    Fast electrophoresis in conventional capillaries by employing a rapid injection device and contactless conductivity detection

    A purpose-made set-up featuring an automated fast injector allowed the easy optimization of the injected amount and the adjustment of the separation length of conventional capillaries from a minimum of 5cm upward. It was found that a compromise in capillary length for separation efficiency and analysis time also has to take into account the injected amount, which in turn affects the sensitivity and hence the detection limit. The versatility of the system was demonstrated by the analysis of the major cations and anions in natural water samples in less than 1min, the concurrent determination of a mixture of amino acids and carbohydrates in 160s, and of three active ingredients in a pharmaceutical preparation in 40s. Plate numbers were typically around 50,000 and detection limits down to 1ÎĽM could be achieve

    Two Optimal Strategies for Active Learning of Causal Models from Interventional Data

    From observational data alone, a causal DAG is only identifiable up to Markov equivalence. Interventional data generally improves identifiability; however, the gain of an intervention strongly depends on the intervention target, that is, the intervened variables. We present active learning (that is, optimal experimental design) strategies calculating optimal interventions for two different learning goals. The first one is a greedy approach using single-vertex interventions that maximizes the number of edges that can be oriented after each intervention. The second one yields in polynomial time a minimum set of targets of arbitrary size that guarantees full identifiability. This second approach proves a conjecture of Eberhardt (2008) indicating the number of unbounded intervention targets which is sufficient and in the worst case necessary for full identifiability. In a simulation study, we compare our two active learning approaches to random interventions and an existing approach, and analyze the influence of estimation errors on the overall performance of active learning

    Appropriate timing of treatment assumes particular importance in critical care. Lange and colleagues recently reported on the time course of the different nitric oxide synthase (NOS) isoforms, nitrosative stress, and poly(ADP-ribosylation) during Pseudomonas aeruginosa pneumonia-induced ovine septic shock. Initially, endothelial NOS expression was increased together with markers of peroxynitrite formation, DNA damage, and nuclear factor-kappa-B activation. Later on, measurable NOS activity and nitric oxide production resulted mainly from inducible NOS activation. These results emphasize the need for long-term, large-animal studies investigated over days so that future therapeutic interventions can be better tailored and matched to the exact time course of the activation of the mediator orchestra

    Determination of ethylene by field asymmetric ion mobility spectrometry

    The determination of ethylene with a field asymmetric ion mobility spectrometer, which can easily be constructed in-house, is described. The device makes use of a Krypton lamp for ionization. A rectangular pulse of 500 V(pp)at 1 MHz was employed as separation waveform in the drift tube rather than the commonly used less efficient bisinusoidal waveform. The calibration curve for the range from 670 ppb((V/V))to 67 ppm((V/V))was found to be highly linear with a correlation coefficient of r = 0.9999. The limit of detection was determined as 200 ppb((V/V)). The reproducibility was 4% (relative standard deviation). The device was found to be suitable for the determination of ethylene given off by fruit; 6 types of climacteric fruit were tested, namely apples, bananas, kiwi fruit, nectarines, pears and plums

    Determination of Binary Gas Mixtures by Measuring the Resonance Frequency in a Piezoelectric Tube

    The composition of gas mixtures may be determined via changes of the speed of sound. As this affects the resonance frequency of the gas inside a tube, indirect measurements through a frequency analysis are also possible. It is demonstrated that this may be carried out with unprecedented simplicity by the novel employment of a piezoelectric tube which serves at the same time as a resonance tube and as transducer into the electronic domain. Experiments were run using a simple diecast aluminum box as the measuring cell, inside which the piezoelectric tube made from lead zirconium titanate with 30-mm length and 5.35-mm inner diameter was suspended. A small loudspeaker placed into the cell served for excitation of the resonance. Peak frequencies between 3910 and 14,590 Hz (for pure CO2 and He, respectively) were obtained. Two component mixtures of O2/N2, CO2/N2, and He/N2 at various composition were tested. A linear frequency change from 4790 to 5100 Hz was observed when going from pure O2 to pure N2

    The publications concerning capacitively coupled contactless conductivity detection for the 2-year period from mid-2016 to mid-2018 are covered in this update to the earlier reviews of the series. Relatively few reports on fundamental investigations or new designs have appeared in the literature in this time interval, but the development of new applications with the detection method has continued strongly. Most often, contactless conductivity measurements have been employed for the detection of inorganic or small organic ions in conventional capillary electrophoresis, less often in microchip electrophoresis. A number of other uses, such as detection in chromatography or the gauging of bubbles in streams have also been reported
