    Práce se zabývá rozborem konstrukčních řešení velkoobjemových překládacích vozů používaných v dnešní době. V první části je proveden vlastní návrh konstrukce velkoobjemového překládacího vozu. Dále je proveden návrh šnekového dopravníku. Dále byla provedena pevnostní kontrola podvozku velkoobjemového překládacího vozu pomocí metody konečných prvků. Na základě výsledků pevnostní kontroly byly navrženy úpravy podvozku. Nakonec byla provedena pevnostní kontrola upraveného podvozku. Přílohy práce obsahují velikost deformace podvozku a výkresovou dokumentaci překládacího vozu. Práce byla vytvořena ve spolupráci s firmou ZDT spol. s.r.o. Nové veselí.This thesis deals with analysis of constructional solution of large-loading wagons used nowadays. In the first part of this thesis is my own construction design of large-loading wagon, furthermore there is a design of screw conveyor. In the following part stress strain analysis of the chassis of large-loading wagon was made using the finite element method. On the basis of these results were suggested modifications of the chassis. Finally the stress strain analysis of the adapted chassis was made. Attachments of the thesis include deformation results of the chassis and design documentation of the large-loading wagon. This thesis was created in cooperation with ZDT spol. s.r.o. Nové Veselí.

    Modified Biochar—A Tool for Wastewater Treatment

    Global deposits of concentrated phosphates, which are a necessary source for the production of phosphate fertilizers, are limited. These reserves keep getting thinner, and every day, large amounts of phosphorus end up in watercourses. In this study, we verified that modified biochar (saturated with FeCl3 solution and then neutralized with NaOH solution) can adsorb significant amounts of phosphorus from wastewater. Moreover, the agrochemical qualities of sludge water from a municipal wastewater treatment plant, struvite, phosphorus-saturated biochar, and iron(III) phosphate from a reused biochar filter were tested in this study. We determined the amount of mobile phosphorus as well as the amount of extractable phosphorus and its five fractions. It was found that modified biochar can hold one-third of the phosphorus amount contained in the commonly used agricultural fertilizer simple superphosphate (1 × 105 g of modified biochar captures up to 2.79 × 103 g of P). Moreover, plants can more easily access phosphorus biochar fractions than struvite, which is formed spontaneously during sludge management. The results of this research prove that the proposed method of recycling phosphorus from wastewater can be applied in technological practice

    Práce se zabývá kolovými nakladači, řeší jejich konstrukci a hledá nejoptimálnější stroj. Pro nalezení optimálního stroje práce obsahuje tabulky s parametry strojů, ve kterých je uvedeno 218 modelů od 26 výrobců. Modely jsou rozděleny do 4 skupin, ve kterých jsou porovnávány pomocí kritérií a následně je proveden výběr nejlepšího a nejhoršího stroje.The thesis is focused on wheel loaders, it solves their construction and seeks for the best machine. To find the optimal machine the thesis contains tables with parameters of the machines, in which 218 models from 26 manufacturers are mentioned. Loaders are divided into four groups, in which they are compared using variable criteria and finally the selection of the best and worst machine is made.

    Tato bakalářská práce se zabývá tématem strategie pro vstup na zahraniční trh relativně malé české firmy s názvem PaperTime s.r.o. a popisuje dopad pandemie COVID-19 na její podnikání. Cílem práce je zjistit, zda by pro malou a rychle rostoucí společnost, jako je PaperTime, bylo možné expandovat a uspět na zahraničním trhu, na základě analýzy vybrat vhodnou zemi pro vstup, a navrhnout formu, jakou na zvolený trh vstoupit. Na základě analýzy makroprostředí a konkurenčního prostředí je provedena komparace vybraných trhů, německého a polského. Výstupem je volba polského trhu z důvodu nízké konkurence a méně rozvinutého niche marketu, na který se PaperTime specializuje. V poslední části je navržena strategie vstupu na zvolený trh na základě krokových modelů internacionalizace, přičemž prvním navrženým krokem je cross-border e-commerce.This bachelor’s thesis deals with the topic of foreign market entry of a relatively small Czech company named PaperTime s.r.o. and describes the impact of the COVID-19 pandemic on its operations. The aim of the thesis can be broken down into three categories: 1) determining whether a small but fast-growing company such as PaperTime can expand and succeed in foreign markets, 2) selecting a suitable foreign market for entry based on the analysis, and 3) suggesting the ideal form of entry into the chosen market. Through the analysis of macro and competitive environments, a comparison was made between the German and Polish markets. Conclusively the decision was made to enter the Polish market as there is minimal competition and a lesser developed niche market, in which PaperTime specializes. The last part focuses on designing a market entry strategy based on stage models of internationalization, while the first stage is cross-border e-commerce

    Mitochondrial genome acquisition restores respiratory function and tumorigenic potential of cancer cells without mitochondrial DNA

    We report that tumor cells devoid of their mitochondrial genome (mtDNA) show delayed tumor growth and that tumor formation is associated with acquisition of mtDNA from host cells. This leads to partial recovery of mitochondrial function in cells derived from primary tumors grown from cells without mtDNA and a shorter lag in tumor growth. Cell lines from circulating tumor cells showed further recovery of mitochondrial respiration and an intermediate lag to tumor growth, while cells from lung metastases exhibited full restoration of respiratory function and no lag in tumor growth. Stepwise assembly of mitochondrial respiratory supercomplexes was correlated with acquisition of respiratory function. Our findings indicate horizontal transfer of mtDNA from host cells in the tumor microenvironment to tumor cells with compromised respiratory function to re-establish respiration and tumor-initiating efficacy. These results suggest a novel pathophysiological process for overcoming mtDNA damage and support the notion of high plasticity of malignant cells
