    Activation of nonselective cation channels by physiological cholecystokinin concentrations in mouse pancreatic acinar cells

    The activation of the nonselective cation channels in mouse pancreatic acinar cells has been assessed at low agonist concentrations using patch-clamp whole cell, cell-attached patch, and isolated inside-out patch recordings. Application of acetylcholine (ACh) (25-1,000 nM) and cholecystokinin (CCK) (2-10 pM) evoked oscillatory responses in both cation and chloride currents measured in whole cell experiments. In cell-attached patch experiments we demonstrate CCK and ACh evoked opening of single 25-pS cation channels in the basolateral membrane. Therefore, at least a component of the whole cell cation current is due to activation of cation channels in the basolateral acinar cell membrane. To further investigate the reported sensitivity of the cation channel to intracellular ATP and calcium we used excised inside-out patches. Micromolar Ca2+ concentrations were required for significant channel activation. Application of ATP and ADP to the intracellular surface of the patch blocked channel opening at concentrations between 0.2 and 4 mM. The nonmetabolizable ATP analogue, 5'- adenylylimidodiphosphate (AMP-PNP, 0.2-2 mM), also effectively blocked channel opening. The subsequent removal of ATP caused a transient increase in channel activity not seen with the removal of ADP or AMP- PNP. Patches isolated into solutions containing 2 mM ATP showed channel activation at micromolar Ca2+ concentrations. Our results show that ATP has two separate effects. The continuous presence of the nucleotide is required for operation of the cation channels and this action seems to depend on ATP hydrolysis. ATP can also close the channel and this effect can be demonstrated in excised inside-out patches when ATP is added to the bath after a period of exposure to an ATP-free solution. This action does not require ATP hydrolysis. Under physiological conditions hormonal stimulation can open the nonselective cation channels and this can be explained by the rise in the intracellular free Ca2+ concentration

    Regularised Volterra series models for modelling of nonlinear self-excited forces on bridge decks

    Volterra series models are considered an attractive approach for modelling nonlinear aerodynamic forces for bridge decks since they extend the convolution integral to higher dimensions. Optimal identification of nonlinear systems is a challenging task since there are typically many unknown variables that need to be determined, and it is vital to avoid overfitting. Several methods exist for identifying Volterra kernels from experimental data, but a large class of them put restrictions on the system inputs, making them infeasible for section model tests of bridge decks. A least-squares identification method does not restrict the inputs, but the identified model often struggles with noisy (non-smooth) kernels, which is deemed to be unphysical and a sign of overfitting. In this work, regularised least-squares identification is introduced to improve the performance of model identification using least-squares. Standard Tikhonov regularisation and other penalty techniques that impose decaying kernels are also explored. The performance of the methodology is studied using experimental data from wind tunnel tests of a twin deck section. The regularised Volterra models show equal or better results in terms of modelling the self-excited forces, and the regularisation makes the models less prone to overfitting

    On the Role of Global Warming on the Statistics of Record-Breaking Temperatures

    We theoretically study long-term trends in the statistics of record-breaking daily temperatures and validate these predictions using Monte Carlo simulations and data from the city of Philadelphia, for which 126 years of daily temperature data is available. Using extreme statistics, we derive the number and the magnitude of record temperature events, based on the observed Gaussian daily temperatures distribution in Philadelphia, as a function of the number of elapsed years from the start of the data. We further consider the case of global warming, where the mean temperature systematically increases with time. We argue that the current warming rate is insufficient to measurably influence the frequency of record temperature events over the time range of the observations, a conclusion that is supported by numerical simulations and the Philadelphia temperature data.Comment: 11 pages, 6 figures, 2-column revtex4 format.     We complete the picture of sharp eigenvalue estimates for the p-Laplacian on a compact manifold by providing sharp estimates on the first nonzero eigenvalue of the nonlinear operator Δp\Delta_p when the Ricci curvature is bounded from below by a negative constant. We assume that the boundary of the manifold is convex, and put Neumann boundary conditions on it.     Calcium oscillations in pancreatic acinar cells, evoked by the cholecystokinin analogue JMV-180, depend on functional inositol 1,4,5- trisphosphate receptors

    It has been reported that the synthetic heptapeptide cholecystokinin (CCK) analogue JMV-180 evokes cytosolic Ca2+ signals in pancreatic acinar cells via mechanisms that do not include either the generation or action of inositol 1,4,5-trisphosphate (InsP3) (Saluja, A. K., Dawra, R. K., Lerch, M. M., and Steer, M. L. (1992) J. Biol. Chem. 267, 11202-11207; Yule, D. I., and Williams, J. A. (1992) J. Biol. Chem. 267, 13830-13835). We have now investigated the CCK- and JMV-180-evoked cytosolic Ca2+ oscillations by measurement of the Ca(2+)-sensitive ion currents in internally perfused mouse pancreatic acinar cells. We find that the InsP3 receptor antagonist heparin (500 micrograms/ml) blocks Ca2+ oscillations induced by both CCK (5-20 pM) and JMV 180 (10-40 nM), whereas de-N-sulfated heparin (500 micrograms/ml), which does not affect InsP3 binding to its receptor, fails to inhibit the responses to the two agonists. We conclude that the cytosolic Ca2+ oscillations evoked by both CCK and JMV-180 are dependent on functional InsP3 receptors

    SHORT COMMUNICATION: Complementary tumor induction in neural grafts exposed to N-ethyl-N-nitrosourea and an activated myc gene

    Using a combination of transplacental carcinogen exposure and retrovirus-mediated oncogene transfer into fetal brain transplants, we have studied complementary transformation by N-ethyl-N-nitrosourea (NEU) and the v-myc oncogene in the nervous system. Previous experiments had demonstrated that both agents will not induce tumors independently whereas simultaneous expression of v-H-ras and v-gag/myc exerted a powerful transforming potential in neural grafts. In order to identify other genetic alterations that co-operate with an activated myc gene, the neurotropic carcinogen NEU was used to generate mutations of cellular genes. On embryonic day 14 (ED14), pregnant donor animals (F344 rats) received a single i.v. dose of NEU (50 mg/kg). Twenty-four hours later (ED15), the fetal brains were removed, triturated and incubated with a retroviral vector carrying the v-gag/myc oncogene. Subsequently, these primary cell suspensions were transplanted stereotactically into the caudate-putamen of syngenic adult recipients. After latency periods of 3-6 months, 5 of 10 recipients harboring ED15 fetal brain transplants developed malignant, poorly differentiated neuroectodermal tumors in the grafts. No tumor development was observed in seven recipients harboring ED16 neural grafts. Cell lines were established from three tumors and the 110 kd gag/myc fusion protein encoded by the retroviral construct was identified in the tumors by Western blotting. Several candidate genes for mutational activation by NEU including the H-ras, K-ras and neu oncogenes were analyzed for specific point mutations by polymerase chain reaction (PCR) and direct DNA sequencing of the PCR products. However, no mutations were found in any of these genes. These findings lend further support to the multistep hypothesis of neoplastic transformation in the brain.     Distinct nature of static and dynamic magnetic stripes in cuprate superconductors

    We present detailed neutron scattering studies of the static and dynamic stripes in an optimally doped high-temperature superconductor, La2_2CuO4+y_{4+y}. We find that the dynamic stripes do not disperse towards the static stripes in the limit of vanishing energy transfer. We conclude that the dynamic stripes observed in neutron scattering experiments are not the Goldstone modes associated with the broken symmetry of the simultaneously observed static stripes, but rather that the signals originate from different domains in the sample. These domains may be related by structural twinning, or may be entirely different phases, where the static stripes in one phase are pinned versions of the dynamic stripes in the other. Our results explain earlier observations of unusual dispersions in underdoped La2−x_{2-x}Srx_xCuO4_{4} (x=0.07x=0.07) and La2−x_{2-x}Bax_xCuO4_{4} (x=0.095x=0.095). Our findings are relevant for all compounds exhibiting magnetic stripes, and may thus be a vital part in unveiling the nature of high temperature superconductivity
