    Regulatory Functions of Pax1 and Pax9 in Mammalian Cells

    Pax1 and Pax9 are paired-box transcription factors, which play vital roles in axial skeletogenesis, thymus organogenesis, palatogenesis and odontogenesis among others. The importance of these closely related transcription factors can be perceived from the various human anomalies associated with their disruption. Vertebral column abnormalities such as kyphoscoliosis, seen in Jarcho-Levine and Klippel-Feil syndromes, secondary cleft palate, oligodontia/ hypodontia (missing teeth) and thymus developmental defects have all been associated with mutations in PAX1 and/or PAX9.     Due to the current environmental situation, the reducing of greenhouse gas emission and the saving energy is the phenomenon. The building sector is still growing and more and more energy is needed. Thermal performance of building envelope has been of great importance in the context of existing global warming issues. Buildings are responsible for 40% of energy consumption and 36% of Carbon Dioxide (CO2) emissions in the member states of the European Union. According to the research project Heartland Green Sheets, the recommended criteria for assessments of sustainable buildings materials are low embodied energy, recyclable, use renewable resources, locally or regionally produced, energy efficient, low environmental impact, durable, minimize waste, positive social impact and affordable. The contribution focuses on life-cycle assessment (LCA) and sustainability assessment of commonly used wall systems. The multi-criteria analysis of the contemporary wall systems in term of sustainable development is presented in the paper.     Assessment of indoor air quality in university classrooms

    This contribution describes the subjective assessment process of indoor environmental conditions in university classrooms. The university campus consisting of several buildings is situated on the outskirts of the town of České Budějovice (South Bohemia, Czech Republic). A high level of indoor environment quality (IEQ) is a key factor to achieve healthy environments in buildings. Suitable indoor air quality is more important in schools and universities (no-industrial civic amenities) than in many other buildings. Indoor air quality in the university environment affects learning processes, concentration, and productivity of students. It also could affect the health of students and teachers in long-term. Students are at greater risk because of the hours spent in educational facilities. The evaluation of the subjective assessment of the quality of the indoor environment at university classrooms is described within the paper

We have previously shown that data from a genome wide binding assay combined with transcriptional profiling is an insightful means to divulge the mechanisms directing cell type specification and the generation of tissues and subsequent organs [1].     COMT val158met Polymorphism and Neural Pain Processing

    A functional polymorphism (val158met) of the gene coding for Catechol-O-methyltransferase (COM) has been demonstrated to be related to processing of emotional stimuli. Also, this polymorphism has been found to be associated with pain regulation in healthy subjects. Therefore, we investigated a possible influence of this polymorphism on pain processing in healthy persons as well as in subjects with markedly reduced pain sensitivity in the context of Borderline Personality Disorder (BPD). Fifty females (25 patients with BPD and 25 healthy control participants) were included in this study. Genotype had a significant – though moderate - effect on pain sensitivity, but only in healthies. The number of val alleles was correlated with the BOLD response in several pain-processing brain regions, including dorsolateral prefrontal cortex, posterior parietal cortex, lateral globus pallidus, anterior and posterior insula. Within the subgroup of healthy participants, the number of val alleles was positively correlated with the BOLD response in posterior parietal, posterior cingulate, and dorsolateral prefrontal cortex. BPD patients revealed a positive correlation between the number of val alleles and BOLD signal in anterior and posterior insula. Thus, our data show that the val158met polymorphism in the COMT gene contributes significantly to inter-individual differences in neural pain processing: in healthy people, this polymorphism was more related to cognitive aspects of pain processing, whereas BPD patients with reduced pain sensitivity showed an association with activity in brain regions related to affective pain processing

    UV stalled replication forks restart by re-priming in human fibroblasts

    Restarting stalled replication forks is vital to avoid fatal replication errors. Previously, it was demonstrated that hydroxyurea-stalled replication forks rescue replication either by an active restart mechanism or by new origin firing. To our surprise, using the DNA fibre assay, we only detect a slightly reduced fork speed on a UV-damaged template during the first hour after UV exposure, and no evidence for persistent replication fork arrest. Interestingly, no evidence for persistent UV-induced fork stalling was observed even in translesion synthesis defective, Polηmut cells. In contrast, using an assay to measure DNA molecule elongation at the fork, we observe that continuous DNA elongation is severely blocked by UV irradiation, particularly in UV-damaged Polηmut cells. In conclusion, our data suggest that UV-blocked replication forks restart effectively through re-priming past the lesion, leaving only a small gap opposite the lesion. This allows continuation of replication on damaged DNA. If left unfilled, the gaps may collapse into DNA double-strand breaks that are repaired by a recombination pathway, similar to the fate of replication forks collapsed after hydroxyurea treatment
