on parameters such as pellet production, combustion and gas emissions was studied for up to 10% of residues incorporation. This approach also deals with the diverting of industrial waste from landfills. The main objectives were: increasing the heat value of the final product, diverting industrial residues with energy potential from landfill and assess the quality of different types of pellets with incorporation of industrial residues. Its implementation was carried out in three phases: selection and characterization of the different industrial residues, production of pellets from different mixtures of wastes and combustion tests. For this purpose a comprehensive characterization of the pellets, the gaseous emissions during combustion and the chemical characterization of the resulting ashes was carried out. The study has shown that the application of industrial residues is a promising route for their incorporation in pellets which should be balanced by the reduction of wastes for landfill. However a few problems were identified: higher difficulty in pelletizing and likely excessive wear of the pellet mill for some of the residues; combustion equipment requiring air supply adjustment and higher ash contents

    A Review of Heat Treatments on Improving the Quality and Residual Stresses of the Ti–6Al–4V Parts Produced by Additive Manufacturing

    Additive manufacturing (AM) can be seen as a disruptive process that builds complex components layer upon layer. Two of its distinct technologies are Selective Laser Melting (SLM) and Electron Beam Melting (EBM), which are powder bed fusion processes that create metallic parts with the aid of a beam source. One of the most studied and manufactured superalloys in metal AM is the Ti–6Al–4V, which can be applied in the aerospace field due to its low density and high melting point, and in the biomedical area owing to its high corrosion resistance and excellent biocompatibility when in contact with tissues or bones of the human body. The research novelty of this work is the aggregation of all kinds of data from the last 20 years of investigation about Ti–6Al–4V parts manufactured via SLM and EBM, namely information related to residual stresses (RS), as well as the influence played by different heat treatments in reducing porosity and increasing mechanical properties. Throughout the report, it can be seen that the expected microstructure of the Ti–6Al–4V alloy is different in both manufacturing processes, mainly due to the distinct cooling rates. However, heat treatments can modify the microstructure, reduce RS, and increase the ductility, fatigue life, and hardness of the components.     The relation between the allowed range of variation of polarization controller wave-plates angles and the respective polarization scattering properties is investigated It is demonstrated that a nearly uniform polarization scattering over the Poincare sphere is obtained using a concatenation of three polarization controllers with angles randomly changed between -pi/4 and pi/4. It is also shown that an improvement of the scattering properties is obtained if the configuration angles are allowed to change between -pi/2 and pi/2

    Two Novel CMY-2-Type β-Lactamases Encountered in Clinical Escherichia Coli Isolates

    BACKGROUND: Chromosomally encoded AmpC β-lactamases may be acquired by transmissible plasmids which consequently can disseminate into bacteria lacking or poorly expressing a chromosomal bla AmpC gene. Nowadays, these plasmid-mediated AmpC β-lactamases are found in different bacterial species, namely Enterobacteriaceae, which typically do not express these types of β-lactamase such as Klebsiella spp. or Escherichia coli. This study was performed to characterize two E. coli isolates collected in two different Portuguese hospitals, both carrying a novel CMY-2-type β-lactamase-encoding gene. FINDINGS: Both isolates, INSRA1169 and INSRA3413, and their respective transformants, were non-susceptible to amoxicillin, amoxicillin plus clavulanic acid, cephalothin, cefoxitin, ceftazidime and cefotaxime, but susceptible to cefepime and imipenem, and presented evidence of synergy between cloxacilin and cefoxitin and/or ceftazidime. The genetic characterization of both isolates revealed the presence of bla CMY-46 and bla CMY-50 genes, respectively, and the following three resistance-encoding regions: a Citrobacter freundii chromosome-type structure encompassing a blc-sugE-bla CMY-2-type -ampR platform; a sul1-type class 1 integron with two antibiotic resistance gene cassettes (dfrA1 and aadA1); and a truncated mercury resistance operon. CONCLUSIONS: This study describes two new bla CMY-2-type genes in E. coli isolates, located within a C. freundii-derived fragment, which may suggest their mobilization through mobile genetic elements. The presence of the three different resistance regions in these isolates, with diverse genetic determinants of resistance and mobile elements, may further contribute to the emergence and spread of these genes, both at a chromosomal or/and plasmid level

    Hydrogen production by catalytic hydrolysis of sodium borohydride in batch reactors: new challenges

    The present manuscript faces the study of H2 generation and storage from catalytic hydrolysis of sodium borohydride (NaBH4) under pressure. We revisit several works on this topic developed (or under development) by our team in the last four years on some of the most critical issues in this research area, namely catalyst durability/reutilization, gravimetric hydrogen storage density and recyclability. New results are also presented. Hydrogen generation rates and yields and hydrogen storage capacities can be augmented to reach 6 wt%, by adding small amounts of an organic polymer (CMC) to the classic NaBH4 hydrolysis, performed with stoichiometric amount of water in a batch reactor with a conical bottom shape and in the presence of Ni-Ru based catalyst, reused from 300 times. Sodium tetrahydroxoborate, NaB(OH)4), was produced in the presence of CMC additive, and did not show crystalline water in its crystal structure. This latter finding has potential to reduce recycling costs of NaBO2 back to NaBH4 and also increase the overall storage density of systems based on NaBH4 as hydrogen carrier
