    Numerical and experimental methods used to evaluate active drag in swimming: a systematic narrative review

    Neonatal alloimmune thrombocytopenia: two different neonatal manifestations of the same disease

    Fetal and neonatal alloimmune thrombocytopenia is a rare disorder in which maternal alloantibodies cross the placenta and cause fetal thrombocytopenia. Most cases are mild but can be potentially fatal if there is an intracranial haemorrhage. Usually, the mother is asymptomatic during pregnancy and no screening is routinely recommended unless there is obstetric or family history. After birth, prompt identification is crucial so that the newborn is closely monitored and treatment is given, if needed, to prevent serious complications. In this article we present two clinical cases of fetal and neonatal alloimmune thrombocytopenia in first-born children, after uneventful pregnancies, with different outcomes and discuss the obstetric management and follow-up in future pregnancies

    Las clases de natación en el contexto escolar durante un período de doce semanas mejoran las habilidades acuáticas y la coordinación motora en niños brasileños

    This study aimed to verify the effects of 12 weeks of swimming lessons implemented in age school Brazilian children’s aquatic skills and motor coordination. Fifty children aged 6 to 10 years old (mean ± SD: 8.34 ± 1.10 years), participated in one swimming lesson per week provided by the school curriculum. Each child was evaluated for their aquatic skills (17 skills checklist) and motor coordination (Körperkoordinationstest Für Kinder test–KTK) before and after the swimming program. This study showed significant improvements in the sum of scores obtained in swimming skills evaluation (31.40 ± 12.89 vs. 46.90 ± 10.73 points, p < .01, d = 3.38). Large effects were found in water orientation and adjustment at the vertical position, breath control, horizontal buoyancy, body position at ventral, dorsal gliding and at longitudinal rotation, front and back somersaults, leg kicking with breath control, feet-first/head-first entries, autonomy in a deep pool and vertical buoyancy and immersion at deep water. The sum of scores in motor coordination assessment increased from 140.64 ± 41.94 to 175.20 ± 41.39 points (p < .01, d = 1.56). These results showed that 12 weeks of swimming practices, even conditioned by state school context (i.e., once a week), allowed Brazilian children aged 6-10 years old to increase aquatic skills and motor coordination.Este estudio tuvo como objetivo verificar los efectos de las clases de natación durante un período de 12 semanas en las habilidades acuáticas y la coordinación motora en niños brasileños. Cincuenta niños entre los 6 y los 10 años de edad (media ± DE: 8.34 ± 1.10 años) participaron una vez por semana en una clase de natación ofrecida por el plan de estudios escolar. Cada niño fue evaluado en cuanto a sus habilidades acuáticas (lista de verificación de 17 habilidades) y coordinación motora (Körperkoordinationstest Für Kinder test - KTK) antes y después del programa de natación. Los resultados del estudio mostraron mejoras significativas en la suma de las puntuaciones obtenidas en la evaluación de las habilidades de natación (31.40 ± 12.89 frente a 46.90 ± 10.73 puntos, p < .01, d = 3.38). Se observaran efectos importantes en la orientación y el ajuste del agua en la posición vertical, en el control de la respiración, en la flotabilidad horizontal, en la posición del cuerpo en la posición ventral, en el deslizamiento dorsal y la rotación longitudinal, en los saltos mortales hacia adelante y hacia atrás, en los empujes con las piernas con control de la respiración, pies primero / cabeza primero entradas, en la autonomía en piscina profunda y flotabilidad vertical y inmersión en aguas profundas. La suma de las puntuaciones en la evaluación de la coordinación motora aumentó de 140.64 ± 41.94 a 175.20 ± 41.39 puntos (p < .01, d = 1.56).     Using wearables to monitor swimmers’ propulsive force to get real-time feedback and understand its relationship to swimming velocity

    Evidence on the role of propulsion compared to drag in swimming, based on experimental settings, is still lacking. However, higher levels of propulsion seem to lead to faster swimming veloci- ties. The aim of this study was to understand the variation in a set of kinematic and kinetic variables between two swimming sections and their relationship to swimming velocity. The sample consisted of 15 young adult recreational swimmers (8 males: 20.84 ± 2.03 years; 7 females: 20.13 ± 1.90 years). Maximum swimming velocity and a set of kinematic and kinetic variables were measured during two consecutive sections of the swimming pool. Differences between sections were measured and the determinants of swimming velocity were analyzed. Swimming velocity, propulsive force, and the other kinematic and kinetic variables did not change significantly (p < 0.05) between sections (only the intra-cyclic fluctuation of swimming velocity decreased significantly, p = 0.005). The modeling identified the propulsive force, stroke length, and active drag coefficient as the determinants of swimming velocity.     Development of a Gold Nanoparticle-Based Lateral-Flow Immunoassay for Pneumocystis Pneumonia Serological Diagnosis at Point-of-Care

GG/001/2014.Pneumocystis jirovecii pneumonia (PcP) is a major human immunodeficiency virus (HIV)-related illness, rising among immunocompromised non-HIV patients and in developing countries. Presently, the diagnosis requires respiratory specimens obtained through invasive and costly techniques that are difficult to perform in all patients or implement in all economic settings. Therefore, the development of a faster, cost-effective, non-invasive and field-friendly test to diagnose PcP would be a significant advance. In this study, recombinant synthetic antigens (RSA) of P. jirovecii’s major surface glycoprotein (Msg) and kexin-like serine protease (Kex1) were produced and purified. These RSA were applied as antigenic tools in immunoenzymatic assays for detection of specific anti-P. jirovecii antibodies (IgG and IgM) in sera of patients with (n = 48) and without (n = 28) PcP. Results showed that only IgM anti-P. jirovecii levels were significantly increased in patients with PcP compared with patients without P. jirovecii infection (p ≤ 0.001 with both RSA). Thus, two strip lateral flow immunoassays (LFIA), based on the detection of specific IgM anti-P. jirovecii antibodies in human sera samples, were developed using the innovative association of P. jirovecii’s RSA with spherical gold nanoparticles (AuNPs). For that, alkanethiol-functionalized spherical AuNPs with ca. ~40 nm in diameter were synthetized and conjugated with the two RSA (Msg or Kex1) produced. These AuNP-RSA conjugates were characterized by agarose gel electrophoresis (AGE) and optimized to improve their ability to interact specifically with serum IgM anti-P. jirovecii antibodies. Finally, two LFIA prototypes were developed and tested with pools of sera from patients with (positive sample) and without (negative sample) PcP. Both LFIA had the expected performance, namely, the presence of a test and control red colored lines with the positive sample, and only a control red colored line with the negative sample. These results provide valuable insights into the possibility of PcP serodiagnosis at point-of-care. The optimization, validation and implementation of this strip-based approach may help to reduce the high cost of medical diagnosis and subsequent treatment of PcP both in industrialized and low-income regions, helping to manage the disease all around the world.publishersversionpublishe

    Introduction: A number of genetic-association studies have identified genes contributing to ankylosing spondylitis (AS) susceptibility but such approaches provide little information as to the gene activity changes occurring during the disease process. Transcriptional profiling generates a 'snapshot' of the sampled cells' activity and thus can provide insights into the molecular processes driving the disease process.     Nutritional characterization of Strychnos madagascariensis fruit flour produced by Mozambican communities and evaluation of Its contribution to nutrient adequacy

    The indigenous fruit Strychnos madagascariensis is usually processed to flour, called nfuma, being highly consumed during staple food shortage. This study aimed to evaluate the nutritional composition of nfuma and its nutrient adequacy. Flours from four districts of Mozambique were analyzed using AOAC methods for proximate composition, HPLC for sugar, amino acids (AA), vitamin E and carotenoids and ICP-MS and FAAS for minerals. The results showed that nfuma stands out for its high content of fat (26.3–27.8%), mainly oleic acid, fiber (>6%), vitamin E (6.7 to 8.0 mg/100 g) and carotenes (2.2 to 2.6 mg/100 g). The main amino acids of nfuma protein were Arg, Asp and Glu, and Lys was the limiting one. The mineral composition reveals K (~1200 to 1700 mg/100 g) as the main macromineral followed by Mg > Ca > Na. The main trace element was Mn (~4 mg/100 g) followed by Fe > Zn > Cu > Cr > Co. Aluminum (~3 mg/100 g) was the main non-essential element and Rb, Ni, Sr, Ba, V, Cd were also quantified. Assuming the daily consumption of 50 g, nfuma provides 82% of Vitamin A dietary reference value for toddlers, while the consumption of 100 g contributes to 132% and 60% of Mn and vitamin A DRV for adults, respectively. Despite the nutritional advantages of nfuma, this flour can be a source of Ni, highlighting the importance of the study of good practices in its preparation to decrease the exposure to non-essential elements.info:eu-repo/semantics/publishedVersio

    Several domains from VAR2CSA can induce Plasmodium falciparum adhesion-blocking antibodies

    <p>Abstract</p> <p>Background</p> <p>Malaria caused by <it>Plasmodium falciparum </it>can result in several different syndromes with severe clinical consequences for the about 200 million individuals infected each year. During pregnancy, women living in endemic areas become susceptible to malaria due to lack of antibodies against a unique <it>P. falciparum </it>membrane protein, named VAR2CSA. This antigen is not expressed in childhood infections, since it binds chondroitin sulphate A (CSA) expressed on the intervillous space in the placenta. A vaccine appears possible because women acquire protective antibodies hindering sequestration in the placenta as a function of parity. A challenge for vaccine development is to design small constructs of this large antigen, which can induce broadly protective antibodies. It has previously been shown that one domain of VAR2CSA, DBL4-FCR3, induces parasite adhesion-blocking antibodies. In this study, it is demonstrated that other domains of VAR2CSA also can induce antibodies with inhibitory activity.</p> <p>Methods</p> <p>All VAR2CSA domains from the 3D7 and HB3 parasites were produced in <it>Baculovirus</it>-transfected insect cells. Groups of three rats per protein were immunized and anti-sera were tested for surface reactivity against infected erythrocytes expressing FCR3 VAR2CSA and for the ability to inhibit FCR3CSA parasite adhesion to CSA. The fine specificity of the immune sera was analysed by VAR2CSA peptide arrays.</p> <p>Results</p> <p>Inhibitory antibodies were induced by immunization with DBL3-HB3 T1 and DBL1-3D7. However, unlike the previously characterised DBL4-FCR3 response the inhibitory response against DBL1-3D7 and DBL3-HB3 T1 was poorly reproduced in the second rounds of immunizations.</p> <p>Conclusion</p> <p>It is possible to induce parasite adhesion-blocking antibodies when immunizing with a number of different VAR2CSA domains. This indicates that the CSA binding site in VAR2CSA is comprised of epitopes from different domains.</p

    MAMMALS IN PORTUGAL : A data set of terrestrial, volant, and marine mammal occurrences in P ortugal

    Mammals are threatened worldwide, with 26% of all species being includedin the IUCN threatened categories. This overall pattern is primarily associatedwith habitat loss or degradation, and human persecution for terrestrial mam-mals, and pollution, open net fishing, climate change, and prey depletion formarine mammals. Mammals play a key role in maintaining ecosystems func-tionality and resilience, and therefore information on their distribution is cru-cial to delineate and support conservation actions. MAMMALS INPORTUGAL is a publicly available data set compiling unpublishedgeoreferenced occurrence records of 92 terrestrial, volant, and marine mam-mals in mainland Portugal and archipelagos of the Azores and Madeira thatincludes 105,026 data entries between 1873 and 2021 (72% of the data occur-ring in 2000 and 2021). The methods used to collect the data were: live obser-vations/captures (43%), sign surveys (35%), camera trapping (16%),bioacoustics surveys (4%) and radiotracking, and inquiries that represent lessthan 1% of the records. The data set includes 13 types of records: (1) burrowsjsoil moundsjtunnel, (2) capture, (3) colony, (4) dead animaljhairjskullsjjaws, (5) genetic confirmation, (6) inquiries, (7) observation of live animal (8),observation in shelters, (9) photo trappingjvideo, (10) predators dietjpelletsjpine cones/nuts, (11) scatjtrackjditch, (12) telemetry and (13) vocalizationjecholocation. The spatial uncertainty of most records ranges between 0 and100 m (76%). Rodentia (n=31,573) has the highest number of records followedby Chiroptera (n=18,857), Carnivora (n=18,594), Lagomorpha (n=17,496),Cetartiodactyla (n=11,568) and Eulipotyphla (n=7008). The data setincludes records of species classified by the IUCN as threatened(e.g.,Oryctolagus cuniculus[n=12,159],Monachus monachus[n=1,512],andLynx pardinus[n=197]). We believe that this data set may stimulate thepublication of other European countries data sets that would certainly contrib-ute to ecology and conservation-related research, and therefore assisting onthe development of more accurate and tailored conservation managementstrategies for each species.
