Influence of substances with acid pH on composite resins microhardness

Resin composites are aesthetic restorative materials widely used in Dentistry and changes in pH can promote amendments in composite surface, compromising their initial characteristics. The aim of this study was to evaluate, in vitro, the effect of low pH beverages on two commercially available materials, the composite resins ICE (SDI) and Durafill VS (Heraeus Kulzer). The materials were evaluated for surface microhardness obtained by Vickers microhardness testing. Fifteen samples were prepared with 11 mm diameter and 2 mm thick, each restorative material. The samples were kept in storage conditions of the following substances: Gatorade ® (pH 2,9); Coca- Cola ® (pH 2,2) and distilled water (pH 6,0; control group) under temperature of 37 ° C, and the environment of immersing exchanged every 24 hours. Five samples of each material were immersed in the beverages tested and monitored for periods of 24 hours, 7, 14 and 21 days. The results were submitted to multifactorial analysis of variance and Tukey test. There were statistically significant differences (p 0.05) for factors environment, material and time, and for the interaction between them. The distilled water resulted in higher values of microhardness, followed by Coca-Cola® and Gatorade®. The highest values of microhardness were obtained at baseline, followed by 7, 14 and 21 days. There was no statistically significant difference (p 0,05) between the times of 14 and 21 days. It was concluded that the nanocomposite better results and that the acidic pH associated with progressive immersion time significantly alter the microhardness of the composites evaluated

Production and purification of an immunogenic G protein of rabies virus from S2 insect cells

Rabies virus is still a major public health concern in under developed countries of Africa, Asia and Latin America. WHO estimates that almost 55.000 people die from rabies annually. Although Rabies virus strains from dogs still are the main responsible of causing human disease, the increasing case numbers of bat virus strains being transmitted to humans in America raises concerns. The called secondary transmission cycle of rabies involves cats or dogs which are exposed to contaminated bats and then transmit it to humans. Combating this type of transmission requires new strategies for disease control, including new vaccines. Particularly for cats, the current rabies vaccines have demonstrated to cause vaccine-associated sarcoma as a serious side effect. New rabies vaccines using recombinant technology have been proposed using the Rabies Glycoprotein (G) as antigen. The expression of this molecule in S2 insect cells has already proved the ability of this cell line to produce an immunogenic protein. Although this antigen is recognized as the main protein necessary for protection upon immunization, its utilization as a recombinant vaccine has been impaired by difficulties in its production and mainly purification steps. The G protein was expressed as a membrane bound protein in Drosophila melanogaster S2 cells. This membrane G protein was further solubilized using a standardized solution containing detergent and purified in a novel methodology using affinity chromatography resulting in protein recoveries up to 96 %. The membrane protein and the purified G were used for mice immunization, using or not ISCOM adjuvant resulting in good levels of specific antibodies and neutralizing antibodies. When immunized mice were submitted to lethal rabies virus challenge, the preparations showed to confer protection levels similar to that of commercially available veterinary rabies vaccine. A flow path from the production, purification, formulation and immunization against Rabies using a recombinant G protein is presented as a promisor veterinary vaccine candidate

Le reti neurali in problemi di classificazione: una soluzione basata sui metodi di segmentazione binaria

In this paper the use of neural networks in the solution of a classification problem is described and a non parametric classifier which combines radial basis function networks (RBF networks) and binary classification trees is proposed. It is shown that from a classification tree a class of RBF networks can be derived, whose elements differ in the degree of dependence from the discriminant rules described by the tree. A simulation study based on a two-class problem shows that the proposal represents a valid solution, particularly in presence of noisy variables
