    A Multi-parameter Model for Effective Configuration of Supply Chains

    In this paper, a mathematical model for the strategic planning and design of supply chains in the globalization context is proposed. A multi-objective function is used to address decisions about capacity sizing, sourcing, and facility location, with the scope of maximizing supply chain profits. The model is dynamic and is applied to a multi- echelon, multi-facility and multi-product supply chain in hypotheses of delocalization. The model is characterized by the specific attention given to cost, revenue and financial factor modelling, which has been obtained by means of an activity-based approach and the inclusion of two drivers that are usually neglected in the literature: energy and labour. The model’s applicability and flexibility have been proved via a scenario analysis in which 12 different scenarios were implemented to reproduce the behaviour of major industries

    Changing the Habitat at Academic Conferences: Using a Learning Ecosystem with Active Learning During a Panel Presentation

    Abstract In order to assess the effectiveness and feasibility of an active learning event during a panel presentation at an academic conference, Mercer University librarians presenting at the Georgia Libraries Conference switched the traditional way panel presentations are modeled. Instead of the question and answer session following a brief overview of the presentation, we moved our physical position in the room, closer to the participants in order to have a more intimate conversation with attendees. Using two active learning techniques, discussion and brainstorming, the presenters started a conversation with attendees about project ideas involving teaching faculty members, librarians, and students and how this type of learning ecosystem would work or already works at their institutions. Though each librarian in the discussion and brainstorming session varied in approach and their types of projects, the model of the ecosystem remained consistent. Our team of librarians then proceeded to test whether or not an active learning event could take place during our presentation

    Mutations in ACTA2, encoding the smooth muscle cell (SMC)-specific isoform of α-actin (α-SMA), cause thoracic aortic aneurysms and dissections and occlusive vascular diseases, including early onset coronary artery disease and stroke. We have shown that occlusive arterial lesions in patients with heterozygous ACTA2 missense mutations show increased numbers of medial or neointimal SMCs. The contribution of SMC hyperplasia to these vascular diseases and the pathways responsible for linking disruption of α-SMA filaments to hyperplasia are unknown. Here, we show that the loss of Acta2 in mice recapitulates the SMC hyperplasia observed in ACTA2 mutant SMCs and determine the cellular pathways responsible for SMC hyperplasia. Acta2−/− mice showed increased neointimal formation following vascular injury in vivo, and SMCs explanted from these mice demonstrated increased proliferation and migration. Loss of α-SMA induced hyperplasia through focal adhesion (FA) rearrangement, FA kinase activation, re-localization of p53 from the nucleus to the cytoplasm and increased expression and ligand-independent activation of platelet-derived growth factor receptor beta (Pdgfr-ÎČ). Disruption of α-SMA in wild-type SMCs also induced similar cellular changes. Imatinib mesylate inhibited Pdgfr-ÎČ activation and Acta2−/− SMC proliferation in vitro and neointimal formation with vascular injury in vivo. Loss of α-SMA leads to SMC hyperplasia in vivo and in vitro through a mechanism involving FAK, p53 and Pdgfr-ÎČ, supporting the hypothesis that SMC hyperplasia contributes to occlusive lesions in patients with ACTA2 missense mutation

    Human Monocytes Exposed to SARS-CoV-2 Display Features of Innate Immune Memory Producing High Levels of CXCL10 upon Restimulation

    Introduction: A role for innate immune memory in protection during COVID-19 infection or vaccination has been recently reported. However, no study so far has shown whether the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) can train innate immune cells. The aim of this study was to investigate whether this virus can induce trained immunity in human monocytes. Methods: Monocytes were exposed to inactivated SARS-CoV-2 (iSARS-CoV-2) for 24 h, followed by a resting period in the medium only and a secondary stimulation on day 6 after which the cytokine/chemokine and transcriptomic profiles were determined. Results: Compared to untrained cells, the iSARS-CoV-2-trained monocytes secreted significantly higher levels of IL-6, TNF-α, CXCL10, CXCL9, and CXCL11 upon restimulation. Transcriptome analysis of iSARS-CoV-2-trained monocytes revealed increased expression of several inflammatory genes. As epigenetic and metabolic modifications are hallmarks of trained immunity, we analyzed the expression of genes related to these processes. Findings indicate that indeed SARS-CoV-2-trained monocytes show changes in the expression of genes involved in metabolic pathways including the tricarboxylic acid cycle, amino acid metabolism, and the expression of several epigenetic regulator genes. Using epigenetic inhibitors that block histone methyl and acetyltransferases, we observed that the capacity of monocytes to be trained by iSARS-CoV-2 was abolished. Conclusion: Overall, our findings indicate that iSARS-CoV-2 can induce properties associated with trained immunity in human monocytes. These results contribute to the knowledge required for improving vaccination strategies to prevent infectious diseases

    A critical guide to the automated quantification of perivascular spaces in magnetic resonance imaging

    The glymphatic system is responsible for waste clearance in the brain. It is comprised of perivascular spaces (PVS) that surround penetrating blood vessels. These spaces are filled with cerebrospinal fluid and interstitial fluid, and can be seen with magnetic resonance imaging. Various algorithms have been developed to automatically label these spaces in MRI. This has enabled volumetric and morphological analyses of PVS in healthy and disease cohorts. However, there remain inconsistencies between PVS measures reported by different methods of automated segmentation. The present review emphasizes that importance of voxel-wise evaluation of model performance, mainly with the SĂžrensen Dice similarity coefficient. Conventional count correlations for model validation are inadequate if the goal is to assess volumetric or morphological measures of PVS. The downside of voxel-wise evaluation is that it requires manual segmentations that require large amounts of time to produce. One possible solution is to derive these semi-automatically. Additionally, recommendations are made to facilitate rigorous development and validation of automated PVS segmentation models. In the application of automated PVS segmentation tools, publication of image quality metrics, such as the contrast-to-noise ratio, alongside descriptive statistics of PVS volumes and counts will facilitate comparability between studies. Lastly, a head-to-head comparison between two algorithms, applied to two cohorts of astronauts reveals how results can differ substantially between techniques

    The history of discovery of bosonic string theory is well documented. This theory evolved as an attempt to find a multidimensional analogue of Euler's beta function. Such an analogue had in fact been known in mathematics literature at least in 1922 and was studied subsequently by mathematicians such as Selberg, Weil and Deligne among others. The mathematical interpretation of this multidimensional beta function is markedly different from that described in physics literature. This paper aims to bridge the gap between the existing treatments. Preserving all results of conformal field theories intact, developed formalism employing topological, algebro-geometric, number-theoretic and combinatorial metods is aimed to provide better understanding of the Veneziano amplitudes and, thus, of string theories.Comment: 92 pages LaTex, some typos removed, discussion section is added along with several additional latest reference
